    Notes: Summary Besides decreased platelet production and an increased platelet sequestration in the spleen chronic DIC is thought to cause thrombocytopenia in chronic liver diseases. To investigate if this mechanism could play a role in patients with severe, but stable hepatic cirrhosis, we studied 32 pts. Twenty-eight of them had portal hypertension, 22 had bleeding episodes from oesophageal varices. To evaluate platelet activity, thrombin generation and fibrinolytic activity in vivo, plasma and platelet levels ofβ-thromboglobulin (β-TG) and platelet factor 4 (PF4) as well as fibrinopeptide A (FPA) and the fibrinogen degradation product FCB-3, generated by the action of plasmin, were determined radioimmunologically. Plasma levels and platelet content of serotonin (5-HT) were measured fluorometrically. The mean platelet count/µl was reduced in pts. (109.000±67.000) compared to a control group (n=25, 264,000±52,000,p〈0.001). Factor VII (45±20%), prothrombin time (63±16%) AT-III activity (65±19%) and AT-III concentration (67±23%) were decreased in the pts'. group (p〈0.01). In contrast, plasma levels ofβ-TG (17±7 ng/ml) and of PF4 (4±2 ng/ml) as well as platelet content (µg/109 platelets) ofβ-TG (59±18), PF4 (21±6) and 5-HT (400±210) were found to be in the normal range. Neither mean FPA plasma concentration (3±2 ng/ml) nor mean FCB-3 scrum level (11±5 pmol/ml) differed significantly from controls. Our results demonstrate that pts. with thrombocytopenia and stable hepatic cirrhosis do not have evidence of chronic DIC, primary fibrinolysis or increased platelet activity in vivo. However, our results are compatible with increased platelet sequestration in the spleen and/or decreased platelet survival due to faster extravascular platelet clearance, or decreased platelet production within the bone marrow.

    Notes: Abstract This study was performed on patients (n=18) suffering from strictly defined hyperdynamic septic shock. Plasma factors (C-reactive protein, acid α1-glycoprotien, fibrinogen, fibrinopeptide A, fibrinogen-fibrin split products, factor XIII, antithrobin III, complement factors C3 and C4, inter-α-trypsin-inhibitor and α2-macroglobulin) measured during hyperdynamic septic shock were highly abnormal. The activation and consumption of clotting, fibrinolytic and complement factors due to system-specific proteinases (such as thrombokinase or plasminogen activators) seemed to be intensified by the nonspecific proteolytic activity of granulocytic proteinases probably released by the action of endotoxins. Possible therapeutic measures to maintain the endogeneous defence mechanism against enhanced proteolysis during septic shock are discussed.

    Notes: Abstract The coralline alga Phymatolithon calcareum was dredged from 13 m in the Kattegatt, Baltic Sea, in December, 1980, and its rate of calcification was measured by 45Ca++-uptake methods. Light-saturated calcification rates at 5°C ranged from 15.8 μg CaCO3 g-1 dry wt h-1 for the basal parts of the plants to 38.7 μg CaCO3 g-1 dry wt h-1 for the tips. These “age” gradients were not apparent when calcification rates were expressed on the basis of surface area. Experiments with salinity (10, 20, 30‰) and temperature (0°, 5°, 10°, 20°C) indicated that optimum conditions for calcification were at 30‰ S and at temperatures above 10°C. Salinity had a greater influence on calcification rate than did temperature, and there was a positive relationship between salinity and calcification rate at all temperatures. In 6 mo old cultures, salinity was again the important factor, with all plants remaining healthy at 30‰ except those at the highest temperature (20°C). These trends, and the low calcification rates at 10‰S (4.6 μg CaCO3 g-1 dry wt h-1 at 5°C to 8.6 μg CaCO3g-1 dry wt h-1 at 20°C) suggest that low salinity may be the explanation for the general absence of P. calcareum from the brackish waters of the Baltic Sea. Short-term experiments in which salinity was kept constant while Ca++ concentration was altered, and experiments in which salinity was varied and Ca++ concentration kept constant, suggest that it is the calcium ion concentration and not salinity per se which affects calcification rates.

    Notes: Estradiol-17β and [4-14C]estradiol-17β in total mass amounts of 0, 10 and 20 pg per analysis in the presence of 1000 pg [2H8]estradiol as an internal standard were simultaneously quantitatively determined by gas chromatography mass spectrometry using the selected ion monitoring technique. The experiment (a nested three way analysis of variance) was designed to investigate the two-dimensional error structure which facilitates the study and comparison of the variances: (1) the variance originating in the device, including gas chromatograph, mass spectrometer and computer system with its interfaces; (2) the variance originating from titration, pipetting and derivatization of the samples. The study revealed that in this concentration range the variance due to sample preparation is smaller than that due to the device. The deviations due to the device were shown to have a two-dimensional normal distribution.

    Notes: A method is proposed for the quantitative estimation of a substance and its labelled analogue using the selected ion monitoring technique of gas chromatography mass spectrometry, in which estimation is achieved without a third labelled analogue as internal standard. In a first measurement the ratio of natural to labelled substance is estabolished. Then a known amount of either labelled or natural compound is added, the isotope ratio is determined a second time and from these two measurements the mass amounts can be calculated. When applied to the heptafluorobutyrates of androstenedione and progesterone about 10 pg of 14C labeled compounds can be determined in the presence of 1 ng of the natural substances.
