    Notizen: Abstract Hydrodynamic and oxygen transfer comparisons were made between two ring sparger locations, draft tube and annulus, in a concentric pilot scale airlift reactor with a baker's yeast suspension. Sectional hydrodynamic measurements were made and a mobile DOT probe was used to characterise the oxygen transfer performance through the individual sections of the reactor. The hydrodynamic performance of the reactor was improved by using a draft tube ring sparger rather than the annulus ring sparger. This was due to the influence of the ratio of the cross sectional area of the downcomer and riser (A D/AR) in conjunction with the effect of liquid velocity and a parameter,C 0, describing the distribution of the liquid velocity and gas holdup across the riser on the bubble coalescence rates. The mixing performance of the reactor was dominated by the frequency of the passage of the broth through the end sections of the reactor. An optimum liquid height above the draft tube, for liquid mixing was demonstrated, above which no further improvement in mixing occurred. The liquid velocity and degree of gas entrainment showed little dependency on top section size for both sparger configurations. Extreme dissolved oxygen heterogeneity was demonstrated around the vessel with both sparger configurations and was shown to be detrimental to the oxygen uptake rate of the baker's yeast. Dissolved oxygen tensions below 1% air saturation occurred along the length of the riser and then rose in the downcomer. The greater oxygen transfer rate in the downcomer than in the riser was caused by the combined effects of a larger slip velocity in the downcomer which enhancedk La and gas residence time, high downcomer gas holdup, and the change in bubble size distribution between the riser and downcomer. The position of greatest oxygen transfer rate in the downcomer was shown to be affected by the reactor from the influence on downcomer liquid linear velocity.

During part of the growth phase the DOT was cycled about the control value and the effect on growth and antibiotic production observed. In fermentations with cycling at 15 and 20% DOT the growth yields were lower than for the fermentations done at constant DOT throughout. There appears to be a complex interaction between growth rate and difficidin production rate which led to a reduced specific production rate at 15% DOT as a result of cycling.

    Notizen: Abstract A complex medium was used to investigate the effects of shear on the S. erythraea fermentation at 7-l scale. Maximum biomass was 11.1 ± 0.5 g l−1 at 1250 rpm (tip speed = 4.45 ms−1), whereas it was 12.7 ± 0.2 g l−1 at 350 rpm (tip speed = 1.07 ms−1). Specific erythromycin production was not stirrer speed dependent in the range of 350 to 1000 rpm and decreased by 10% at stirrer speed of 1250 rpm. Morphological measurements using image analysis showed that the major axis of the mycelia (both freely dispersed and clumps) decreased after the end of the rapid growth phase to a relatively constant value (equilibrium size) dependent on the stirrer speed. The mechanical properties of the cell wall were examined by disruption of fermentation broth in homogeniser and it was shown that mechanical strength of the cell wall increased in a large extent during deceleration phase.

However, the specific erythromycin production is virtually identical to that of a culture where the DOT did not fall below 65%. In addition, at constant DOT (10%) a stirrer speed of 750 rpm in a 7 litre causes mechanical damage to the mycelia in comparison with result at 500 rpm.

    Notizen: Abstract Proteolytic cleavage of recombinant human interferon-γ (IFN-γ) expressed in Chinese hamster ovary (CHO) cells during batch fermentation has been monitored by mass spectrometric peptide mapping. IFN-γ was purified from cell-free culture supernatant by immunoaffinity chromatography and cleaved by endoprotease Asp-N. Peptide fragments were resolved by reverse-phase HPLC and identified by a combination of matrix-assisted laser desorption/ionization time-of-flight (MALDI-TOF) mass spectrometry (MS) and automated N-terminal peptide sequencing. Using this approach, a peptide was identified as the C-terminal fragment of the IFN-γ polypeptide. Analysis of this peptide by MS indicated that the recombinant IFN-γ polypeptide secreted by CHO cells was truncated by at least ten amino acids, initially at Gln133-Met134. No full length (143 amino acids) polypeptide molecules were observed at any stages of the fermentation. Additional proteolytic cleavages at basic amino acids N-terminal of Gln133 occurred during the later stages of the culture resulting in a heterogeneous IFN-γ polypeptide population with 'ragged' C-termini.
