    Notes: Summary The genePDC1 ofSaccharomyces cerevisiae coding for pyruvate decarboxylase (E.C. 4.1.1.1.) was used as a hybridization probe to detect gene sequence homologies in different strains ofSaccharomyces cerevisiae and in other yeast species. Additionally six other genes coding for glycolytic enzymes as well as the genesURA3 of the pyrimidine synthetic andTRP1 of the tryptophan synthetic pathways were used. A restriction polymorphism for the BamH1 fragments carrying thePDC1 gene was evident within the speciesSaccharomyces cerevisiae. All strains definitely declared asSaccharomyces cerevisiae showed the same restriction patterns. Hybridizations of different intensities were observed only with species in the familySaccharomycetaceae. Hybridizations with different genes showed different degrees of conservation for certain DNA sequences.

    Notes: Abstract We report six cases of presenile (five) and senile (one) progressive dementia with a mild-to-marked frontal or frontotemporal atrophy and ventricular dilation (Frontal Lobe Degeneration [FLD]). The most prominent microscopic features were layer-dependet neuronal depletion of the cortex, spongiosis, and cortical and subcortical gliosis. Five cases showed additional degeneration of the S. nigra, and two also had motor neuron disease. Despite the absence of Pick cells and bodies, such cases have many features in common with Pick atrophy. Because Pick cells and bodies are inconstantly occurring features in other-wise typical cases of Pick atrophy, they cannot be regarded as inevitable markers of the latter. In our opinion, cases with mild frontal or frontotemporal atrophy as described herein and by others match the grades 1 and 2 in terms of Schneider's classification of Pick atrophy [37]. As long as the etiology of both Pick atrophy and the socalled FLD is unknown, and we finally have to follow morphological criteria for classification, there is apparently no convincing reason to introduce a separate category, such as FLD or FTA, for the cases with moderate or mild frontal atrophy and dementia of frontal lobe type, which can be sufficiently classified with the Pick spectrum of lobar atrophy.

    Notes: Abstract Polyadenylated transcripts homologous to highly repetitive DNA were found in root tips ofVicia faba by Northern blot hybridization. Electron microscope autoradiography using [3H]uridine as a probe revealed transcription of condensed chromatin in various higher plants. This is consistent with the general rule that highly repetitive DNA is located within condensed chromatin, but it is new that this chromatin fraction is active in RNA synthesis to a considerable amount. Semi-quantitative comparison of the intensity of transcription in species with widely differing 2C DNA contents by means of light microscope autoradiography revealed an inverse relationship between the amount of 2C DNA (and condensed chromatin), and the rate of RNA synthesis.

    Notes: Abstract Low-voltage-activated (1-v-a) and high-voltage-activated (h-v-a) Ca2+ currents I Ca were recorded in whole-cell voltage clamped NG108-15 neuroblastoma x glioma hybrid cells. We studied the effects of arachidonic acid (AA), oleic acid, myristic acid and of the positively charged compounds tetradecyltrimethyl-ammonium (C14TMA) and sphingosine. At pulse potentials 〉−20 mV, AA (25-100 μm) decreased 1-v-a and h-v-a I Ca equally. The decrease developed slowly and became continually stronger with increasing time of application. It was accompanied by a small negative shift and a slight flattening of the activation and inactivation curves of the 1-v-a I Ca. The shift of the activation curve manifested itself in a small increase of 1-v-a I Ca at pulse potentials 〈−30 mV. The effects were only partly reversible. The AA effect was not prevented by 50 μm 5, 8, 11, 14-eicosatetraynoic acid, an inhibitor of the AA metabolism, and not mimicked by 0.1–1 μm phorbol 12, 13-dibutyrate, an activator of protein kinase C. Probably, AA directly affects the channel protein or its lipid environment. Oleic and myristic acid acted similarly to AA but were much less effective. The positively charged compounds C14TMA and sphingosine had a different effect: They shifted the activation curve of 1-v-a I Ca in the positive direction and suppressed 1-v-a more than h-v-a I Ca; their effect reached a steady-state within 5–10 min and was readily reversible. C14TMA blocked 1-v-a I Ca with an IC50 of 4.2 μm while sphingosine was less potent.

    Notes: Abstract Experimental results on the optical absorption in Li doped NiO are presented. Upon Li doping, considerable oscillator strength is found to appear throughout the fundamental absorption gap until at 2% doping the d-d excitations usually observed within the gap are completely overwhelmed by these new excitations. Low energy REELS measurements on the same set of samples do not show the same behaviour. We briefly discuss the compatibility of our findings with results of previous electron spectroscopic results and with theoretical scenarios.

    Description / Table of Contents: Zusammenfassung Während viele Berichte belegen, daß Pertussis eine häufige Krankheit auch im Erwachsenenalter ist, gibt es nur wenige Studien zum klinischen Verlauf der Krankheit jenseits der Kindheit. Im Rahmen einer Haushalt-kontaktstudie zum Nachweis der Wirksamkeit einer azellulären Pertussis-Vakzine wurden in einem Studienzentrum mit bekannt niedriger Pertussis-Durchimpfungsrate Erwachsene mit Keuchhusten identifiziert und Symptome erfragt. Innerhalb von 2 Jahren wurden 257 Patienten mit Pertussis in 121 Familien gefunden. Davon waren 79 Erwachsene (30,7%) im Alter zwischen 19 und 83 Jahren (Durchschnittsalter: 36 Jahre). Das Verhältnis männlich zu weiblich betrug 1:1,8. Husten wurde von 91% der Erwachsenen angegeben (durch-schnittliche Dauer: 54 Tage). Er dauerte in 80% der Fälle ≥ 21 Tage an. Inspiratorischer Stridor war selten (8%), dagegen war der Husten häufig von Erbrechen und/oder Würgreiz gefolgt (53%) oder störte den Schlaf der Patienten (52%). Dies ist die erste Studic, in der anfallsweise auftretender Schweißausbruch als Symptom bei Pertussis beschrieben wird (14%). Über pharyngeale Symtome, (37%), Influenza-ähnliche Symptome (30%), Niesanfälle (22%), Heiserkeit (18%), schmerzhafte Sinus (16%) und Kopfschmerzen (14%) wurde ebenfalls berichtet. Komplikationen wurden bei 23% der erwachsenen Patienten beobachtet. Unabhängig vom Alter sollte die mikrobiologische Diagnostik bei allen symptomatischen Kontaktpersonen eines Patienten mit Pertussis und ebenso bei jedem Patienten mit lang anhaltendem Husten konsequent durchgeführt werden, damit die weitere Ausbreitung des Erregers zu einem möglichst frühen Zeitpunkt verhindert werden kann.
    Notes: Summary There is increasing evidence that pertussis occurs frequently in adults, but there is limited information on the clinical course of this disease beyond childhood. A household contact study on the efficacy of an acellular pertussis vaccine was used to study the symptoms of pertussis in adults. Among 257 patients with pertussis identified in 121 families during a two-year period in one study center with a low whole-cell pertussis-vaccine uptake, 79 (30.7%) were adults, aged 19–83 years (mean age: 36 years) with a 1:1.8 male to female ratio. Ninety-one percent of the adults suffered from coughing (mean duration: 54 days), and in 80% this cough lasted ≥ 21 days. Whoops were rare (8%), whereas cough followed by vomiting and/or choking (53%) and cough disturbing sleep (52%) were common. This is the first report to describe sweating attacks as symptom of pertussis (14%). Pharyngeal symptoms (37%), influenza-like symptoms (30%), sneezing attacks (22%), hoarseness (18%), sinus pain (16%) and headaches (14%) were also observed. Various complications were seen in 23% of the patients. In order to minimize the spread of the organism, micro-biological diagnostics should be vigorously applied to all symptomatic contacts of a patient with pertussis but also to all patients with long lasting cough — irrespective of age.

The molecular conformation is right-handed helical with three α-(5 → 1 hydrogen bonds) and three β-turns (4 → 1 hydrogen bonds). All but two of the hydrogen bonds are significantly longer than the usual value and show bifurcation to some extent. The α/310r-helical nonapeptide molecules are arranged head-to-tail along the a direction. The resulting linear antiparallel chains are linked by a weak intermolecular hydrogen bridge, thus forming a two-dimensional layer structure in the ab plane. The conformation of this nonapeptide is almost identical with that of the corresponding C-terminal part found by x-ray crystallography of the eicosapeptide alamethicin.

    Notes: The x-ray structure of Boc-L-Ala-Aib-Ala-Aib-Ala-Glu(OBzl)-Ala-Aib-Ala-Aib-Ala-OMe(I) represents the first α-helix determined by direct methods. This undecapeptide is a model of the N-terminus of alamethicin, and it exhibits voltage-dependent pores in bilayer membranes at a higher voltage and concentration than alamethicin. The molecule crystallizes in the monoclinic space group P21 with a = 10.602(1), b = 23.884(3), c = 13.622(1) Å, β = 95.61(6)°, and Z = 2. It adopts a right-handed α-helical conformation in the solid state with intramolecular 5 → 1 hydrogen bonds. An additional intramolecular hydrogen bond is bifurcated, forming a stronger 4 → 1 interaction (i.e., a β-turn III) and a weaker 5 → 1 interaction, thus prolonging the α-helical part up to 9 residues. The α-helix radius of 2.1 Å, the height per residue (distance Ni … Ni + 4) of 1.53 Å, the resulting length of the α-helical part of 13.8 Å (9 residues) resp. 15.3 Å (10 residues), the van der Waals radius (4.7 Å), and the minimal diameter of pores formed by aggregation of 3-10 α-helices were calculated omitting the Glu(OBzl) side chain. In the crystal, the α-helices are linked head to tail via two hydrogen bridges forming continuous chains. Adjacent helices are oriented in antiparallel with their helix axes and have only van der Waals contacts.
