Notizen: Abstract  This study compared two recombinant human (rh) hematopoietic growth factors in healthy volunteers for stem cell stimulation. Granulocyte colony-stimulating factor (G-CSF,n=9) or granulocyte-macrophage colony-stimulating factor (GM-CSF,n=8) was given subcutaneously for 5 days (5 μg/kg/day). Controls (n=5) received no growth factor. Laboratory parameters and side effects were monitored for 8 days. Within 24 h, both cytokines led to a rapid increase of leukocytes, the majority of which were granulocytes. Compared with the controls (n=5), the increase on day 5 in the G-CSF/GM-CSF groups was 37-/10-fold (CD34+ cells), 5.2-/2.4-fold (leukocytes), 7.2-/3.0-fold (granulocytes), 7.4-/4.4-fold (monocytes), 1.7-/1.1-fold (lymphocytes), 9.8-/2.7-fold (basophils), 2.3-/9.6-fold (eosinophils), and 1.9-/1.6-fold (reticulocytes). The mobilization of myeloblasts, promyelocytes, myelocytes, and metamyelocytes coincided with the pronounced increase of CD34+ PBPC observed on day 4. Serum levels of uric acid (UA) and lactic dehydrogenase (LDH) increased under G-CSF, and platelets decreased after G-CSF discontinuation. Rash at the injection site occurred in 50% of the GM-CSF-treated volunteers. Seven volunteers in the GM-CSF group and six in the G-CSF cohort complained of flu-like symptoms, including musculoskeletal pain. We conclude that, in terms of tolerance and mobilization of CD34+ cells and leukocytes, G-CSF is superior to GM-CSF, but higher levels of UA and LDH and late decrease in platelets make monitoring of these parameters necessary.

Notizen: Abstract  A large group of patients relapsing after allogeneic bone marrow transplantation (BMT) have obtained remission after infusion of leukocytes from their original donor, suggesting a graft-versus-myeloma effect. However, side effects such as graft-versus-host disease and myelosuppression are severe, and sometimes fatal, complications of this therapeutic approach. Previously we demonstrated that patients with leukemia who lack donor hematopoiesis in relapse after BMT experience severe and lasting aplasia after infusion of donor leukocytes. In two patients – one with extramedullary and one with marrow relapse after a sex-mismatched transplantation – we analyzed hematopoietic chimerism by cell sorting and bone marrow cultures. CD34-positive cells, CD4-CD8-positive cells, committed progenitors, and LTC-IC were of donor origin, as demonstrated by two-color fluorescence in situ hybridization (FISH). Additionally, in relapse complete donor T-cell chimerism was seen. In contrast, plasma cells were of recipient origin in the patient who had a relapse in the bone marrow. Both patients were treated with infusions of donor leukocytes from their original donor. Neither patient suffered myelosuppression, and one achieved a stable complete remission.

Notizen: Abstract We report on new measurements of the electrical conductivity, Hall effect and thermoelectric power in the temperature range from 2 K to 440 K for AlCuFe quasicrystals of different compositions and annealing treatments. Particularly, the Hall coefficient and the thermopower show a strong dependence on composition and also on heat treatment. The increase of sample perfection on annealing between 810° C and 825° C is demonstrated by analysing X-ray measurements. All measured quantities can be explained in terms of a semimetal containing both electrons and holes at low temperatures, at least for samples near the ideal quasicrystalline structure and composition.

Notizen: Abstract A premature infant who developed respiratory distress syndrome required artifical ventilation. A right-sided tension pneumothorax at the age of 78 h was treated by insertion of a P.V.C. chest tube reaching deep into the right upper chest. At discharge on day 51 a Horner's syndrome of the right eye was noticed. Detailed examination at the age of one year revealed a typical peripheral preganglionar Horner's syndrome. Because other causes could be ruled out we assume that the lesion of the sympathetic nerve fibers was caused by the tip of the chest tube near the first thoracic intervertebral space.
