    Notes: Abstract Background. The purpose of this study was to investigate the correlation between P-glycoprotein status and outcome in adult patients with high-grade osteosarcomas and soft-tissue sarcomas. Methods. P-glycoprotein status was determined im-munohistochemically in specimens from 28 patients with osteosarcoma and 34 patients with soft-tissue sarcoma. The polyclonal antibody mdr(Ab-1) was used for either decalcified or undecalcified tissue samples which were formalin-fixed and paraffin-embedded. The expression of P-glycoprotein mRNA was also determined by the polymerase chain reaction in 23 fresh sarcoma specimens. P-glycoprotein status was analyzed in relation to the duration of event-free survival. Results. Positivity for P-glycoprotein was found in 29% of the osteosarcomas and 34% of the soft-tissue sarcomas. Consistent results were obtained at both the immunohistochemical and reverse transcriptase-polymerase chain reaction (RT-PCR) levels in 19 of 23 sarcomas (83%). In patients with osteosarcoma, the presence of increased levels of P-glycoprotein was significantly associated with a decreased probability of event-free survival after diagnosis (P = 0.022). In contrast, in patients with soft-tissue sarcoma there was no correlation between the level of P-glycoprotein and prognosis. Conclusions. In patients with high-grade osteosarcomas, the presence of increased levels of P-glycoprotein detected by polyclonal antibody mdr(Ab-1) was associated with a significantly increased risk of adverse events. This association was not found in patients with soft-tissue sarcomas.

    Notes: Abstract Animal studies have suggested the involvement of the adrenergic system in drinking behavior. The present study investigated the involvement of the α2-adrenergic system in the polydipsia of patients with chronic schizophrenia by use of an α2 agonist and an antagonist. Four patients with schizophrenic disorders accompanied by intermittent hyponatremia and polydipsia were the subjects of, and completed, this study. Drinking behavior was assessed by calculating the percent of maximum weight gain [PMWG: (maximum diurnal weight – standard weight) × 100/standard weight]. Standard weight was defined as body weight after 8 h of water restriction. Clonidine (75, 150, and 225 mg/day) increased the PMWG in a dose-dependent manner in the four subjects. In contrast, in three of the subjects, mianserin (30, 60, and 90 mg/day) decreased PMWG, and the severe polydipsia disappeared almost completely. These findings indicate clearly that the α2-adrenergic system is involved in the drinking behavior of schizophrenic patients. Mianserin appears to be clinically useful in treating such patients with polydipsia.

    Description / Table of Contents: Résumé On a étudié les modifications de l'aspect des métastases pulmonaires chez des malades porteurs d'un ostéosarcome et traités par chimiothérapie complémentaire. Trente-deux malades traités à la fois par chirurgie radicale et chimiothérapie ont été examinés (groupe avec chimiothérapie). Soixante-deux malades traités seulement par chirurgie radicale (groupe sans chimiothérapie) ont également été suivis, constituant un groupe de contrôle. Le groupe avec chimiothérapie a présenté une diminution du nombre ainsi qu'un retard à l'apparition des métastases. Le temps de doublement de ces tumeurs ne diffère pas entre les deux groupes. La localisation initiale des métastases dans le groupe avec chimiothérapie est le plus souvent la base du poumon, tandis que dans le groupe sans chimiothérapie c'est habituellement la partie moyenne du poumon. Après apparition des métastases pulmonaires la durée de survie est plus importante dans le groupe avec chimiothérapie que dans l'autre. L'examen histologique montre que les foyers métastatiques sont identiques à la localisation initiale de la tumeur.
    Notes: Summary Alterations in the pattern of pulmonary metastasis of patients with osteosarcoma treated with adjuvant chemotherapy were studied. Thirty two patients who were treated with both radical surgery and adjuvant chemotherapy were observed (chemotherapy group). As a control, sixty two patients treated with radical surgery alone were also assessed (non-chemotherapy group). The chemotherapy group demonstrated a reduction in the number and a delay in the appearance of metastases. The tumour doubling time did not differ between the chemotherapy and non-chemotherapy groups. The initial site of metastasis among the chemotherapy group was most commonly in the lower lung field, whereas among the non-chemotherapy group it was usually to the middle lung field. The chemotherapy group survived longer than the non-chemotherapy group after developing pulmonary metastases. Histological examination showed that the metastatic foci simulated the primary sites.
