    Notes: Preferential transport of phosphate through macropores increases the significance of phosphate sorption to macropore wall materials compared with bulk soil materials. Therefore we studied the kinetics of phosphate sorption to soil bulk materials from the Ap and Btg horizons, from the iron oxide-poor (Albic) centres and from iron oxide-rich (Red) exteriors of the fractures in a clayey pseudogley in batch at initial phosphate concentrations (P0) up to a maximum of 650 μm and at sampling times up to 7 days. Uptake of phosphate was least in the phosphate-rich Ap, and the Albic material also adsorbed little. By contrast the Red material sorbed phosphate strongly. By plotting the logarithm of the solution phosphate concentration against log time, two or three different kinetic regions could be distinguished: a fast reaction within the first minute of reaction; a lag period at intermediate to large initial values of phosphate concentration (P0); and a steady slow reaction. The lag phase cannot be described satisfactorily by the well-known adsorption models. The Langmuir equilibrium expression produced rather good fits at any fixed time of reaction, but the estimated adsorption maximum increased steadily with time. Empirical models were fitted to the data, among them a ‘lag-linear’ model developed to include the lag phase. The relative contribution of the fast reaction to total adsorption during 7 days decreased from 50 to 80% at small P0 to less than 10% at large P0 values. The fraction of fast-adsorbed phosphate followed the order Red 〉 Btg 〉 Albic 〉 Ap regardless of P0; the slow reaction is, by a log–log rate, ranked Albic ≅  Btg 〉 Red 〉 Ap for P0 〈 20 μm and Red 〉 Btg 〉 Albic 〉 Ap at larger initial concentrations. The Albic material does not minimize the risk of phosphate leaching through macropores, as demonstrated by its small P sorption capacity, the relatively slow adsorption rate and its small actual P content.

    Notes: The rate at which minerals in the soil weather is affected by pH and concentration of organic solutes (DOC). The rates of release of Al, Ca, Fe, K, Mg, Na, P and Si from a mineral soil sample to solutions of natural organic solutes and HCI (control) were determined at pH 3 and 4 for up to 17 weeks. Soil solutions were collected by centrifuging materials of O horizons from various soil types under four tree species (spruce, birch, oak, beech) and passed through a cation-exchange resin to yield H+-saturated organic solutes. The acceleration of the elements' release by the organic solutes was shown directly by the relative ligand effect (RLE), that is, the release rate in the organic solute solution divided by the release rate in the HCI solution (control) at the same pH. The RLE was greater at pH 4 than at pH 3, and it decreased for the elements in the order Fe 〉 Ca 〉 Mg 〉 Al ≈ Si 〉 K ≈ Na. This indicates that natural organic solutes are more important weathering agents at higher than lower pH and for weathering of mafic minerals rich in Ca, Fe and Mg than of felsic minerals such as K- and Na-feldspars. For all elements and at both pHs, RLE was strongly correlated with the concentration of DOC, which was also closely correlated with titratable acidity of the organic solutes. The important effect of soil type and tree species in producing weathering-promoting organic solutes therefore seems to be expressed through the concentration and not the composition of the organic soil solutes.

    Notes: Colonization or emergence of microbial pathogens may result in tissue destruction by activation of one or more of five distinct host degradative pathways (matrix metalloproteinase pathway, plasminogen-dependent pathway, phagocytic pathway, PMN-serine proteinase pathway and osteoclastic bone resorption) or by direct cleavage of extracellular matrix constituents by microbial proteinases. Activation of endogenous destructive pathways may be mediated by immune responses resulting in expression of degradative cellular phenotypes among both immigrant and resident cell populations. In addition, expression of degradative phenotypes may be triggered by direct influences on host cells of microbial products (LPS, enzymes, toxins). A body of evidence suggests that each of these mechanisms involves local production of proinflammatory cytokines and growth factors. The matrix metalloproteinase pathway is centrally involved in dissolution of all unmineralized connective tissues and perhaps in resorption of bone as well. The matrix metalloproteinase family consists of nine or more genetically distinct Zn++ endopeptidases which collectively cleave all of the constituents of the extracellular matrix. Recent studies have uncovered many essential elements of a complex, but still incomplete, regulatory network that governs tissue destruction. Proinflammatory cytokines and growth factors induce signalling pathways several of which are dependent on protein kinase C and result in transient expression of the transcription factors c-jun and c-fos. Initiation of transcription of most matrix metalloproteinase genes requires binding of the transcription factor AP-1 (c-jun/c-fos) to a specific promoter sequence but attainment of maximal transcription rates is dependent on interaction with other promoter elements as well. Several matrix metalloproteinases have been detected in crevicular fluids and tissues of inflamed human gingiva as have the proinflammatory cytokines (IL-1 and TNF-α) which regulate their transcription. Although the mere presence of enzymes and cytokines does not necessarily impart function per se, these observations suggest that some level of spatial or temporal linkage exists between metalloproteinase/cytokine expression and gingival inflammation.

    Notes: summary Twenty-two commercially available endodontic posts were examined with regard to stiffness, elastic limit and resistance to fracture. Differences in mechanical properties were explained by differences in width, shape and surface structure. In the selection of an endodontic post, its stability rather than its retention should be of primary clinical concern. Accordingly, the authors advocate the use of endodontic posts that have a cylindrical coronal portion and a conical apical portion.

    Notes: Summary Hairless mice were exposed to UVB irradiation from a Philips T1 12 light source. Mice that were exposed to UV-light and not protected with 5%p-aminobenzoic acid (PABA) showed a significantly higher number of peripheral blood granulocytes (P〈0·001) and a significantly higher mean weight of both the spleen and the liver (P〈0·001) than the non-irradiated controls. light microscopy of the histology of the enlarged liver and spleen showed a proliferation of granulocytes and the reticuloendothelial cells. Treatment with topical PABA during the whole period of UV-exposure prevented the peripheral blood granulocytosis. These protected mice also had a significantly (P 〈0·001) lower mean weight for the liver and spleen than UV-exposed and non-protected mice.

    Notes: Crystals, diffracting best to around 3 Å, have been grown from intact large and small ribosomal subunits. The bright synchrotron radiation necessary for the collection of the higher-resolution X-ray diffraction data introduces significant decay even at cryo temperatures. Nevertheless, owing to the reasonable isomorphism of the recently improved crystals of the small ribosomal subunits, reliable phases have been extracted at medium resolution (5–6 Å) and an interpretable five-derivative MIR map has been constructed. For the crystals of the large subunits, however, the situation is more complicated because at higher resolution (2.7–7 Å) they suffer from substantial radiation sensitivity, a low level of isomorphism, instability of the longest unit-cell axis and nonisotropic mosaicity. The 8 Å MIR map, constructed to gain insight into this unusual system, may provide feasible reasoning for the odd combination of the properties of these crystals as well as hints for future improvement. Parallel efforts, in which electron-microscopy-reconstructed images are being exploited for molecular-replacement studies, are also discussed.
