Notes: Summary The intestinalβ-galactosidases in suction biopsy material of 4 lactose-tolerant adult Europeans were separated after solubilization with Triton X-100 in a linear 5–20% mannitol gradient by ultracentrifugation at 4°C and 44000 rpm. After a centrifugation time of 12 h 3 fractions were identified. The 2 fractions which sedimented more rapidly were active with lactose as substrate; all 3 fractions hydrolized p-nitrophenyl-β-galactoside. The 3 isolatedβ-galactosidases probably correspond to the “neutral” brush border lactase, the “acid” lysosomal lactase, and a cytoplasmatic hetero-β-galactosidase.
