    Notizen: A model is proposed that accounts for the decreases in yield which occur in chemostat cultures of mesophilic yeasts at superoptimal growth temperatures. Two yield depressing effects were identified, one due to increased maintenance requirements by the viable fraction of the population, the other due to energy substrate dissipation by the nonviable fraction. The two effects are functions of the dilution rate, as is the fraction of nonviable cells. Experimental results were obtained on the yield, maintenance, and dissipation of energy substrate in a glucose-limited chemostat culture of a respiration-deficient mutant of Saccharomyces cerevisiae at 39°C. The rates of glucose utilization for maintenance and for dissipation constituted, respectively, 33-28% and 15-9% of the total glucose utilization rate over the range of dilution rates tested (0.038-0.064 hr-1), while the yield varied over this range from 0.066-0.085 g of biomass (dry wt) per gram of glucose.

    Notizen: The models of Monod and Williams, for the growth of unicellular organisms in chemostats, give strongly damped transients in the biomass and cell number when the flow rate of the chemostat is changed. A simple trick is used to incorporate time delay in these models while still allowing a conventional stability analysis. For long enough time delays the equilibrium point is unstable and limit cycles can be computed. Results obtained using Williams' model, with weakly damped transients as a result of using moderately long time delay, are compared with his data in which cell numbers show weak damping but biomass shows strong damping.

    Notizen: An efficient method to grow Escherichia coli W to high cell concentrations on the pilot scale is described and discussed. The method involves growth linked introduction of glucose; and ammonia to the culture, sparing with oxygen, and maintenance of aerobic conditions by gradually decreasing the temperature in the culture in order to keep the oxygen demand within the limits of the capacity of supply. Under these conditions the linear rate of cell mass production is actually the result of exponential growth with a gradually decreasing growth-rate constant.About 10 kg packed cells were produced in a 50 liter working-volume fermentor in one run of 13 hr. The concentration of the cells at the end of the growth was about 47 g dry cells/liter. The expenditure for nutrients was minimal and the controls were of simple automatic nature. From the determined yield constants for glucose, nitrogen, phosphorus, and oxygen it may be inferred that the cells grown by this method are similar to those grown exponentially at constant temperature.

    Notizen: An analytical expression for the rate efficiency factor of planar localized enzyme systems is derived. The derivation takes into account the isothermal kinetic effect under the externally imposed perturbation of combined electrostatic and high frequency time-varying fields. The contribution of each individual field to the enzyme reaction is examined through the basic mechanism in which charged substrates interact, with the specific perturbing field. The interaction mechanisms for the electrostatic and for the time-varying fields are found to be different. This difference regulates the different manners in which enzymatic reaction rates are altered. Enzymatic reactions under electrostatic perturbation can be retarded or enhanced depending on the field polarization. At sufficiently high field intensities the reaction rate may approach zero or approach a maximum value equal to the turnover number of the enzyme. Time-varying field perturbations, on the other hand, always enhance the enzymatic reactions if bunching effects are negligible. At sufficiently high field intensities, the reaction may approach a value equal to that of the free enzyme system. Several typical numerical examples on pure electrostatic field perturbations, pure time-varying field perturbations, and combined field perturbations are also presented.

    Notizen: The pressure required for initiation of flow when freeze-pressing with the X-press is related to the phase boundaries of water, particularly those between ice I and liquid even at temperatures around -25°C and lower. Widening the orifice of the pressure chamber to diameters larger than 2.5 mm leads to lower pressures and less extensive cell disintegration.Pressing Saccharomyces cerevisiae slowly with the aid of a manual hydraulic jack at -25°C produces a disintegration of 60-75% irrespective of cell concentration. Pressing at -35°C shows no clear differences.Pressing more rapidly with the aid of a motor-driven hydraulic press produces a similar extent of disruption of diluted cell suspensions (5.4 mg/g) as slow pressing. However, freeze-pressing a paste of baker's yeast (270 mg/g) increases the degree of disintegration. Under these conditions the disintegration is further enhanced by a lower temperature, -35°C, and by a high velocity of flow through the orifice, such that more than 95% of the S. cerevisiae is disrupted by one pressing at less than 2 × 108 Pa.Mechanisms for flow through the X-press are suggested and discussed in relation to the phase diagram of water.

    Notizen: The characteristic flavor of hard Italian cheeses is associated with the presence of fatty acids, particularly butyric acid, liberated from milk fat during the ripening process. To ensure proper development and control of flavor, animal pregastric esterases or lipases are routinely added to the milk before coagulation of the curd. Such esterases are also used to generate flavor in enzyme modified cheese and other dairy products.Esterases from microbial sources have been investigated as agents to enhance flavor in cheese. We have found that an esterase from Mucor miehei exhibits the type of lipolytic activity needed for this application. Romano and fontina cheeses of excellent quality have been prepared by the use of this esterase. It has also been used successfully in the preparation of enzyme modified cheese, and, in turn, processed American cheese.

A third example is presented to illustrate the use of a chelating agent in growing corn to increase the sugar content by over 50%, presumably by influencing enzyme activity in the corncob itself.     Notizen: Over 130 volatile components have been identified in aromas of film (F), submerged-culture flor (S), and baked (B) sherries. The composition of these volatiles, which include more than 25 alcohols, five carbonyls, ten acetals, three amides, 35 esters, and five lactones, is influenced by the variety of grape used, the nature of the primary alcoholic fermentation, type of cooperage, aging and blending system, and filmphase yeast metabolism in (F) and (S) or reactions occurring during “baking” in (B). The influence of these factors on the aroma and several volatile formation mechanisms which contribute to the characteristic flavor of sherry, particularly during the film growth and during baking, are discussed.

    Notizen: A cephalosporin acetylesterase produced by Bacillus subtilis catalyzes the deacetylation of 7-aminocephalosporanic acid (7-ACA). Previous reports from our laboratory described the kinetic constants that characterize the reaction: Km = 2.8 × 10-3M, Kia acetate = 5 × 10-2M, and Kid deacetyl-7-ACA = 3.6 × 10-2M. These constants were used to predict the time course of the reaction using the following equation for dual competitive product inhibition. \documentclass{article}\pagestyle{empty}\begin{document}$$ \frac{{dS_t}}{{dt}} = \frac{{- V_{\max}}}{{1 + \left({K_m /S_t} \right)\left({1 + A_t /K_{{\rm ia}} + D_t /K_{{\rm id}}} \right)}} $$\end{document} where St = mg/ml 7-ACA, At = mg/ml acetate, Dt = mg/ml deacetyl-7-ACA. The predicted time course closely matched the time course measured experimentally. The equation also was solved without the inhibition terms and the solution indicated that product inhibition caused about a 30% increase in the time required for complete (〉97%) hydrolysis of a 24 mg/ml 7-ACA solution. The esterase was immobilized by containment within an ultrafiltration device. With this technique the enzyme was reused 20 times over an 11 day span to deacetylate 7-ACA solutions containing 4 to 24 mg/ml 7-ACA. The specific activity after the 20th use was the same as the activity prior to the first use, indicating little enzyme inactivation occurred.

    Notizen: The design of a continuous multistage tower fermentor is described. The fermentor consists of five stages separated by perforated plates. Each stage includes mechanical mixing provided by two disc turbine impellers and has its own impeller shaft with bearing assembly and flexible coupling that enables the operation of an arbitrary number of stages. The normal operation of this system enables the co-current flow of gas and liquid, but the system can function countercurrently as well.The purpose of this study was to examine the hydrodynamic performance, i.e., the pressure gradient along the tower, the mixing time, gas holdup, the residence lime distribution of the continuous phase, the value of the backflow coefficient, and the oxygen transfer rate under conditions usually used during fermentations. From the interrelations between parameters influencing the proper performance of this system, an optimal design of plate geometry for processes requiring high oxygen transfer rate was formulated.

    Notizen: Flow system technology enables the biological and medical experimenter to investigate the distribution spectra of various cellular characteristics separately or in parameter combination on the basis of ultrarapid single cell measurements. A typical rate of recognition is about 1000 to 5000 cells per second and the precision of measurements and their statistical relevance has been previously unobtainable. According to the approach of the multiparameter analysis and high data rate, computer assistance in flow system technology is given primary consideration. In this study three different kinds of software controlled modes in data acquisition are demonstrated: normal acquisition and linear accumulation of single parameters, spectra accumulation of two correlated parameters of each single cell and documentation as linear, two- or three-dimensional distribution pattern, and linear accumulation of two correlated parameters simultaneously with their actual signal-to-signal ratio. A first attempt to analyze distribution spectra was the application of the entropy of the structure routinely used in cybernetics. This function seems to be a measure for determining the degree of synchrony in an appropriate pretreated cell population. A special mathematical strategy has been applied to the linear spectra of cellular DNA content, whose advantage is the quantitative extraction of the fractions concerning the various phases of the life cycle cells. The validity of this special curve fitting procedure has been proven on various experimental cell populations.

    Notizen: The biosynthesis of invertase by Saccharomyces carlsbergensis LAM 1068 was studied in relation to its glucose effect at both unsteady and steady states of growth. Experimental correlations between the dilution rate and invertase specific activity (E/X) in chemostat, cultures led to an optimum for the enzyme synthesis at a particular intermediate growth rate. The value of E/X increased from 1.1 (U/mg biomass) in batch cultures to 13 (U/mg biomass) in chemostat cultures. A mutant strain A3 showed the highest value for E/X = 25 (U/mg biomass) at high dilution rates where glucose repression was observed with the wild strain.

    Notizen: Five marine phytoplankton species (Phaeodactylum tricornutum, Thalassiosira pseudonana, Skeletonema costatum, Monochrysis lutheri, and Dunaliella tertiolecta) were grown in enriched laboratory continuous cultures and natural populations were mass cultured outdoors for 16 months. Competition among the species was shown to be highly dependent on temperature, although the actual production of plant organic matter at the low growth rates used was relatively independent of this variable. Control of marine species in mass cultures does not appear economically feasible, but this drawback may be overcome by selecting herbivorous shellfish that are capable of assimilating those temperature-dependent phytoplankton species dominating in a particular locale.

    Notizen: Hexyl-groups have been introduced into crosslinked Sepharose 6B, yielding gels with degrees of substitution which range from 0.02 to 0.70 mol hexyl-side chain per mole galactose residue.The gels were exposed to β-amylase in solution, and the resulting adsorbates indicated a monotonic increase in adsorption capacity with an increasing hexyl-content. Adsorbate activity, by contrast, displayed a maximum for a carrier gel with a hexyl-galactose ratio of 0.51. Adsorbates based on gels with different hexyl-content were used in column reactors for continuous maltose production from a soluble starch substrate.

    Notizen: Amyloglucosidase from Aspergillus niger (α-1,4 and 1,6 glucan glucohydrolase, EC 3.2.1.3) was immobilized through adsorption onto a hexyl-Sepharose, containing 0.51 mol hexyl-group per mole of galactose.The adsorption limit of the carrier with respect to this enzyme was about 17 mg per gram wet conjugate. The retention of activity upon immobilization was high, varying from essentially full activity at low enzyme content down to 68% at the adsorption limit.The immobilized preparation, as well as the soluble enzyme, showed apparent zero order kinetics within 60% of the substrate's conversion limit. Product inhibition of the soluble enzyme showed a k1 of 5 · 10-2M.In the presence of 3M NaCl, adsorbates were formed more rapidly and with a higher yield of immobilized protein, but with lower specific activity. Conjugates resulting from adsorption of amyloglucosidase in identical concentrations, but at different salt contents, showed comparable activities and operational stabilities. Continuous operation for three months reduced conjugate activity to 40%.The thermal stability of the adsorbate was inferior to that of the soluble enzyme, but was noticeably enhanced in the presence of substrate.

    Notizen: Transient experiments were conducted on a Pseudomomas utilizing phenol in a continuous culture by disturbing the influent substrate concentration and dilution rate. Two stable steady states existed for some ranges of the parameters. Highly damped oscillations were observed in approaching a new high conversion steady state or in returning to a new high conversion steady state following a small disturbance. When a large disturbance was applied there was a smooth (overdamped) approach to a new low conversion steady state.The observed oscillatory behavior for small disturbances was predicted by a modified Powell-Ierusalemskii bottleneck model, but could not be predicted by a Monod-Haldane model; neither model was accurate for predicting the effect of large disturbances.A constant wall growth factor was used to account for microbial film activity, and the existence of two stable states was directly due to the presence of the film.

    Notizen: This analytical study deals with the steady-state behavior and control of microbial growth in continuous cultures. A second order Haldane-Monod model of continuous cultures is used as a basis for study of the effects of the adjustment of pH by the addition of acidic (or basic) materials. The treatment of a hydrogen ion concentration, in addition to substrate and microbial concentrations as state variables, results in a third order system of equations describing the process.The analysis of the system in equilibrium yields several admissible steady states, that is, steady states which satisfy all constraints. An optimal control problem is formulated and subsequently solved to maximize steady-state microbial production.

    Notizen: A soluble-insoluble form of nicotinamide adenine dinucleotide (NAD+), which can be rendered either soluble or insoluble by simply adjusting the pH, has been prepared by covalently coupling NAD to alginic acid using 1,2,7,8-diepoxyoctane. The NAD bound to the alginic acid showed the coenzymic function in the soluble state and could be collected for further use as a precipitate by lowering the pH to below 3. Coupling soluble-insoluble coenzymes with insolubilized apoenzymes is possible in fluidized and fixed-bed reactors.

    Notizen: Glucoamylase was immobilized to porous silica and its kinetics and stability were observed with acid- and α-amylase-hydrolyzed dextrin as feed. The enzyme was found to be extremely stable in both laboratory and pilot plant operations. When the feed had been previously only lightly hydrolyzed, pore diffusion limitation caused appreciable decreases in glucose production rate. The severity of starch hydrolysis to dextrin markedly affected ultimate glucose yields. The diffusional gradients present in the carrier pores caused the immobilized enzyme to yield lower glucose concentrations than the free enzyme at similar feed conditions.

    Notizen: There are four main sources of enzymes in foods - these being the inherent enzymes, enzymes from microbial contaminants, enzymes elaborated by microorganisms added to foods, and specific enzymes added to foods. This study primarily deals with the latter two sources of enzymes in food. Although both plants and animals serve as sources of enzymes, they are not as economical or versatile sources as are enzymes obtained from microorganisms. In the meat industry, proteases are used to tenderize muscle and to obtain flavor precursors. In the preparation of cured meat products such as sausages, lipases, and proteases from bacterial cultures are utilized. Similarly, proteases and lipases are used in the dairy industry to develop flavor compounds. Proteases and amylases also have applications in the baking and milling industries where they are used to produce precursors for the nonenzymatic browning reactions. Carbohydrases such as amylase, amyloglucosidase, and glucose isomerase have found usage in the starch and syrup industry for the production of high dextrose and high fructose syrups. Other enzymes such as glucose oxidase, pectinase, and naringinase are of value to the wine and fruit juice industries. A better understanding of the mode of action of enzymes as well as the mechanisms of development of flavor compounds will further enhance the use of microbial enzymes to develop specific and desired flavors in foods.

    Notizen: The development of the unique flavor of blue type cheese depends on the concerted action of numerous enzymes of Penicillium roqueforti involved in protein and lipid metabolism. Protease(s) by degrading casein modify the texture and background flavor of the ripening cheese. Lipase by hydrolyzing milk triglycerides provides flavorful fatty acids and precursors of methyl ketones. The enzyme complex involved in the partial oxidation of free fatty acids and the properties of β-ketoacyl decarboxylase which generates the major flavor components of blue cheese are discussed. Fermentation of P. roqueforti for the rapid production of methyl ketones is briefly reviewed.

    Notizen: A method for the measurement of the affinity of the cell surface to hydrocarbons is described. The affinity was basically unaffected by different pH values and temperature as well as by the chain length of the substrate. The contact time required for saturation of the cell surface with substrate was 30 sec. Cells grown on glucose showed a 25% lower adsorption capacity compared to those grown on n-alkane. The glucose grown cells showed also a more distinguished dependence of the amount of adsorbed hydrocarbon on the quality of the emulsion. The interaction between the substrate and cell surface turned out to be an adsorption that did not involve an enzymatic reaction. These results led to the conclusion that a lipopolysaccharide moiety present at the cell surface is responsible for the affinity.

    Notizen: The “malty” flavor defect that commonly develops in raw milk produced in certain areas of North America and Europe has long been known to be due to the metabolic activity of Streptococcus lactis var. maltigenes. The identification of the aldehydes and alcohols responsible for this flavor defect and the mechanisms involved in their formation from amino acids are discussed.Pseudomonas fragi, a common psychrophilic recontaminant, is responsible for development of “fruity” flavors in processed dairy products by virtue of the organisms ability to hydrolyze milk fat and esterify certain of the lower fatty acids with ethanol. A similar esterase is present in certain lactic cultures used in the manufacture of cheddar cheese.The “musty potato” aroma first described in eggs and milk and other dairy products due to the growth of Pseudomonas graveolens (Pseudomonas taetrolens) continues to be reported as a defect in eggs and carcass meats. Pseudomonas perolens has been found to produce a similar aroma in spoiling fish. Vapors entrained from milk and fish tissue cultures of these organisms, collected on porous polymer traps and analyzed by GLC-alkali flame and GLC-MS systems, revealed both organisms produce 2-methoxy-3-alkylpyrazines. 2-Methyoxy-3-isopropylpyrazine was found to be responsible for the musty potato aroma. A possible mechanism for the formation of pyrazines is discussed.

    Notizen: An analysis of the pore diffusion model involving a two-substrate enzymatic reaction is presented. The resulting equations have been applied to the case of galactose oxidase catalyzed oxidation of galactose when the enzyme is immobilized on porous glass particles. The physical constants of the system were obtained by theoretical predictions and the enzyme concentration in the porous medium was derived from the experimental results. The calculations were performed with the assumption that the kinetic parameters of the enzyme remain unchanged upon immobilization. The theoretically calculated effectiveness factors were compared with the experimental effectiveness factors determined from the batch kinetic experiments and were found to be in agreement. The results are presented as effectiveness factor plots graphed as functions of bulk galactose and oxygen concentrations. The model was extended in order to study the effect of external mass transfer coefficients and pore enzyme concentrations on the effectiveness factors.

    Notizen: A mathematical model is suggested for growth of microorganisms under limitation by “conservative” substrates such as inorganic ions or vitamins that are not broken down after uptake into the cells, but that wholely or partly remain available for production of biomass. The specific growth rate is expressed here as a function of the intracellular “concentration” of the limiting substrate, defined as the amount of substrate within the cells per unit of cell dry weight.In the model, the intracellular substrate is divided into two parts. One part is a “structural” substrate not available for further growth. The other part is an “excess” or “functional” substrate that is used for biomass production and is assumed to be converted into structural substrate proportionally to growth. The rate of growth is believed to be controlled by the intracellular concentration of excess substrate.

    Notizen: A conceptual model of aerobic microbial growth in semisolid matrices was developed as a first step in the prediction of the rate of breakdown of semisolid cellulosic material. The conceptual model was described by a series of equations simplified by the assumption of steady-state microbial activity, and heat and mass transfer limitation. Temperature and oxygen distribution in compost piles were measured experimentally at the Butler County Mushroom Farm, Butler County, Pennsylvania, to test the validity of these assumptions. The compost piles consisted of ground corn husks, straw, and race horse manure. The data fit with the model was excellent with deviations between model predictions (as solved by an analog computer) and actual temperature measurements never exceeding 3°C. The effects of compost pile geometry, external temperature, compost density, external oxygen concentration, and insulation at the bottom of the pile were then predicted using a digital computer to solve the model. The predictions show that the maximum breakdown rate occurs for an optimum height (which depends upon the system), insulating the base increases the breakdown rate, increasing the external temperature increases the initial breakdown rate but decreases the pseudo-steady-state breakdown rate and the uniformity, and any increase in the external oxygen concentration increases the breakdown rate but decreases the uniformity.

    Notizen: Shellfish processing wastewaters were characterized and subjected to laboratory-scale treatability investigations using continuous flow chemostats operated under both aerobic and anaerobic conditions with and without biological solids recycle. System operation and evaluation employed hydraulic retention time as the controlled variable with substrate removal and biomass production as dependent variables. Additional variables specific to anaerobic operation included total and methane gas production rate and volatile acids generation. Application of a kinetic model incorporating the Monod substrate concentration- growth rate function allowed estimation of kinetic coefficients μmax, kd, Y, and Ks.Aerobic treatment of the wastewaters could be adequately described by the kinetic model presented. However, total efficiencies were notably affected by the inherent morphological differences induced by changing hydraulic flows as reflected by poor solids separation at lower and higher retention times. Anaerobic treatment, although not amenable to kinetic description by the model and data presented, exhibited a limited potential for wastewater stabilization in that soluble substrate removals and methane production occurred at all retention times. Difficulties in successfully describing the system kinetically stemmed from an inability to quantify influent solids liquifaction rates. Moreover, evidence of some substrate associated inhibition of methanogenesis was apparent. Solids recycle tended to reduce the inhibitory influences and to increase treatment efficiencies in both the aerobic and anaerobic processes.

    Notizen: A mathematical model was developed for percent removal of a pure, non-adsorbable, biodegradable substrate in a submerged biological filter using the nonlinear Monod expression for the substrate utilization rate. Analytical solutions were obtained using zero and first order linearizations of the Monod expression and were compared to the numerical solution of the nonlinear system. The relevant dimensionless parameter groups describing the system were identified and their effects on substrate flux into the biolayer, effectiveness factor, and percent removal were also examined. Criteria were established for determining the relative accuracy of the two biokinetic approximations and for determining values of “limiting” biolayer thickness.There are two major results of engineering significance. (1) The approximation of the nonlinear Monod expression by a first order rate expression yields accurate results for a much wider range of bulk concentrations than might be expected from inspection of the Monod half-velocity coefficient for a given substrate. (2) Percent removal of substrate is a weak function of biolayer thickness (and thus total cell mass) when the thickness is greater than a rather small limiting value but is a strong function of particle radius (and thus biolayer surface area) for a wide range of parameter values including those of common interest.

    Notizen: Reactor systems based on tapered fluidized beds are being developed for aqueous bioprocesses in which adhering microorganisms or immobilized active biological fractions are used. The use of a fluidized bed prevents biomass buildup, accommodates particulates in the feed stream, is compatible with gas sparging, and allows easy removal or addition of the active materials. The tapered reactor tends to stabilize the fluidized bed, thus allowing a much wider range of operating conditions. Preliminary experimental results and an empirical mathematical model of the tapered bed indicate that bed stability is associated with a decreasing velocity and void-fraction profile up the bed and the pressure drop across the bed decreases with increasing flow rates.The tapered fluidized bed bioreactor is being evaluated for use in the enzymatic production of hydrogen, microbiological denitrification, and microbiological degradation of coal conversion aqueous waste streams. The enzyme catalyzed conversion of lactose to glucose and galactose was used in the evaluation of the reactor concept.

    Notizen: The immobilization of Rhodospirillum rubrum chromatophores was successfully performed by entrapping them in polyacrylamide. Their photophosphorylating activity was about 40% of native chromatophores. The temperature and pH optima for immobilized chromatophores were similar to the native photosynthetic apparatus and kinetic parameters showed that the rate of photophosphorylation in polyacrylamide particles was diffusion controlled. Light penetration of the gel particles was not a limiting parameter. Immobilization considerably increased the stability of the chromatophores towards denaturation.

    Notizen: The construction and use of an automated membrane filtration system is described which produces 300 liters per day of sterile medium over at least four months of continuous fermentation. The capacity of the system is limited only by choice of filter type and has applications in continuous pilot plant work or medium-scale batch fermentation, in the bulk supply of communally used laboratory medium, in supplying banks of small-scale continuous fermentors, and for remote control of medium supply in hazardous fermentations (e.g., methane) with great savings in time and materials. Only two sterile storage jars are needed, irrespective of the total demand for the medium.

    Notizen: The response of a polarographic oxygen electrode to a step change and to an exponential change in bulk oxygen concentration was studied theoretically and experimentally for the case where there is a significant liquid film resistance at the outer side of the membrane-covered electrode. The probe response has been described by a mathematical model which takes into account the oxygen mass transfer through the liquid film and through the membrane. It is shown that substantial error could be introduced by neglecting the liquid film resistance. A test procedure is suggested to evaluate the error significance.

    Notizen: Sweet potato β-amylase (α-1,4 glucan maltohydrolase, EC 3.2.1.2) was immobilized through adsorption onto an agarose gel to which nonpolar side chains had been introduced via ether bridges.The adsorbent showed evidence of saturation at an enzyme content of 35 mg per milliliter of packed gel. The adsorption was rapid and yielded a product whose operational stability depended on the initial content of β-amylase. Activity leakage was low. The relative activity of immobilized enzyme was inversely related to the amount of enzyme adsorbed to a given gel volume, having a maximal value of around 50% at low enzyme contents.

    Notizen: Several bacteria utilizing C1-compounds as sole carbon sources were grown on these substrates in continuous culture. The molar yield values (g of cell dry wt/mol of substrate utilized) of bacteria which utilize C1-compounds via the ribulose monophosphate pathway were between 15.7 to 17.3 when grown on methanol; while the molar yield values of bacteria which use the serine pathway for the assimilation of C1-compounds varied between 9.8 and 13.1. The molar yield values of different bacteria which use the serine pathway decreased as the oxidation levels of the C1-growth substrates increased. On formaldehyde the values were between 7.2 to 9.6, whereas on formate the values varied from 3.3 to 6.9.It appears that bacteria utilize Cl-compounds more efficiently via the ribulose monophosphate pathway than via the serine pathway. The oxidation step from methanol to formaldehyde (and from methylamine to formaldehyde) in the bacteria studied may be energy yielding. A comparison has been made between the experimental yield values obtained and theoretical values.

    Notizen: The black yeast-like fungus Rhinocladiella mansonii NRRL Y-6272 elaborates an extracellular polysaccharide composed of N-acetyl D-glucosamine and N-acetyl D-glucosaminuronic acid residues in a 2:1 molar ratio. Production of this polymer was studied in 300 ml flasks and 20 liter fermentors. On the basis of glucose utilized, 13% conversion to polymer was obtained in four days at 25°C in medium containing yeast autolysate paste, K2HPO4, MgSO4, ZnSO4, L-asparagine or urea, and D-glucose. Nitrogen sources, aeration, agitation, temperature, and salts affected yields.The polymer in diluted fermentation broths was precipitated with ethanol in the presence of an electrolyte and collected by centrifugation. The viscosities of aqueous resolutions containing 1 and 1.5% of the polymer were 9400 cP and 21,500 cP, respectively.

    Notizen: Galactose oxidase (E.G. 1.1.3.9) was covalently immobilized to chemically modified porous silica particles by reaction of the native enzyme with pendant benzoyl azide groups on the carrier. The enzyme loading on the carrier was 100-150 units per milliliter. The immobilized enzyme was incorporated into a hardware assembly suitable for the determination of galactose or lactose concentrations in complex biological fluids. The prototype instrument as described is suitable for continuous, on-line monitoring or discrete sample analysis. Reaction conditions can be readily provided which maintain global first order kinetics within the reactor and strict linearity of the procedure over a wide range of sample concentrations. Auto-inactivation of the immobilized enzyme can be prevented by K3Fe(CN)6 and long-term reactor stability can be achieved by the periodic application of the reagent to the enzyme reactor in situ.

    Notizen: For better operational control of the completely mixed oxygen activated sludge process (CMOAS), a study concerning the kinetics, performance, and operational stability of the Ramanathan-Gaudy model was conducted. Short-term experiments were conducted at various dilution rates (1/9, 1/6, 1/3, 1/1.5, and 1/1.0 hr-1) by using two recycle solids concentration values (5000 and 10,000 mg/liter). The influent substrate was an actual industrial organic wastewater (soft drink waste) and its concentration was maintained at 1000 mg/liter COD. The hydraulic recycle ratio, α, was maintained at 0.30.It was found that for CMOAS system with constant recycle cell concentration, a “steady state” with respect to reactor biological solids and effluent COD at different dilution rates could be attained. No appreciable dilute-out of reactor biological solids and substrate was observed up to the dilution rate of 1 hr-1 for both systems of different XR (5000 and 10,000 mg/liter). For the system of XR = 5000 mg/liter, except the dilution rate of hr-1, the effluent filtrate COD was lower than 100 mg/liter, the aerator biological solids concentration was about 1550 mg/liter, and the COD removal efficiency was higher than 90% for all dilution rates. For the system of XR = 10,000 mg/liter, the effluent filtrate COD was lower than 71 mg/liter, the aerator biological solids concentration was about 2750 mg/liter, and the COD removal efficiency was higher than 90% throughout all the dilution rates selection in the present study. The value of the Sludge Volume Index (SVI) was the range of 37.0 to 58.5 and provided good settleability of sludge. The sludge yield was 0.53 for the system of XR = 5000 mg/liter and 0.57 for the system of XR = 10,000 mg/liter. The carbohydrate and the protein content of the cells were 10.1-21.6% and 35.6-50.6%, respectively.For predicting the reactor biological solid and effluent COD of the CMOAS system by using the Ramanathan-Gaudy model, two sets of values for the biological kinetic constants should be considered since it provided the best fit of predicted values of the observed values. In the present study, μm = 0.4 hr-1, ks = 92 mg/liter for 1/3 ≤ D ≤ 1, and μm = 0.05 hr-1, ks = 11.1 mg/liter for 1/9 ≤ D 〈 1/3 were used to calculate the predicted values of reactor biological solid and effluent filtrate COD.

    Notizen: A new device is designed that is suitable for both laboratory- and industrial-scale cultivation of microorganisms, particularly filamentous fungi, on the surface of liquid media. The device reliably ensures the sterility of cultivation, regardless of its duration. Its installation costs are relatively low and maximum production is reached per unit of installation space. In comparison with submerged cultures in fermentors, the proposed mode does not use a high consumption of energy for stirring and aeration (consequently, the duration of cultivation is not an economically relevant factor) and does not require maintenance of complicated equipment. Due to the building and construction of the technological equipment, the production plant is capable of very fast expansion and, when the need arises of very fast and economical liquidation.

    Notizen: Due to the loss of enzymatic activity as a function of time, an alkaline protease, selected for the continuous preparation of protein hydrolysates (J. Boudrant and C. Cheftel, Biotechnol. Bioeng., 18, 1735, 1976), was chemically stabilized by a simple treatment with glutaraldehyde. Two fractions, soluble and insoluble, were obtained. The activities of these two fractions were measured with casein and N-benzoyl-L-arginine ethyl ester (BAEE) as a function of glutaraldehyde concentration used. It was noted that the insoluble fraction was practically inactive with the first substrate and that the heat stability of the soluble form was likewise enhanced. Molecular weights of these two forms were unchanged, but the uv-spectrum of the soluble form was modified. From amino acid analysis, it appears that this treatment mainly provokes a decrease in lysine content.

    Notizen: Batch proteolysis experiments were performed in order to choose a protein-protease system to prepare a correct hydrolysate suitable for the enrichment of soft-drinks. The system eventually studied was casein-Alcalase.Comparative batch and continuous proteolysis of casein by Alcalase showed that the reaction, which does not exactly follow first order kinetics with respect to the substrate concentration, is inhibited by the reaction products. Furthermore, experiments were done in order to determine the reaction conditions (pH 8.8 in the reactor, casein concentration 5%, 40°C). Determining the molecular weight of Alcalase (43,000) suggested the choice of ultrafiltration membrane PM 30.Studies of continuous proteolysis with the chemically stabilized enzyme retained by the ultrafiltration reactor showed that protease reuse for seven days at 40°C is possible and that the growth of microorganisms is practically inhibited under these conditions. Gel chromatography showed the molecular weight range of the peptides to be less than 2,000. Triangular taste tests showed that the threshold identification concentration of the dry hydrolysate in orange juice is about 0.65%.

    Notizen: The apparent viscosity of baker's yeast suspensions was employed to control the cell concentration of the mixing of baker's yeast and distilled water. The apparent viscosity of baker's yeast was found to be a function of cell concentration and independent of the flow rate through the rotational viscometer. The viscometer and mixing unit were found to have first order dynamics. The open loop dynamics of the control system were nonlinear but could be linearized for small perturbations. The control system was employed to successfully control the cell concentration but exhibited nonlinear behavior.
