    Notes: Summary The gene responsible for xeroderma pigmentosum (XP) group A has recently been cloned and designated XPA gene. Previous studies have shown that most Japanese XPA patients have homozygous mutations for the splicing site of intron 3 of the XPA gene, which was recognized by restriction endonuclease (RE) AlwNI (AlwNI mutation). Other mutations found to date have been the nonsense mutation at codon 228 in exon 6, recognized by RE Hphi (HphI mutation), and at codon 116 in exon 3, recognized by RE Msel (Msel mutation). Using polymerase chain reaction restriction fragment length polymorphism (PCR-RFLP) analysis, we examined the point mutations of the XPA gene in 16 XPA patients, their parents, and their four asymptomatic siblings. We found that eight patients were homozygous for the AlwNI mutation, two were compound heterozygotes for the AIwNI mutation and the Hphl mutation, one was a compound heterozygote for the AIwNI mutation and the MseI mutation, three were compound heterozygotes for the AlwNl mutation and an unidentified mutation, and two were compound hcterozygotes for the Hphl mutation and an unidentified mutation, investigation of their clinical features suggested that the four patients who were heterozygous for the Hphl mutation and the AlwNI or an unidentified mutation had milder clinical manifestations such as later development of skin cancers and milder neurological deterioration, than those patients who were either homozygous for the AlwNI mutation or heterozygous for the AlwNI mutation and MseI mutation.PCR-RFLP analysis of the XPA gene in the four asymptomatic siblings of the XPA patients revealed that two were carriers of the mutated XPA allele, one was not a carrier, and one was not diagnosed because of the presence of an unidentified mutation.These data indicate that determination of the point mutation of the XPAC gene is important in predicting the clinical course in XPA patients. In addition, this method is useful for the detection of asymptomatic carriers in affected families, who have not been identified with conventional techniques.

    Notes: Abstract A new type of PbO2-coated metal anode, useful for electrochemical processing requiring the use of high-overpotential anodes, was prepared by applying to a titanium substrate an undercoating of Ti-Ta oxides, then covering the undercoating with an intermediate coating of stress-free α-PbO2 deposit, and finally covering the intermediate coating with a Ta2O5 particle-admixed β-PbO2 deposit. The anode thus produced was found to be superior to the anodized Pb or platinized Ti anode in respect of oxygen overvoltage, and corrosion resistance, even in solutions containing organic solvents. The possibility for use in chromium plating is also indicated.

    Notes: Abstract We have carried out a simultaneous observation of radar and optical meteors with the MU radar (Middle and Upper Atmosphere Radar), Shigaraki and TV camera systems. We usually obtained about 20 meteors per an hour with 85 mm lens, but very small part of them are simultaneously observed by the MU radar (〈 5%), suggesting the significance of rectangular scatterring. We have analyzed about 20 simultaneous meteors with magnitudes from 0 to +5.5, most of which are overdense meteors. For Geminid meteors, a linear relation between the logarithm of the echo duration and the absolute magnitude of the TV meteor, was deduced.

    Notes: Abstract We have carried out double-station TV meteor observations between 1990 and 1994. The orbits of 326 meteors have been determined from doubly observed meteors, and radiant distributions are studied. The mean magnitude of the observed meteors was as faint as +4.7, since I.I. (Image Intensifier) and Video cameras were used. Radiants were widely distributed over the celestial sphere. The velocity distribution showed some similarity with the distribution predicted by the theoretical radiant distribution from comets rather than that from asteroids. In all 13 showers including both major and minor meteor showers were detected from radiant distributions of the observed meteors; from the orbital elements and meteor velocities as well as from the radiant directions.

    Notes: Abstract  When the isocitrate lyase gene, containing 5′-upstream and 3′-flanking regions, of an n-alkane-assimilating yeast Candida tropicalis was introduced into Saccharomyces cerevisiae, the enzyme was functionally overexpressed in the cells grown on acetate. The amount of the recombinant isocitrate lyase expressed in S. cerevisiae was as much as 30% of the total soluble proteins in the cells, being comparable to that with GAL7 functional under the control of galactose. The expression was also observed when the cells were grown on glycerol, lactate, ethanol or oleate. These facts indicate that the isocitrate lyase gene upstream region (UPR-ICL) contains a strong promoter functional in S. cerevisiae. UPR-ICL is active as a promoter on cheap carbon sources such as acetate and non-conventional carbon sources such as oleate, whereas many conventional strong promoters demand relatively expensive sugars or sugar derivatives. Therefore, it is promising to construct an economical recombinant protein production system by using UPR-ICL.

    Notes: Abstract The stabilization mechanism of Sodium Polystyrene Sulphonate (PSSNa) on coal water mixture (CWM) has been examined using the following colloid chemical concept. It is realized that the stabilization of the CWM is due to an increase in electrostatic repulsion between the coal particles and the electrostatic repulsion is influenced strongly by the concentration of metal cations, especially Ca2+ eluted from the coal surface. The adsorption isotherm of PSSNa on the coal surface indicates a weak affinity type and the desorption amount of PSSNa is tremendously small in compared with the amount of adsorption. This indicates that a lot of PSSNa adsorbed weakly has been eliminated from the surface in the pre-washing process of desorption experiment. Furthermore, it appears that the ζ-potential determined by the Acoustosizer for concentrated coal suspension shows higher values than the values determined by the usual electrophoresis, and that the high values hold for a wide range of salt concentrations in the medium. All these results indicate that much PSSNa is adsorbed weakly on the coal surface and the component plays a role in the stability character of CWM, where a large contribution of depletion stabilization effect can be expected.
