    Notizen: Abstract: The rat substance P (SP) receptor (SPR) was expressed in insect Sf9 cells by infection with recombinant baculovirus. The receptor bound SP with high affinity (KD = 360 pM) and had a rank order of affinity of SP 〉 neurokinin A 〉 neurokinin B. Ligand activation of the receptor resulted in an increase in both inositol lipid hydrolysis and intracellular Ca2+ concentration ([Ca2+]i). However, high-level expression of the receptor, in the absence of ligand, was correlated with increased basal turnover of inositol lipids and an elevated rate of Ca2+ influx. These results demonstrate that the Sf9 cells provide a suitable environment for the high-level expression of a functionally active SPR. Two carboxy-terminal epitope-tagged receptors (SPR-KT3 = SPR-TPPPEPET, COOH; SPR-Glu = SPR-EEEEYMPME, COOH) were also expressed. The affinity of the KT3-tagged receptor for ligand was similar to that of the wild-type receptor (KD = 405 pM), and that of the Glu-tagged receptor was slightly lower (KD = 1,082 pM). The high-affinity SP binding site of all three receptors was sensitive to guanosine 5′-O-(3-thiotriphosphate) pretreatment. The maximal signal-transducing ability of the epitope-tagged receptors was comparable to that of the wild-type receptor ([Ca2+]i rise as a percentage of wild-type: SPR-KT3, 80–100%; SPR-Glu, 88–100%). These data show that heterologous expression in the baculovirus system results in high expression of functional wild-type and tagged receptors.

    Notizen: A first-principles calculation of the magnetic anisotropy energies of X/Co (X=Pt, Cu, Ag and Au) metallic multilayers has been performed to investigate the effect of strain on the magnetic anisotropy. It is successfully predicted that Pt/Co and Au/Co multilayers exhibit perpendicular magnetic anisotropies in accordance with experiments. The strength of the hybridization of electronic states at the interface determines the relative position of the Fermi energy to the local density of states (LDOS) of ||m||=2 character of Co d electrons of minority spin. If the LDOS of ||m||=2 character is large at the Fermi energy, a system shows a perpendicular anisotropy. The anisotropy energies of all the systems increase as a function of interatomic distance in the in-plane direction. The magneto-elastic constants are more than twice the value of pure Co. The origin of this large value is explained in terms of the change of relative position of the Fermi level to that of the LDOS of ||m||=2 character as the value of strain increases. © 1996 American Institute of Physics.

    Notizen: Although postmortem inspection (organoleptic examination by seeing, feeling, and smelling) of market poultry contributes significantly to the assurance of wholesomeness of the meat, such assurance is relative and not absolute. A significantly higher incidence of salmonellae in the cecal area of the intestines of condemned carcasses than in passed carcasses was found in chicken fryers but not in chicken hens or turkeys. Although no single genus of bacteria in the liver was correlated with disposition, the presence of “one or more species” of bacteria in livers was significantly correlated with gross pathology sufficient to cause condemnation. Each of eight separate species of organisms in livers was significantly correlated with condemnation of the carcass for septicemia-toxemia or inflammatory process. Certain isolates were found in a significantly greater percent of normal livers, suggesting that these are “normal” flora and not associated with disease. Livers that were grossly normal were found to have significantly fewer histopathologic lesions than livers condemned for gross pathology, which, in turn, had fewer lesions than livers from condemned carcasses. No single histopathologic process in the liver was correlated with its condemnation, whereas the category of “one or more lesions” was so correlated. The presence of each of three histopathologic processes as well as “one or more lesions” in the liver was also significantly correlated with condemnation of the carcass. The presence of at least one or more species of bacteria was correlated with at least one or more forms of histopathology, but no specific species of bacteria was correlated with specific histopathologic lesions. Inspection failed to detect numerous livers with histopathologic lesions of varying degrees of unwholesomeness, and, conversely, resulted in condemnation of livers in which no histopathologic evidence of unwholesomeness was detected. Inspection did not detect livers harboring a variety of bacteria of unknown significance.

    Notizen: 1. The mechanisms involved in the fine adjustment of iris sphincter muscle tone are largely unknown. The aim of the present study was to clarify the effects of adrenomedullin on the resting tension of the bovine isolated iris sphincter muscle.2. The motor activity of the bovine isolated iris sphincter muscle was measured isometrically. The effects of adrenomedullin on resting tension were analysed in the presence of indomethacin. The presence of adrenomedullin mRNA in the preparation was determined by reverse transcription–polymerase chain reaction. Immunolabelling for adrenomedullin was also performed.3. Adrenomedullin significantly decreased the resting tension of the muscle. The relaxant effect of adrenomedullin was significantly inhibited by adrenomedullin (22–52), a putative antagonist for the adrenomedullin receptor, or calcitonin gene-related peptide (CGRP) (8–37), a putative antagonist for the CGRP1 receptor. The relaxant effect was almost completely blocked by a combination of adrenomedullin (22–52) and CGRP (8–37).4. The relaxant effect of adrenomedullin was also significantly diminished by 2′,5′-dideoxyadenosine, an inhibitor of adenylate cyclase, NG-nitro-l-arginine, an inhibitor of nitric oxide synthesis, or 1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-1-one, an inhibitor of soluble guanylate cyclase.5. Reverse transcription–polymerase chain reaction analysis showed that adrenomedullin mRNA was expressed in the muscle strip. Immunopositive staining for adrenomedullin was detected in blood vessel cells and in the iris sphincter muscle cells.6. These results suggest that adrenomedullin may be an autocrine and paracrine regulator of the resting tension of the iris sphincter muscle. Its biological effects may be due to the direct involvement of adrenomedullin receptors and also to the stimulation of CGRP1 receptors. The stimulation of these receptors by the peptide leads to the activation of adenylate cyclase and soluble guanylate cyclase and subsequent relaxation of the muscle strip.
