    Notes: Abstract. Sprague-Dawley rats received intragastrically a single dose of 226Ra or 224Ra once weekly for 21 weeks. During this period the animals of both groups were thus exposed to comparable doses. After 12 weeks, a shortening of the maxillary incisors was observed, and this progressed during the course of the 55–week experiment. Radiologically, a loss of pulpal transparency and an apical irregular opacity in the maxillary incisors were demonstrable. Histologically, dysplastic changes of the maxillary incisors and multiple resorptions at the cemento-enamel junction of the molars were seen. The observed alterations were more pronounced in the 226Ra group than in the 224Ra treated group. The experimentally induced resorptions were comparable to those observed in patients who had incorporated 224Ra or 226Ra. Clinical, radiological, histological, γ-spectrometrical, and autoradiographical findings, as well as dose-estimations, are described.

    Description / Table of Contents: Zusammenfassung Die biologische Wirkung von 1-Acetoxypropylpropylnitrosamine (1-APPN) wurde an Syrischen Hamstern nach subcutaner Gabe überprüft. 1-APPN induzierte sowohl mesenchymale und epitheliale Geschwülste an der Injektionsstelle wie auch epitheliale Tumoren in entfernt liegenden Organen. Lokale Neoplasmen erwiesen sich als subcutane Sarcome, Adenocarcinome der Mamma und Schwannome, während Tumoren des Respirationstraktes (papilläre Polypen, Papillome, Adenome, Plattenepithelcarcinome, Adenocarcinome) wie auch der Pankreasgänge einer systemischen Wirkung entsprachen. Neoplasmen des oberen Verdauungstrakts und des Genitaltrakts weiblicher Hamster (Papillome, Plattenepithelcarcinome) sind möglicherweise Folge sowohl systemischer wie lokaler Wirkung von 1-APPN.
    Notes: Summary 1-Acetoxypropylpropylnitrosamine (1-APPN) was synthesized and its biological effect examined in Syrian hamsters after subcutaneous (s.c.) administration. 1-APPN induced mesenchymal and epithelial neoplasms at the injection sites, as well as epithelial tumors in remote organs. Local neoplasms were s.c. sarcomas, mammary adenocarcinomas and Schwannomas, whereas tumors of the respiratory tract (papillary polyps, papillomas, adenomas, epidermoid carcinomas, and adenocarcinomas) were attributed to a systemic effect as were pancreas duct tumors. Neoplasms which originated in the upper digestive and genital tracts of females (papillomas, epidermoid carcinomas) may be related to a systemic and local effect of 1-APPN.

    Description / Table of Contents: Zusammenfassung Die transplazentare Wirkung von Dibutylnitrosamin (DBN) and Nitrosohexamethylenimin (N-6-MI) wurde an Syrischen Hamstern untersucht. Beide Substanzen erreichten teilweise unverändert das fetale Gewebe. Bei den Jungen wurden keine makroskopisch nachweisbaren Mißbildungen gefunden. Die postnatale Mortalität war jedoch hoch. Induzierte Tumoren zeigten sich im Respirationstrakt. Einzeldosen von 30 mg DBN/kg KG und 2 Dosen von 10 mg N-6-MI/kg KG erzeugten keine Neoplasmen in der P-Generation, führten aber noch zu einer geringen Tumorrate in der F1-generation (DBN, 7,0%; N-6-MI, 2,0%). Bei Behandlung (bis zu 8 x) während der zweiten Hälfte der Tragzeit war die Häufigkeit der Tumoren in der P-Generation (DBN: 22%, N-6-MI, 20%) höher als in der F1-Generation (DBN, 6,0%, N-6-MI, 10%).
    Notes: Summary The transplacental carcinogenic effects of dibutylnitrosamine (DBN) and nitrosohexamethyleneimine (N-6-MI) were examined in Syrian hamsters. A proportion of both substances reached the fetal tissue unaltered. No macroscopic malformations were observed in the offspring; however, postnatal mortality was high. Respiratory tumours were found upon histologie examination of surviving animals. Single doses of 30 mg/kg body weight (b.w.) DBN and 2 doses of 10 mg/kg b.w. N-6-MI did not induce tumours in the P-generation, but led to a low tumour incidence in the F1-generation (DBN, 7.0%; N-6-MI, 2.0%). Treatment for up to eight days during the second half of pregnancy led to a higher tumour incidence in the P-generation (DBN, 22%; N-6-MI, 20%), than in the F1-regeneration (DBN, 6.0%; N-6-MI, 10%).

    Description / Table of Contents: Zusammenfassung Die aliphatischen Nitrosamine Dimethylnitrosamin (DMN), Diäthylnitrosamin (DEN), Dipropylnitrosamin (DPN) und Dibutylnitrosamin (DBN) erreichen nach subcutaner Verabreichung an trächtige, Syrische Hamster in quantitativ bestimmbaren Mengen das fetale Gewebe. Die Verbindungen waren im mütterlichen Blut, in Placenta, Fetus und Fruchtwasser wenigstens 2 Std nachweisbar; DBN zeigte meßbare Werte noch nach 6 Std. Der Vergleich der Häufigkeit und Latenzzeit der Tumoren, die in den Luftwegen und im Verdauungstrakt der F1-Generation gefunden wurden, mit denen in der P-Generation nach Behandlung mit einer Einzeldosis von DMN oder DPN ließ nur eine schwache oder grenzwertige, transplacentare Wirkung erkennen. Jedoch entwickelten sich bei den Jungen einige Tumorarten, die nur gelegentlich bei den Müttern oder anderen Hamstern dieser Kolonie gefunden wurden.
    Notes: Summary The aliphatic nitrosamines dimethylnitrosamine (DMN), diethylnitrosamine (DEN), dipropylnitrosamine (DPN), and dibutylnitrosamine(DBN) reached fetal tissue in quantitatively measurable amounts after subcutaneous administration to pregnant Syrian hamsters. The compounds were present for at least 2 h in maternal blood, placenta, fetus, and amniotic fluid; DBN was still measurable after 6 h. Only a weak or borderline transplacental effect was seen when incidences and latencies of neoplasms in the respiratory and digestive tracts of the F1-generation were compared with those of the P-generation after exposure to a single dose of DMN or DPN. However, some tumor types occurred at relatively high rates in the young, but were seen only occasionally in their mothers or in this hamster colony in general.

    Description / Table of Contents: Zusammenfassung An Syrischen Hamstern wurde gezeigt, daß sich Nitrosopiperidin (NP) quantitativ nach subcutaner Injektion im mütterlichen Blut für mehr als 8 Std nachweisen läßt, während die Substanz zu diesem Zeitpunkt in Placenta, Fetus und Fruchtwasser nicht mehr meßbar war. In gleichen Geweben fanden sich nur Spuren von N6MI nach 2 Std. Die langzeitige transplacentare Wirkung einer Einzeldosis von NP war schwach. Dies zeigt sich an der geringen Rate von Tumoren im Respirationstrakt (P-Generation: 54%; F1 Generation: 4%). Einige Neoplasmen, die bei den behandelten Müttern nicht gefunden und gewöhnlich nicht bei Kontrollen beobachtet wurden, traten im Verdauungstrakt der Jungen auf. Diese Tumoren wurden als Ausdruck einer transplacentaren Grenzwirkung gewertet. Geschwülste anderer Organe (Urogenitaltrakt, reticulo-endotheliales System, endocrine Organe und andere Gewebe) zeigten Häufigkeitsschwankungen, wie sie in unserer Hamsterkolonie beobachtet wurden.
    Notes: Summary Nitrosopiperidine (NP) was found in Syrian hamsters quantitatively in the maternal blood for more than 8 h after subcutaneous injection, whereas it disappeared from placenta, fetus and amniotic fluid within the same time period. For N6MI, only traces were seen after 2 h in the same tissues. The long-term transplacental effect of a single dose of NP was weak, as demonstrated by a low respiratory tract tumor incidence (P-generation: 54%, F1 generation: 4%). Some tumors occurring in the digestive tract of exposed young were not found in their mothers and not commonly observed in controls. These tumors were considered a borderline transplacental effect. Tumors of other sites (i.e., the urogenital and genital tracts, reticuloendothelial system, endocrine organs and other tissues) corresponded in incidences to the overall fluctuations observed in this hamster colony.

    Description / Table of Contents: Zusammenfassung Die biologische Wirkung von N-Nitroso-N-Methyl-N-Dodecylamin (NMDA) wurde bei Syrischen Hamstern in einer vergleichenden Untersuchung geprüft. Die Ergebnisse zeigten, daß nach intragastrischer Verabreichung von NMDA vorwiegend die Harnblase von der carcinogenen Wirkung betroffen ist. Übergangsepithel-Geschwülste entwickelten sich, und eine positive Dosis Wirkungsbeziehung wurde beobachtet. Lungentumoren entstanden nur bei weiblichen Hamstern, während männliche Tiere häufiger Geschwülste der Nasenhöhle und des Verdauungstraktes zeigten.
    Notes: Summary The biological effect of N-nitroso-N-methyl-N-dodecylamine (NMDA) was examined in Syrian hamsters as part of a comparative study. Data obtained show that after intragastric administration of NMDA, the urinary bladder was the main target organ for the carcinogenic effect. Transitional cell neoplasms developed and a positive dose response relationship was observed. Lung tumors occurred only in female hamsters, whereas males more frequently showed neoplasms of the nasal cavity and digestive tract.

    Description / Table of Contents: Zusammenfassung Die tumorerzeugende Wirkung von Dimethylnitrosamin wurde nach einmaliger Gabe in 5 unterschiedlichen Dosen bei ausgewachsenen Swiss, ASW/SN-und A-Mäusen verglichen. Eine erhöhte Tumorhäufigkeit fand sich nur in der höchsten Dosierungsgruppe bei Swissund ASW/SNMäusen, jedoch nicht bei Tieren des A-Stammes. Die Wirkung der Behandlung war allerdings bei Lungentumoren eindeutig. Die Rate der Carcinome stieg signifikant in allen 3 Stämmen, obwohl eine klare Dosis-Wirkungsbeziehung nur bei Swissund ASW/SN-Mäusen gefunden wurde. Die Häufigkeit von Adenomen war nur bei Swiss-Mäusen dosisabhängig.
    Notes: Summary The tumorigenic effect of dimethylnitrosamine given once at 5 different dose levels was compared in adult Swiss, ASW/SN and A-strain mice. Considering the overall tumour incidence, at the highest dose level an increase was found in Swiss and ASW/SN mice, but not in A-strain animals. However, the effect of treatment was unequivocal for lung neoplasms. The rate of carcinomas rose significantly in all 3 strains, although a clear dose response-relationship could be established only in Swiss and ASW/SN mice. A dose-dependent effect for adenomas was found solely in Swiss mice.
