    Notes: —The effects of various cations and cholinergic ligands on the rate of α-bungarotoxin-acetylcholine receptor complex formation has been studied by means of a DEAE-cellulose disk assay technique. The reaction rate is reduced to one half the initial rate in the presence of 2·5 · 10−6m acetyleholine. a value close to the observed KD of ACh measured by equilibrium dialysis. Inhibition constants of about 5 mM were obtained for most monovalent cations whereas divalent cations gave inhibition constants of 0·05-0·2 mm. The rate and extent of toxin-receptor complex formation was also investigated as a function of hydrogen ion concentration; the rate of formation reaches a maximum at pH 7·5 and a group with a pK about 6 inhibits toxin binding to the receptor when protonated.These data can be correlated with the observed effects of inorganic cations on the binding of cholinergic ligands in vivo at the neuromuscular junction. Given the affinities of the individual cations, it is possible to predict how the apparent affinity of a cation-sensitive cholinergic ligand will change with variations of buffer composition.

    Notes: SUMMARY– A study was carried out with light and electron microscopic techniques to discover the effect of heating and carcass maturity upon the connective tissue and myofibrillar proteins of longissimus from veal (5–6 months), A (12–20 months) and D (54–60 months) maturities. Longissimus was heated to internal temperatures of 1) 50°C; 2) 60°C; 3) 70°C; 4) 80°C and 5) 90°C. Light microscopy indicated that connective tissue fibers increased in size and degree of aggregation with carcass maturity. Also, the more aggregated fibers from D maturity were more heat resistant than the less aggregated fibers of veal and A maturity. Endomysial connective tissue shrinkage was initiated at 50°C and it was completed at approximately 70°C. On the other hand, perimysial connective tissue shrinkage required internal temperatures of 70°C and higher before any significant fiber changes were observed. However, if endomysial and perimysial connective tissue fibers showed similar degrees of aggregation, they appeared to react similarly to heat regardless of maturity group. Electron and phase contrast microscopy showed that myofibrillar proteins compressed and sarcomeres shortened at 50°C. Heating to 60°C caused loss of M-line structure, initiation of disintegration and coagulation of thin and thick filaments and further myofibrillar protein shrinkage. Heating to 70 and 80°C caused more disintegration of thin filaments and coagulation of thick filaments. At 90°C, an amorphous structure resulted, but regardless of the internal temperature the principal banding features of the sarcomere could be identified. These changes are discussed in relationship to changes in meat tenderness.

Es fand sich ein in mehreren Gruppen reproduzierbarer signifikanter Anstieg, der nicht mit Änderungen von Hämatokrit, Serum Gesamteiweiß, Gewichtsbilanz, Blut- und Plasmavolumen korreliert war. Als mögliche Ursache des PB127I-Anstieges wird eine TSH-induzierte Mehrsekretion von Schilddrüsenhormon nach initialem Absinken von freiem und Gesamt-Thyroxin während der Peritonealdialyse diskutiert.
    Notes: Summary A significant increase of serum protein-bound (PB)127I was observed after 22 peritoneal dialyses in 16 patients with terminal renal failure. No correlation with changes in body weight, haematocrit, total serum protein nor blood or plasma volume was found.—The increase of PB127I during peritoneal dialysis may be ascribable to an initial fall of free and total thyroxine followed by a rise in TSH secretion.

    Notes: Abstract A standard procedure for the determination of the diamine oxidase (DAO) activity is described as a modification of the method ofOkuyama andKobyashi [2]. The principle of this method is the assay of14C-Δ 1-pyrroline and its polymers formed by the oxydative deamination of14C-putrescine in the presence of DAO. This assay was shown to possess a high sensitivity and precision. Now its accuracy could be enhanced by the comparison with the NADH test for DAO activity. This kind of a reference method made the direct correlation between cpm/min and deamination rates possible. Thus International Units (mU/sample) could be introduced into the isotope assay: In our test system 1 mU corresponded to 215 cpm/min, this value depending on the specific radioactivity of the substrate solution. The DAO activity from dog's small intestine was investigated with these two methods. The pH-optimum for this enzyme was 7.6, the K m for putrescine as substrate 1×10−4M , the optimum substrate concentration was 1×10−3M . In the small intestine of dogs and rabbits the DAO activity increased from proximal to distal parts, but in human beings the distribution of the enzyme in the gut did not show such a characteristic gradient. In the discussion, the advantages and disadvantages of the two tests for the determination of DAO activity were compared with each other. The isotope assay was the more sensitive and convenient test. But due to its absolute specificity the NADH test was useful as a reference method and for the investigation of the substrate specificity of the DAO in various tissues and body fluids.

    Description / Table of Contents: Summary Iteration methods are given for the solution of nonlinear equations in normed spaces requiring functionvalues and first-order divided differences. The latter may be replaced by derivations. The linear equations of each iteration step are solved only approximatively but this does not diminish the order of convergence.
