    Notes: Summary The oral bioavailability of methotrexate is variable and may be dose-dependent. The absorption of ‘interval’ oral methotrexate, which is given between cycles of chemotherapy, is unknown. The bioavailability of oral methotrexate has been studied in eight patients, acting as their own controls, to assess the effect of subdivision of the dose, the formulation, and the timing of the methotrexate within the chemotherapy cycle. The mean bioavailability for all the oral methods of administration was 28.2%±3.7% compared with the same dose given IV. Absorption was uninfluenced by subdivision of the dose, liquid or tablet formulation, or administration on day 1 or day 10 of the chemotherapy cycle.

    Notes: Summary A range of clinical and industrial gloves have been evaluated to determine their ability to exclude penetration by cytotoxic drugs. Radiolabelled cyclophosphamide, methotrexate, daunomycin, cytosine arabinoside, and vincristine sulphate were studied in an equilibrium dialysis system in which the glove material was used as the dialysis membrane. The thicker gloves were the most effective, but very little drug crossed any of the gloves tested under laboratory conditions. Further studies to evaluate gloves in conditions of clinical use are now needed.

    Notes: Summary Experimental and clinical evidence indicates that bleomycin by continuous infusion is superior to intermittent administration. Continuous infusion is less convenient, however. It has been suggested that a suspension of bleomycin in sesame oil, given by IM injection, simulates a continous infusion. The pharmacokinetics of this formulation have been compared with those of bleomycin in saline following IM injection, in six patients. The pharmacokinetic profiles of the two formulations were similar. The only difference between the profiles was the long terminal half-life at very low concentrations between 12 and 48 h after injection of the oil suspension. This difference is of unknown, but doubtful, clinical significance.

    Notes: Summary Six healthy volunteers received an iv infusion of 317 mg lysine theophylline (equivalent to 197 mg anhydrous theophylline) in order to calculate theophylline clearance by standard methods. They subsequently received a 20 minute inhalation of nebulised lysine theophylline. Serum and salivary theophylline concentrations were measured and all saliva was collected for the first hour. From these concentrations estimates were made of the distribution of theophylline into the blood and saliva with 40% to 94% identified in the blood. Very high salivary concentrations were reached during the inhalation phase with saliva: serum concentration ratios of between 60 and 1600.

    Notes: Abstract The development of orthopyroxene-Fe/Mg ferrite symplectites associated with olivine is discussed with respect to the chemical reactions by which they form. Previously proposed reactions are presented graphically and the differences between them are reviewed. With the exception of exsolution, these are all discontinuous reactions in the sense that olivine is replaced by the two-phase symplectite assemblage. Olivine-hosted symplectites developed in the margins of lherzolite xenoliths from Kauai, Hawaii, demonstrate a reaction mechanism which has not been previously documented from natural samples. Original Fo90 olivine in these samples oxidized to a new assemblage consisting of orthopyroxene (En92–95)-Fe/Mg ferrite (Mf35–50) symplectites developed within more magnesian olivine (Fo92–96) hosts. Thus, by this mechanism, olivine of a different composition persists as part of a final three-phase assemblage. As oxidation advanced, the compositions of all three product phases became continuously more magnesian and the stoichiometric coefficients of the orthopyroxene and Fe/Mg ferrite continuously increased, whereas those of the product olivine decreased in the mass-balance equations. These characteristics demonstrate that the reaction was controlled by oxygen diffusion into the xenoliths from the highly oxidized alkali picrite melt in which they were entrained. Thermodynamic calculations suggest that a gradient in oxygen fugacity of 100.9 bars existed across the xenolith rims and resulted in compositional gradients of 4 mol% fayalite and ferrosilite and 15 mol% magnetite.

    Notes: Summary Some primitive pea lines, e.g. cultivar Afghanistan, are resistant to nodulation by most strains of Rhizobium leguminosarum. However the Turkish strain TOM can nodulate cv. Afghanistan in addition to commercial pea varieties, and this extended host range is a property of its symbiotic plasmid, pRL5JI. A gene bank was constructed using DNA from a strain of R. leguminosarum containing pRL5JI. Following transfer to a strain of R. leguminosarum that had been cured of its symbiotic plasmid, two derivatives were isolated that contained cloned nodulation determinants, and were able to nodulate both cv. Afghanistan and a commercial pea variety. In addition, these clones conferred the ability to nodulate peas to a strain of R. phaseoli that had been previously cured of its symbiotic plasmid. One of these clones was subjected to mutagenesis with transposon Tn5, and 11 mutants were identified that were affected in nodulation ability. The sites of Tn5 insertions were mapped using restriction endonucleases and all were found to be within a region of 5 kb. The mutants fell into three classes on the basis of their map positions and their phenotypes on the two different pea lines tested. One class of mutants was affected in gene functions that were common to the nodulation of both pea hosts; a second class was impaired specifically in the nodulation of the commercial pea variety; a third class of mutant failed to confer on a ‘normal’ strain of R. leguminosarum the supplementary ability to nodulate cv. Afghanistan.

    Notes: Summary Selection was made for the transposition of the kanamycin resistance transposon Tn5 from a location on the chromosome of R. leguminosarum into a transmissible, bacteriocinogenic plasmid that also carries genes required for the induction of nitrogen-fixing nodules on peas. One hundred and sixty independent insertions into transmissible plasmids were isolated. When these plasmids were transferred by conjugation into a non-nodulating strain, which carries a deletion in one of its resident plasmids, of the 160 isolates tested 14 yielded transconjugants that formed nodules that did not fix nitrogen (Fix-) and in a further 15 cases the transconjugants were unable to form nodules (were Nod-). When transferred to a symbiotically proficient strain (i.e. Nod+ Fix+) none of the transconjugants was symbiotically defective; thus the mutations were not dominant. When kan was transduced from the clones that generated Fix- transconjugants into a Fix+ recipient the majority of transductants inherited Fix- indicating that the insertion of Tn5 had induced the symbiotic mutations. Transduction of kan, from the clones that failed to donate Nod+ by conjugation to strain 6015, occurred at barely detectable frequencies and it was not possible to demonstrate transduction of Nod-. kan was co-transduced with Nod+ from some of the clones and some of these transductants also inherited the ability to produce medium bacteriocin and to transfer at high frequency by conjugation. Thus the genes for all these characters are closely linked.
