    Notes: Summary Knowledge of the voltage dependencies of apical and basolateral conductances is important in determining the factors that regulate transcellular transport. To gain this knowledge it is necessary to distinguish between cellular and paracellular currents and conductances. This is generally done by sequentially measuring transepithelial current/voltage (I t /V t ) and conductance/voltage (g t /V t ) relationships before and after the abolition of cellular sodium transport with amiloride. Often, however, there are variable time-dependent and voltage-dependent responses to voltage perturbation both in the absence and presence of amiloride, pointing to effects on the paracellular pathway. We have here investigated these phenomena systematically and found that the difficulties were significantly lessened by the use of an intermittent technique, measuringI t andg t before and after brief (〈10 sec) exposure to amiloride at each setting ofV t .I/V relationships were characterized by these means in frog skins (Rana pipiens, Northern variety, andRana temporaria). Cellular current,I c , decreased with hyperpolarization (larger serosa positive clamps) ofV t . DerivedI c /V t relationships betweenV t =0 and 175 mV (serosa positive) were slightly concave upwards. Because values of cell conductance,g c , remained finite, it was possible to demonstrate reversal ofI c . Values of the reversal potentialV' averaged 156±14 (sd,n=18) mV. Simultaneous microelectrode measurements permitted also the calculation of apical and basolateral conductances,g a andg b . The apical conductance decreased monotonically with increasing positivity ofV t (andV a ). In contrast, in the range in which the basolateral conductance could be evaluated adequately (V t 〈125 mV),g b increased with more positive values ofV t (andV b ). That is, there was an inverse relation betweeng b and cellular current at the quasi-steady state, 10–30 sec after the transepithelial voltage step.

    Notes: Summary The incidence of polyneuronal innervation in the cutaneous pectoris muscle of the adult frog,Rana esculenta, was determined quantitatively using electrophysiological and morphological techniques. The mean percentages of multiple innervated endplates obtained with both techniques from a series of 19 muscles examined at all times of the year were in very good correlation: 30.7% (196/639 endplates) by electrophysiology and 30.5% (478/1569 endplates) by morphology. In nine muscles examined during the period from December to March the mean percentages, 36.8% (110/299) by electrophysiology and 38.6% (281/727) by morphology, were significantly higher than those obtained for 10 muscles investigated during the period from May to November, 25.3% (86/340) and 23.4% (197/842) with both techniques respectively. The higher incidence of collateral sprouted branches detected at polyinnervated endplate sites in muscles of winter frogs might be related to these seasonal variations. Most of the 1688 fibres from 26 muscles examined throughout the year exhibited one centrally located endplate. However, around 11% of them were found to be innervated at two separate endplate sites. Muscle fibres exhibiting this type of innervation were invariably the largest fibres in each muscle tested, having an apparent diameter greater than 48 μm. The distance between the endplates of these fibres represented between 10 and 30% of their total length. No significant seasonal variations were observed in the incidence of these dually innervated fibres. In conclusion, both electrophysiological and morphological results show that the normal incidence of polyneuronal innervation in the cutaneous pectoris muscle of adultRana esculenta is affected by seasonal related factors which influence the nodal sprouting activity. Moreover, they show that a dual pattern of innervation is a common feature in large fibres of this muscle.

    Notes: Abstract The effect of inoculum size and gene dosage on the level of antibiotic resistance mediated by TEM-1β-lactamase was measured. From the results it seemed that gene dosage is a more efficient mechanism than inoculum size for increasing TEM-1 mediated resistance toβ-lactam antibiotics. It also seemed that the two mechanisms for enhancing antibiotic resistance are synergistic. The clinical implications of these results are discussed.

    Notes: Abstract The expressions for quantitative phase analysis from permeability versus temperature curves are discussed, and the results are illustrated by the determination of magnetic Co2 Y remnants (2CoO·2BaO·6Fe2O3) in Co2Z (2CoO·3BaO·12Fe2O3) samples. The method is well suited for the determination of small inclusions of a magnetic phase in a given magnetic material, in a range where X-ray methods become useless as a rule.
