    Notes: This paper reports the results of X-ray powder diffraction experiments on C60Br24(Br2)2. This material is a C60 derivative and shows rhombohedral symmetry at room temperature. At low temperature, a rhombohedral-to-triclinic phase transition can be observed that involves a large distortion of the cell while the volume of the unit cell remains almost constant. This is a sufficiently complicated system that high-resolution synchrotron data and a combination of different tactics were required to reach a structure solution.

    Notes: Abstract A strain of Pseudomonas putida that can express a nitrate reductase that is located in the periplasmic compartment was isolated from freshwater. The enzyme was active in vivo during arginine fermentation and at the onset of oxygen limitation in batch cultures. The activity of the enzyme increased the yield of bacteria following fermentative growth under anoxic conditions with arginine, but nitrate reduction did not support growth on non-fermentable carbon substrates under anoxic conditions. Cells expressing the periplasmic nitrate reductase were capable of reducing nitrate in the presence of oxygen. Nitrate reduction under oxic conditions was clearly coupled to a respiratory electron transport chain because: (1) the process was sensitive to the respiratory inhibitors rotenone and 2-n-heptyl-4-hydroxyquinoline N-oxide, and (2) membrane-bound and periplasmic cytochromes were involved. This is the first report of the presence of a periplasmic nitrate reductase in a member of the γ proteobacteria.

    Notes: Abstract Hydroxylamine oxidation was measured in four recently isolated heterotrophic nitrate-reducing bacteria belonging to the genera Pseudomonas, Moraxella, Arthrobacter and Aeromonas. A hydroxylamine-cytochrome c oxidoreductase activity was detected in periplasmic fractions of the Pseudomonas and Aeromonas spp. and in total soluble fractions of the Arthrobacter sp. A monomeric 19-kDa non-haem iron hydroxylamine-cytochrome c oxidoreductase was purified from the Pseudomonas species and shown to be similar to hydroxylamine-cytochrome c oxidoreductase of Paracoccus denitrificans.

    Notes: Summary Combinatorial libraries have yielded high-affinity ligands for SH3 domains of a number of different proteins. We have shown that synthetic peptides containing these SH3 ligand sequences serve as specific probes of SH3 domains. Direct binding of the N-terminal biotinylated peptide ligands was conveniently detected in ELISA, filter-blotting, and dot-blotting experiments with the use of streptavidin-conjugated enzymes. In some cases, detection of peptide-SH3 interactions required that the biotinylated peptides first were preconjugated with streptavidin to form a multivalent complex. Interestingly, these nominally tetravalent SH3 peptide ligands cross-react to varying degrees with different SH3 domains. We have used such complexes to screen λcDNA expression libraries and have isolated clones that encode both known and novel SH3-domain-containing proteins. Based on the success of this methodology, we propose a general strategy by which ligands of a modular domain-containing protein can be isolated from random peptide libraries and used to screen cDNA expression libraries systematically for novel modular domain-containing proteins.

    Notes: Abstract The sinterability of (La,Ca)CrO3 is analysed by comparing the liquid phase behaviour in A-site excess and A-site deficient compositions (referring to the ABO3 formula). The analysis shows that a series of A-site excess and deficient compositions form distinct liquid phases belonging to the CaO-Cr2O3 phase system. Although both series experience grain growth and densification due to the presence of a liquid phase, the A-site excess compositions exhibit greater shrinkage and sinter to closed porosity, whereas the A-site deficient compositions remain porous. During the final stages of sintering, surplus liquid from the A-site excess material exudes to the free-surface forming a layer of uniform thickness. By comparison, the liquid in the A-site deficient composition segregates to the free-surface forming islands and leaving porous regions in the bulk matrix. It is concluded that the different liquid phase compositions in the A-site excess and A-site deficient (La,Ca)CrO3 have dissimilar wetting characteristics during the later stages of sintering. The A-site deficient liquid forms a premature solid phase, which accounts for the difference in sintering behaviour.

    Notes: Abstract Lanthanum chromate (LaCrO4) was synthesized as a low temperature ( 〈 700 °C) intermediate to perovskite-type, lanthanum chromite (LaCrO3). The lattice parameters, atom positions and bond lengths determined from X-ray powder diffraction show that LaCrO4 forms a monazite-type crystal structure. Lanthanum chromate forms a solid solution with Ca, having a solubility between 10 and 20 at%. Thermal analysis shows that at ambient oxygen pressure, LaCrO4 transforms to LaCrO3 at a temperature near 700 °C. It also indicates that (La,Ca)CrO4 initially transforms to LaCrO3 and CaCrO4 with the subsequent formation of a (La,Ca)CrO3 solid solution. Evidence of Ca segregation to the surface of (La,Ca) CrO3 particles is given by Auger electron spectroscopy and scanning electron microscopy.[/p]

    Notes: Summary A protocol was developed to observe plant microtubules and actin microfilaments in large tissue samples without physical sectioning. Rye (Secale cereale L. cv. Rymin) root tip pieces from two-day-old seedlings were fixed and processed for immunolabeling. Incubation times of 24–48 h were required to insure adequate penetration of fixatives, antibodies, and washing buffers. Clearing of the tissue with methyl salicylate reduced background auto-fluorescence that would otherwise interfere with the resolution of cytoskeletal structures. Microtubules or microfilaments in 5–7 cell layers were visualized using the optical-sectioning capability of laser scanning confocal microscopy (LSCM) and projected as three-dimensional images. The three-dimensional character of the cytoskeletal elements is retained when viewing stained cells of intact tissue. The net-like character of a microfilament array radiating out from a single point into the cytoplasm is maintained when the cells are stained in intact root tip pieces and imaging is accomplished in situ.
