    Notes: Abstract: The injection of phorbol esters into the eyes of dark-adapted teleost fish can mimic light effects in the retina and induces corresponding synaptic plasticity of horizontal cells (HCs). It is therefore very likely that protein kinase C (PKC) mediates light-induced synaptic plasticity. In the present study, we investigated the distribution of PKC, the phorbol ester receptor, in isolated HCs and in the whole retina by using tritiated phorbol 12,13-dibutyrate ([3H]PDBu). The binding characteristics analyzed for HC homogenates and retinal homogenates revealed that [3H]PDBu binding is time dependent, specific, saturable, and reversible. Binding sites in HCs displayed a dissociation constant of 11.5 nM and a total number of 2.8 pmol/mg of protein. Autoradiography revealed that [3H]PDBu labeling is present in all retinal layers, including HCs, where it is associated with the somata. Furthermore, the treatment with PDBu strongly affected the endogenous phosphorylation of several membrane, cytosolic, and HC proteins and led to PKC activation as measured by H1 histone phosphorylation. In HCs, the treatment with PDBu in particular affected the amount of 32P incorporated into a group of phosphoproteins (68, 56/58, 47, 28, and 15 kDa) that were recently shown to be affected by light adaptation. These proteins might therefore be considered as important components of the observed morphological and physiological synaptic plasticity of HCs in the course of light adaptation.

    Notes: The nucleation of diamond on Si is enhanced for negative substrate bias of 200–250 V. We show that the ion flux is the critical factor causing the enhanced nucleation. The ion energy distribution has a maximum at about 80 eV, the optimum to subplant C ions into a-C. We propose that subplantation causes deposition of nanocrystalline graphitic C, and that diamond nucleates where the graphitic planes are locally oriented perpendicular to the surface. An atomic model is proposed that allows a matching of the diamond, graphite, and Si lattice. © 1995 American Institute of Physics.

    Notes: It is shown that for a cathode consisting of compressed graphite powder, the cathodic arc discharge is confined within deep erosion holes and the macroparticle emission is greatly reduced. Electron energy loss spectroscopy and scanning electron microscopy show that smooth tetrahedral amorphous carbon films with up to 85±10% sp3 bonding can be deposited without the use of magnetic filters. The new carbon arc discharge process holds potential for deposition of smooth tetrahedral amorphous carbon films on large area substrates due to the elimination of the magnetic filtering stage. © 1995 American Institute of Physics.

    Notes: European domestic pigs are derived from the European wild pig by genetic selection and differ in their muscular-growth ability. Thus, it was the aim of this study to investigate the consequences of genetic selection on muscle composition and fibre size in the gracilis muscle. Fibre typing based on the ATPase reaction revealed that this muscle in wild pigs is composed mainly of type-I and -IIa fibres, whereas, in domestic pigs, type-IIb fibres were predominant. For all fibre types, domestic pigs had about threefold larger fibres than wild pigs. Type-I fibres tended to be the smallest fibres in domestic pigs, but the largest fibres in wild pigs, which may be due to long-term training effects in the free-ranging animals. Giant fibres as an indicator of degeneration were obvious only in the domestic-pig samples. Their occurrence, as well as the larger fibre size and the predominance of type-IIb fibres in domestic pigs, may be ascribed to high concentrations of growth hormone. It is concluded that selection for muscular growth may favour metabolic dysbalances within the muscle and, therefore, degenerative processes.

    Notes: The immune response of adult BALB/c mice against the bacterial antigen dextran B13558 (Dex) is well characterized as thymus independent (TI type 2) and Igha linked. The antisera consist of mainly IgM/ antibodies directed against the λ(1 → 3) glucosidic linkage. This study describes the immune reponse against the α(1 → 3) linkage in the thymus dependent form (TD), i. e. tetra- or heptasaccharides (N4 or N7) of glucose as hapten coupled to chicken serum albumin (CSA) as carrier. Whereas athymic BALB/c-nu/nu mice did not respond to the TD antigens N4-CSA and N7-CSA, euthymic BALB/c showed high anti-Dex antibody titres of IgM and, after 2° immunization, a class switch to IgG (mainly IgGl) isotypes with λ light chains. The hapten N4 inhibited Dex-binding of M104E or of antisera from Dex or N4-CSA or N7-CSA immunized mice at 1.7–10 × 104M. The idiotype composition of these antibodies resembled those after Dex immunization. We conclude that the same Dex-specific precursor B cells have been stimulated by either form of antigen. The ontogenic development of a Dex-specific response could not be accelerated by the aid of T cells, even of adult origin. It seems, therefore, that the maturation of antigen specific B cells is the limiting step in ontogeny.

    Notes: The formation of spinules at the terminal dendrites of retinal horizontal cells with the onset of light and their subsequent retraction during darkness is a remarkable example of synaptic plasticity where sensory experience modifies reversibly, and on a time scale of minutes the ultrastructure of synaptic connectivity. The signals and the subsequent intracellular cascades underlying the prominent morphological alterations are only partially understood. We show here that lowering the external calcium concentration did prevent dark- and AMPA-induced retraction of spinules in an eyecup preparation. Furthermore, spinule retraction was prevented in vivo by the injection of calmidazolium, an inhibitor of calmodulin, into the eyeball, and also by the injection of KN-62, an inhibitor of Ca2+/calmodulin-dependent protein kinase II (CaMkll). We conclude that local Ca2+ influx through AMPA-gated channels followed by activation of CaMkll is an important step for spinule retraction during dark adaptation. The phosphorylation patterns of phosphoproteins derived from purified horizontal cells was affected by the inhibitors of camodulin and CaMkll respectively. Some of the affected phosphoproteins appeared to be cytoskeleton-associated proteins, including GAP-43. Based on these observations, a putative scenario for the retraction of spinules is proposed.

    Notes: To test whether eosinophil recruitment after pulmonary allergen challenge is associated with interleukin (IL)-5 in patients with asthma, we performed segmental bronchoprovocation (SBP) with saline, and with low and high dosages of ragweed extract in six patients with allergic asthma. Bronchoalveolar lavage (BAL) of the challenged segments was performed 5 min after challenge (immediate BAL fluid) and repeated 24 h later (late BAL fluid). Allergen challenge resulted in recruitment of eosinophils, and increased levels of eosinophil-active cytokines. A bioassay showed the predominant eosinophil-active cytokine in the late BAL fluids to be IL-5. Analysis of the late BAL fluids revealed that IL-5 levels correlated with the numbers of eosinophils and lymphocytes. This study provides evidence that IL-5 is a critical cytokine associated with eosinophil and lymphocyte recruitment into the airways of patients with asthma following exposure to allergen.

    Notes: Abstract For the purposes of a thesis at the Institute of avalanche and torrent control at the university of agriculture in Vienna a comparison of two avalanche-models, a hydraulic one from Voellmy/Salm/Gubler and a statistical one from Laatsch/Zenke/Dankerl with exemplary exactly known avalanches of Tyrol and Switzerland was started in 1993. The result of this work was that both models failed by the calculation of avalanches with high recurrence intervals (over 300 years). For the calculation of avalanches with recurrence intervals under 300 years the results of both models are regular. The conclusion is that a combination of two models i.e. a hydraulic one improved by statistical calculations will be the best.
