Summary A DNA linker with TAA translational stop codons in all three reading frames was inserted into the polylinker region of pUC12. The new plasmid pUC12-STOP is useful for the expression of DNA in cases where defined translational stops are desired. The STOP linker is flanked by unique restriction sites and thus can be excised as ‘portable STOP linker fragments’. The STOP linker was used to express in Escherichia coli a truncated form of the Herpes simplex virus type 1 glycoprotein D antigen.
