    Notes: Non-specific antibody production usually accompanies the T-cell-regulated B-cell response. In this papaer the Machanisms involved in non-MHC-restricted T-cell interaction were studied. As previously shown for NK cells, activated B cells induce IFN-α and TNFα production in non MHC-restricted cytotoxic T lymphocytes (NrCTL). Using an in vitro model system, we demonstrate that direct cell cell interactions are required to induce these cytokines in NrCTL. Receptor ligand systems involed are leucocyte function antigen-1/intercellular adhesion molecule-1 (LFA-1/ICAM-1)(CDlla, CD18/CDM).Tn/LFA-3(CD2, CD58), andtheclonoty-pic T-cell receptor (TCR) strueture NKTa of JT9/JTI0 with ils no!i-MHC-related target antigen TNKtar(4F2), Cytokine produclion can be induced by activating motiockmahmtibodics against CD2R, Antibodies againsi the elonotypic TCR (NKTa) or CD3 had no cylokine-iiiduciiig effect on NrCTL cullured alone, but were able to retrieve the effect of blocking the target antigen on co-cuhured B eells. We eould lurther demonstrate that the inhibition of the TCR/targel antigen interaction eould be overcome by elose cell cell contae! culture conditions. From these findings il i s concluded that the role of the TCR in non-MHC-reslricted cell cell interaction is to facilitate LFA-l. lCAM-1-mediated elfectorlarget adhesion in a specific way rather than lo mediate direct activating signals upon lymphokine produclion or cytoxicity.

    Notes: Abstract Using a selection of ‘classic’ haptens (dinitrohalobenzenes and picryl chloride) and related nonsensitizing analogous, we examined changes in levels of glutathione (GSH) and glutathione disulphide (GSSG) in mouse skin 12 h after their epicutaneous application. We observed that elevation of GSSG levels and/or depletion of GSH levels correlated well with contact allergenic potential. Non-sensitizing analogous failed to perturb GSH/GSSG status. In vitro assays using mouse skin and rat liver microsomal preparations indicated that only the allergenic nitrohalobenzenes initiated NADPH-dependent oxygen utilization, with the activity falling off in the order picryl chloride ≫ DNIB 〉 DNBB 〉 DNCB 〉 DNFB. In addition, an examination of the colour of mouse skin homogenates ex vivo after application of the dinitrohalobenzenes showed significant yellowing (consistent with aromatic nucleophilic substitution) only with DNFB. Our results indicate that, while an aromatic nucleophilic substitution reaction with skin protein can possibly account for the allergenicity of DNFB, it does not seem to occur with DNCB, DNBB or DNIB. These may instead behave mainly as prohaptens which are activated enzymically by NADPH-dependent reductase (s) within the skin, with the concomitant generation of superoxide and hydrogen peroxide, to form potentially protein-reactive free radical and other metabolites. Picryl chloride appears capable of both conjugating directly with proteins by aromatic nucleophilic substitution and undergoing NADPH-dependent metabolism to other potentially protein-reactive metabolites.

    Notes: Abstract The dynamics of coupled biological oscillators can be modeled by averaging the effects of coupling over each oscillatory cycle so that the coupling depends on the phase difference φ between the two oscillators and not on their specific states. Average phase difference theory claims that mode locking phenomena can be predicted by the average effects of the coupling influences. As a starting point for both empirical and theoretical investigations, Rand et al. (1988) have proposed dφ/dt=Δω — K sin φ), with phase-locked solutions φ=arcsin(Δω /K), where Δω is the difference between the uncoupled frequencies and K is the coupling strength. Phase-locking was evaluated in three experiments using an interlimb coordination paradigm in which a person oscillates hand-held pendulums.Δω was controlled through length differences in the left and right pendulums. The coupled frequency ωc was varied by a metronome, and scaled to the eigenfrequency ωv of the coupled system K was assumed to vary inversely with ωc. The results indicate that: (1) Δω and K contribute multiplicatively to φ (2) φ =0 or φ = π regardless of K when Δω=0; (3) φ ≈ 0 or φ ≈ π regardless of Δω when K is large (relative to Δω); (4) results (1) to (3) hold identically for both in phase and antiphase coordination. The results also indicate that the relevant frequency is ωc/ωv rather than ωc. Discussion high-lighted the significance of confirming φ=arcsin(Δω/K) for more general treatments of phase-locking, such as circle map dynamics, and for the 1∶1 phase-entrainment which characterizes biological movement systems.

    Notes: Summary An Ac-derived, two-component transposable element system has been developed and analyzed with respect to its use in Arabidopsis thaliana. This system consists of an immobilized Ac element (“Ac clipped wing”, Accl) as the source of transactivating transposase and a nonautonomous “Ds” element, DsA, which is inserted into a chimaeric neomycinphosphotransferase gene used as excision marker. After separate introduction of Acc1 and DsA into Arabidopsis thaliana, progeny analysis of crosses between five different Accl lines and seven different DsA lines shows that: (1) different Accl lines differ greatly in their capacity to transactivate DsA; (2) different DsA lines do not differ significantly with respect to DsA transactivation by one Accl line; (3) reintegration of excised DsA elements, both at (genetically) linked and unlinked sites, occurs in about 50% of the excision events; and (4) plants with a high rate of somatic excisions can be used as source of new DsA transpositions, allowing the creation of a large number of independent DsA insertions.

    Notes: Abstract Collective properties of206,208Pb have been investigated with gamma-ray spectroscopy using nuclear and electromagnetic excitation by208Pb projectiles. From the measurement of particle-particle-γ and particle-particle-γ γ coincidences a new energy level in208Pb at 5.683 MeV was observed and the data suggest an interpretation as a 2-phonon octupole vibrational state with spin 4+ or 6+. For this state one finds the same dynamical deformation parameterβ 3 as for the first-excited collective 3− state. The results are compared to predictions of models which consider, in particular, the aspects of the quadrupole interaction and the interaction of the 2-particle pairing vibration with the 2-phonon octupole vibration. For the206Pb+208Pb system at 6.2 MeV/u the nuclear potential could be determined from the measured inelastic cross sections.

    Notes: Abstract Nasal absorption of O-(N-morpholino-carbonyl-3-L-phenylaspartyl-L-leucinamide of (2S,3R,4S)-2-amino-1-cyclohexyl-3,4-dihydroxy-6-methylheptane (I), a renin inhibitor, was evaluated in two rat nasal models, one involving surgery and the other requiring no surgical intervention. Oleic acid/monoolein emulsion formulations were tested along with a control PEG 400 solution. The percent absolute bioavailability of the compound was enhanced from 3–6% (PEG 400 solution) to 15–27% when the emulsion formulations were used. The different nasal model techniques (with and without surgery) did not produce any statistical difference in the absolute bioavailability values for I. Emulsion formulations did not produce appreciable damage as assessed morphologically. It is suggested that emulsion formulations containing membrane adjuvants such as oleic acid and monoolein can be used to enhance the nasal delivery of low-bioavailable, lipid-soluble drugs.____________________
