    Notes: We have investigated magneto-transport and cyclotron resonance (CR) of two-dimensional electron gas in silicon δ-doped p-InSb under a magnetic field of up to 12 T at 4.2 K. Because there are multiple subbands occupied, Shubnikov–de Haas oscillations show a beating behavior. The CR spectra also display several peaks originating from different subbands. Effective masses of electrons associated with the lowest three subbands can therefore be directly determined, and they are in excellent agreement with a self-consistent calculation, which takes into account the electrostatic Poisson equation, the Schrödinger equation, and realistic sample parameters. Furthermore, we observed an absorption peak, whose resonance position has anomalous angle dependence. It is attributed to impurity CR where donors are in the vicinity of the δ-doped sheet.

    Notes: We have performed optical transmission measurements on radiatively heated GaAs substrates as a function of molecular beam epitaxial growth of InAs, GaSb, AlSb, and GaAs films. The energy gap of the GaAs substrate is observed to decrease strongly in energy when materials with band gaps smaller than GaAs are deposited. This decrease in energy gap is a consequence of a substantial increase in growth temperature induced by the deposition of the film. We have observed increases in temperature of over 150 °C from the temperature measured before film growth. Because the thermocouple is weakly coupled to the radiatively heated substrate, conventional temperature controllers are ineffective at measuring or accounting for this change in temperature.

    Notes: We have studied far infrared transmission spectra of various AlSb/InAs/AlSb single quantum wells, where the well width ranges from 60 to 200 A(ring). Because the quantum well is thin, the far infrared absorption due to transverse-optical phonons is not saturated. We obtained excellent fitting to the transmission spectra by taking into account the complex dielectric functions with a transfer matrix formalism. The thickness of InAs quantum wells can be determined with monolayer resolution. High-resolution transmission electron microscopy shows clear lattice images in a region away from the AlSb/InAs interfaces. The thickness of the pure nonintermixed InAs region is in excellent agreement with our far infrared absorption results. Phonon absorption can therefore provide a nondestructive method to effectively determine the number of pure InAs monolayers, in spite of interdiffusion occurring at the interface.

    Notes: InAs/Ga1−xInxSb superlattices have been investigated by magneto-optical and magnetotransport techniques. Band gaps, determined from interband magneto-optical measurements and from the temperature dependence of the intrinsic carrier concentration, are in the long wavelength infrared region (8.3–12.4 μm) and are in good agreement with gaps calculated from a two-band model. Both electron and hole effective masses were measured by cyclotron resonance and the electron effective mass is found to be a factor of 4–5 larger than in HgCdTe (the industry standard IR material). This is necessary for reduced dark currents and good optical absorption coefficients in this material.

    Notes: Transcription elongation catalysed by DNA-dependent RNA polymerase does not occur at a constant rate. Instead, during the transcription of many genes pausing occurs at defined template positions. Pausing is known to be influenced by the intracellular NTP concentration, the secondary structure of the growing transcript or by transcription factors like NusA. We have investigated the effects of the template topology of transcriptional pauses in the presence and absence on purified NusA protein. Taking advantage of a method for quantifying transcriptional pauses we have studied pausing behaviour during in vitro transcription of the early region of a plasmid-encoded ribosomal RNA operon. Plasmid templates with different super-helical densities (σ between +0.0017 and -0.055) were employed in transcription elongation assays. If linearized or relaxed templates are used, some of the characteristic pauses can no longer be detected. For the stronger pauses we could demonstrate a direct correlation between pause strength and the negative superhelical densities of the templates used. This correlation is observed regardless of whether or not pauses are dependent upon NusA. Changes in the average transcription elongation rate, caused by variations in the NTP concentration or the temperature, do not appear to have a comparable effect on transcription pausing. The results are consistent with the assumption that the template topology has a regulatory function in transcription elongation of rRNA genes in Escherichia coli.

    Notes: A new polarized target for neutron scattering has been designed by CERN and tested successfully using the reactor FRG-1 at the GKSS Research Centre. The nuclear spins are aligned with respect to the external field – parallel or antiparallel – by dynamic nuclear polarization (DNP). To avoid absorption of neutrons by 3He, the frozen solutions of biomolecules are immersed in liquid 4He which in turn is thermally coupled to the cooling mixture of 3He/4He of the dilution refrigerator. Compared with earlier experiments where the sample had been cooled directly by 3He, the rate of detectable neutrons increased by a factor of 30. Another factor of 30 is due to the installation of the cold source and the beryllium reflector in FRG-1. Polarized neutron scattering from apoferritin in deuterated solvent shows that the proton spin polarization is homogeneous in apoferritin molecules. After saturation of proton nuclear magnetic resonance (NMR), polarized neutron scattering is dominated by deuteron spin contrast. With the deuterated large subunit of E. coli ribosomes, three different basic scattering functions are derived from spin-contrast variation, reflecting the known scattering-length-density distribution of the architecture of rRNA and ribosomal proteins. The planned in situ structure determination of a mRNA fragment is discussed in the light of the present results.

    Notes: Summary The cytoplasmic islet cell antibody patterns of sera from islet cell antibody positive non-diabetic and diabetic endocrine autoimmune patients, and newly-diagnosed Type 1 (insulin-dependent) diabetic patients were characterised using four layer immunofluorescence with monoclonal antiproinsulin or anti-glucagon antibodies. Two distinct islet cell antibody types were identified. One gave a diffuse cytoplasmic staining in both Beta and Alpha cells (‘whole’ islet pattern), and was not affected by pre-incubation with rat brain homogenate. The other had a granular appearance with staining restricted predominantly to Beta cells (‘selective’ islet pattern) and was completely inhibited by pre-incubation with rat brain homogenate. Some sera appeared to have a ‘mixed’ islet pattern, in which glucagon-positive cells gave a weaker cytoplasmic staining than proinsulin-positive cells. The granular ‘selective’ pattern was found in sera from 19 (79%) of 24 non-diabetic endocrine autoimmune patients, in two (22%) endocrine autoimmune patients who developed Type 1 diabetes (p〈0.0001 vs non-diabetic endocrine autoimmune patients), and in none of 19 newly-diagnosed diabetic patients. The ‘whole’ islet pattern was found only in sera from patients who had, or who subsequently progressed to, Type 1 diabetes. This study has identified a novel islet cell antibody specificity and demonstrates that in islet cell antibody positive endocrine autoimmune patients, only islet cell antibodies which stain both Beta and Alpha cells are associated with progression to Type 1 diabetes.

    Notes: Summary Antigens in a particulate conformation were shown to be highly immunogenic in mammals. For this reason, the particle forming capacity of derivatives of the HIV-1 group specific core antigen p 55gag was assayed and compared dependent on various expression systems: recombinant bacteria, vaccinia-and baculoviruses were established encoding the entire core protein p 55 either in its authentic sequence or lacking the myristylation consensus signal. Moreover, p 55gag was expressed in combination with the protease (p 55-PR) or with the entire polymerase (p 55-pol), respectively. Budding of 100–160 nm p 55 core particles, resembling immature HIV-virions, was observed in the eucaryotic expression systems only. In comparison to the vaccinia virus driven expression of p 55 in mammalian cells, considerably higher yields of particulate core antigen were obtained by infection ofSpodoptera frugiperda (Sf9) insect cells with the recombinantAutographa californica nuclear polyhedrosis (AcMNPV) baculovirus. Mutation of the NH2-terminal myristylation signal sequence prevented budding of the immature core particles. Expression of the HIV p 55-PR gene construct by recombinant baculovirus resulted in complete processing of the p 55gag precursor molecule in this system. The introduction of an artificial frameshift near the natural frameshift site resulted in constitutive expression of the viral protease and complete processing of p 55, both inEscherichia coli and in vaccinia virus infected cells. Interestingly, significant processing of p 55 resembling that of HIV infected H 9 cells could also be achieved in the vaccinia system by fusing the entirepol gene to thegag gene. Moreover, processing was not found to be dependent on amino-terminal myristylation of thegag procursor molecule, which is in contrast to observations with type C and type D retrovirus. However, complete processing of p 55 into p 24, p 17, p 9 and p 6 abolished particle formation. Purified immature HIV-virus like particles were highly immunogenic in rabbits, leading to a strong humoral immune response after immunization. Empty immature p 55gag particles represent a noninfectious and attractive candidate for a basic vaccine component.
