On Amino Acids and Peptides, X. Synthesis of 2,5-Cyclohexadiene-1-alanine (1,4-Dihydrophenylalanine) from 2,5-Cyclohexadien-1-ylmethylmagnesium Chloride2,5-Cyclohexadiene-1-acetaldehyde (1) is synthesized in high yield from 2.5-cyclohexadien-1-ylmethylmagnesium chloride. Ugi reaction leads to the formation of N-trifluoroacetyl-2,5-cyclohexadiene-1-alanine methylamide (2) which on saponification yields 2,5-cyclohexadiene-1 -alanine methylamide (1,4-dihydrophenylalanine methylamide, 3).
Notes:
2,5-Cyclohexadien-1-acetaldehyd (1) wird aus 2,5-Cyclohexadien-1-ylmethylmagnesiumchlorid in guter Ausbeute hergestellt. Ugi-Reaktion führt zu N-Trifluoracetyl-2,5-cyclohexadien-1-alanin-methylamid (2). Dessen Verseifung ergibt 2,5-Cyclohexadien-1-alanin-methyl-amid (1.4-Dihydrophenylalanin-methylamid, 3).
