    Notes: Thin films of LiyV2O5 (0〈y〈1) were made by reactive dc magnetron sputtering of V followed by electrochemical treatment in LiClO4. X-ray diffractometry showed an orthorhombic structure that varied from nanocrystalline (n-X ) for unheated substrates to polycrystalline (p-X ) for substrates at 180 °C. Optical absorption was measured at 0.3〈λ〈2.5 μm. The semiconductor bandgap lay fixed at ∼0.5 μm for p-X LiyV2O5 and appeared blue-shifted in proportion with y for n-X Liy V2O5. At λ〉0.5 μm there are absorption peaks associated with V4+ ions both for p-X and n-X materials, and a broad absorption feature for p-X Liy V2O5 that is tentatively ascribed to electron hopping in the crystallographic b direction. The absorption goes up for increased y, as expected from the higher V4+ density.

    Notes: The MAX laboratory at Lund University, Sweden, today operates an accelerator system consisting of a 100 MeV racetrack microtron and a 550 MeV storage ring (MAX I). At the moment (July 1994) a new storage ring MAX II is near completion and will start first injections within 2 months. This work gives an overview of the MAX II project including the first beamlines and a description of the accelerator system. MAX II is a 1.5 GeV third generation light source optimized for the VUV and soft-x-ray region. It consists of a ten cell double bend achromat lattice forming the 90 m circumference ring. Injection is done at 500 MeV from the existing storage ring MAX I, and ramping up to full energy will take place in MAX II. The straight sections have a length of 3.2 m and eight sections are free to house insertion devices. At start up the ring will be equipped with one 7.5 T superconducting wiggler and one 1.8 T multipole wiggler. Two more undulators are ordered and under construction. To be able to achieve the project a few shortcuts have been made in the design of the storage ring: (1) Nonzero dispersion is allowed in the straight sections, (2) chromaticity correction is built into the quadrupole magnets, and (3) the length of the straight sections is limited to 3.2 m. The project is progressing on time. Extraction of an electron beam from the MAX I storage ring has been achieved and has successfully been transported into the MAX II building. The MAX II ring is under assembly with most of the sections already mounted. First injection is planned to take place in August 1994. © 1995 American Institute of Physics.

    Notes: The remodeling process in the lung of asthmatic patients results in a deposition of connective tissue and extracellular matrix components (ECM), which has been described as a subepithelial fibrosis. Fibroblasts, with the phenotypic appearance of myofibroblasts, are considered as the key source of the ECM in the fibrotic tissue. Furthermore, has the ECM molecule biglycan shown to correlate to the hyperactivity of the lung of asthmatic patients. In this study, we report the novel finding of a stretched fibroblast phenotype from bronchoalveolar lavage fluid (BALF) in 30% of the asthmatic subjects (n = 13). No fibroblasts were obtained in BALF from any of the nonasthmatic control subjects (n = 17). These BALF fibroblasts with several characteristics of a myofibroblast displayed increased migratory capacity, accompanied by an induced expression of the small GTPases RhoA and Rac1, when compared to myofibroblasts from corresponding lung biopsies. Data also shows that patients with BALF myofibroblast also have more cells in their lung tissue near to the basement membrane that stain for markers for mesechymal progenitor cells. Moreover, the BALF-myofibroblasts showed an increase in production of several types of proteoglycans like biglycan. By adding biglycan to normal fibrobalsts it is further possible to mimic the migratory type of myofibroblast found in BALF. To study differences in protein expression pattern between the two phenotypes, we used a gel-based proteomic technique in combination with MALDI-TOF mass spectrometry. BALF-myofibroblast displayed an increase in heat shock protein 20 (HSP20) and myofibroblasts from biopsies displayed an increase in chloride intracellular channel protein (CLIC) and cofilin. These proteins are all known to regulate cell migration by interacting with the actin cytoskeleton. In summery, data demonstrates that biglycan and mesenchymal progenitor cells might have an important role in the early -remodeling process in asthmatic patients and are potential future biomarkers for sub-epithelial fibrosis formation.

    Notes: We investigated whether the development of spontaneous T-cell-mediated type I diabetes in NOD mice is influenced by B cells and immunoglobulin (Ig). During the first 4 weeks of life. B-cell development was suppressed by repeated administration of rabbit anti-mouse IgM (RaIgM), while controls received polyclonal rabbit Ig (NRIg).A reduction in the incidence of diabetes, as well as in development of insulitis, was observed after either of these treatments. However, the effect on insulitis was more pronounced in mice treated with RaIgM compared with those treated with NRIg. Furthermore, while the optimal effect of NRig was obtained after a single injection at birth, the additional effect of RaIgM on development of insulitis was observed only after continued treatment for the first 4 weeks of life. Taken together these data suggest a possible role of Ig/B cells in the development of autoimmunity In the NOD mouse. The additional effect observed after continued suppression of the neonatal B-cell development suggests that this population may contribute significantly to the establishment of an auto-aggressive lymphocyte repertoire in the NOD mouse.

    Notes: High-mobility group box chromosomal protein 1 (HMGB1) is a protein with both intranuclear functions and extracellular cytokine-like effects. In this report, we study possible candidate receptors for HMGB1 on macrophages (Mφ) and define pathways activated by HMGB1 binding. Bone marrow Mφ were prepared from Dark Agouti (DA) rats and stimulated in vitro with HMGB1. The kinetics of tumour necrosis factor (TNF) production, NO production, activation of p38 mitogen-activated protein kinase (MAPK), p44/42 MAPK- and SAPK/JNK-signalling pathways, nuclear translocation of nuclear factor kappa B (NF-κB) and HMGB1-induced upregulation of major histocompatibility complex (MHC) class II and CD86 were analysed. Mφ from interleukin (IL)-1 receptor type I–/–, Toll-like receptor 2 (TLR2–/–) and RAGE–/– mice were used to investigate the role of these receptors in HMGB1 signalling. HMGB1 induced TNF and NO production by Mφ, phosphorylation of all investigated MAP kinase pathways and NF-κB translocation, and expression of MHC class II was increased. Mφ from RAGE–/– mice produced significantly lower amounts of TNF, IL-1β and IL-6, while IL-1RI–/– and TLR2–/– Mφ produced cytokine levels comparable with wildtype controls in response to HMGB1 stimulation. We conclude that HMGB1 has the potential to induce a proinflammatory phenotype in Mφ, with RAGE as the major activation-inducing receptor.
