    Notizen: The desorption of Trichoderma reesei cellulase from Avicel by a wide range of desorbents was measured. Emphasis was placed on desorption at alkaline pH. A maximum desorption of 65-68% Avicelase activity was achieved by contact with NaOH, pH 10.0, at 40°C for 5 min in the presence of 0.005% Triton X-100 or Tween 80. The design of a suitable desorption process using these conditions is discussed. Glycerol was also effective as a desorbent either alone or in combination with alkali and detergent. However, relatively high concentrations of glycerol were needed and the maximum desorption achieved, 68%, was not significantly greater than that with only alkali and detergent.

    Notizen: Scalopus membranes are characterized by: Superficial nidation; antimesometrial orientation of the embryonic disc; amniogenesis by folding; an extensive but transitory choriovitelline placenta; a large yolk sac with late and incomplete inversion; large persistent allantoic vesicle; a very broad, thin, villous, epitheliochorial chorioallantoic placenta of annular shape interrupted mesometrially, dotted with numerous areolae, and bordered by a non-villous sparsely vascular chorioallantoic membrane connected with the persistent bilaminar omphalopleure by a very narrow rim of chorion. There is no decidua. Electron microscopy shows that at 8 mm, CR, (limb bud embryo) the uterine epithelium of the interhemal membrane may be 0.5 μm or less in thickness, but that it shows no signs of degeneration. Trophoblastic microvilli often penetrate the epithelium to within 0.2 μm of its base. At this time there is active secretion by the uterine glands, and cellular hypertrophy and cytolysis of the epithelium at the gland mouths, with active phagocytosis by the areolar cytotrophoblast. The occurrence of absorptive areolae in an insectivore emphasizes the probable primitiveness of this widely distributed placental mechanism. In spite of similarities of the yolk sac to that of rabbits and rodents, the bilaminar omphalopleure produces no invasive trophoblastic giant cells. The definitive membranes of Parascalops breweri and Scapanus latimanus are like those of Scalopus. The placentae of Talpa europaea, Condylura cristata, and Neurotrichus gibbsii are discoid and relatively much smaller, thicker and more complex in internal structure. There is some reason to believe that the fetal membrane systems of moles and shrews (Soricoidea) are more like those of the ancestral mammalian stock than are those of any other recent eutherians.

    Notizen: A variety of computational methods, including the semiempirical techniques AM1, PM3, and MNDO, and the thermochemical basis sets of Benson and Stine, was used to calculate and compare heats of formation (ΔHf°) data for optimized geometries of a variety of aromatic and nonaromatic heterocycles. Detailed analyses, including 6-31G* and MP2/6-31G* ab initio calculations, were performed for the oxazole and thiazole heterocycles. The results indicate a scatter among the methods sensitive to the nature of the heterocycle. This was in particular evident in the oxazole molecule, where AM1 gave a singularly high value of ΔHf° consistent with longer calculated bond lengths, particularly about the oxygen atom. Aromatic stabilization energy appears to be addressed differently among the employed methods. Implications of this contrast applied to calculation of macromolecular systems containing heterocyclic units are discussed.

    Notizen: We have compared the biochemical expression of cholinergic enzymes with the morphological differentiation of efferent nerve fibers and endings in the cochlea of the postnatally developing mouse. Choline acetyltransferase (ChAT) and acetylcholinesterase (AChE) are present in the newborn cochlea at specific activities 63% and 25%, respectively, of their mature levels. The relative increases in ChAT, in AChE, and in its molecular forms over the newborn values start about day 4 and reach maturity by about day 10. The biochemical results correlate well with the massive presence of nerve fibers stained immunocytochemically for ChAT and AChE or enzymatically for AChE in the inner and outer hair cell regions. Ultrastructural studies, however, indicate the presence of only few vesiculated fibers and endings in the inner and outer hair cell regions. The appearance of large, cytologically mature endings occurs only toward the end of the third postnatal week. The discrepancy may be resolved in the electron microscope using the enzymatic staining for AChE. Labeling is seen on many nonvesiculated fibers and endings in the hair cell regions, suggesting that the majority of the efferent fibers in the perinatal organ may be biochemically differentiated but morphologically immature. The results may imply that the efferents to inner and outer hair cells develop earlier than indicated by previous ultrastructural studies. Moreover, the pattern of development suggests that in the cochlea, as in other tissues, the biochemical differentiation of the efferent innervation may precede the morphological maturation.
