    Notes: Diamond films were successfully synthesized in both parallel-plate radio frequency (rf: 13.56 MHz) CH4 and CH3OH plasmas with injection of H and OH radicals generated in the remote microwave (2.45 GHz) H2/H2O plasma. Effects of H, OH, and CH3 radicals on the diamond film formation in the rf plasma reactor were investigated by the formation of diamond films employing radical injection technique and the measurement of density in the plasma. Under the condition of diamond film formation, CH3 density was measured by infrared diode laser absorption spectroscopy (IRLAS). The kinetics of CH3 in rf CH4 and CH3OH plasmas with injection of H and OH radicals were evaluated from the results of optical emission spectroscopy and lifetime of CH3 radicals estimated by IRLAS. © 1997 American Institute of Physics.

    Notes: This work demonstrates that the technique of guiding atoms through hollow optical fiber escorted by evanescent light is useful as a novel scheme of optical atom deposition. To show the feasibility of fabricating micron-sized structures with nanometric depth on a substrate, we measure the spatial distribution of the guided atom flux with a hot-wire detector. Moreover, precise control of the deposition rate is illustrated with a 1.4-μm-hollow fiber via photoionization spectroscopy. The ratio of the guided atom flux relative to the background transmission is enhanced up to 80-fold with a slightly tilted hollow fiber. © 1997 American Institute of Physics.

    Notes: Lysozyme has a bactericidal activity for some strains of Gram-positive bacteria, by enzymatic cleavage of peptidoglycans that constitute the cell wall. Hen egg-white lysozyme (HEL) was tested in vitro for effects on biological activities of lipopolysaccharides from periodontopathic Gram-negative bacteria, Actinobacillus actinomycetemcomitans, Prevotella intermedia and Porphyromonas gingivalis. HEL inhibited a wide range of activities of these lipopolysaccharides, including activation of Limulus amoebocyte lysate, stimulation of human leukocytes to secrete tumour necrosis factor-alpha, polyclonal activation of mouse B cells, and promotion of osteoclastic differentiation in mouse bone marrow cultures. The anti-endotoxic activity of HEL may be worthy of being intended for periodontal therapy.

    Notes: Abstract Using immunocytochemistry we have analyzed 8 pituitary oncocytomas, 14 null cell adenomas, and 2 oncocytomas of the parotid gland (Warthin’s tumor). The proportions of adenoma cells that are positive for mitochondrial protein (MP), cytochrome oxidase (COX), and manganese-superoxide dismutase (Mn-SOD) were significantly higher in pituitary oncocytomas than in null cell adenomas (MP P 〈 0.001, COX P 〈 0.001, Mn-SOD P 〈 0.05). In pituitary oncocytomas, MP-positive cells were distributed unevenly but in clusters or in islets admixed with some MP-negative cells, and corresponded to COX-positive cells. In contrast, almost all of the oxyphilic epithelial cells of Warthin’s tumor were positive for MP, COX, and Mn-SOD. On the other hand, both pituitary tumors displayed similar findings with regard to the proportion of adenoma cells immunoreactive for copper/zinc-SOD and adenohypophysial hormones, the Ki-67 (MIB-1) proliferating cell index, and the mean number of argyrophilic nucleolar organizer regions. It was confirmed that immunocytochemical identification of MP and COX is useful for distinguishing pituitary oncocytomas from null cell adenomas. Although it remains to be determined whether oncocytomas originate from oncocytic changes of tumor cells or from neoplastic transformation of oncocytic cells, it appears that tumorigenesis of pituitary oncocytomas differs from that of Warthin’s tumor.

We previously generated a monoclonal antibody (mAb) against a putative pathogenic epitope on native type II collagen (CII) for the induction of collagen-induced arthritis in mice (mAb1), and an anti-idiotypic mAb which appears to possess the internal image of the CII epitope (mAb2). In the present study, the structural basis of the antigen/mAb1 and mAb1/mAb2 interactions was examined. When partially SH-reduced mAb1 was analysed on Western blots, only fragments containing both heavy (H) and light (L) chains were recognized by mAb2. When mAb2 was partially SH-reduced, only fragments containing both H and L chains were recognized by mAb1. H and L chains were separated from mAb1 in a reduced, denatured condition, and each chain and a mixture of the two were refolded. mAb2 reacted specifically to the renatured whole IgG molecule of mAb1, but not to the refolded L or to H chains. Recombinant single chain Fv (scFv) generated from mAb1 and mAb2 had properties of the original mAbs, whereas genetical ly constructed chimeric scFvs, consisting of VH from mAb1 and an irrelevant VL , or VL of mAb1 and an irrelevant VH , did not react either to CII or to mAb2. Thus, interactions among CII, mAb1 and mAb2 appear to depend on quaternary structures containing different protein subunits. These observations support the internal image property of the mAb2. In addition, this dependency on quaternary structure for recognition of proteins may also be relevant to other protein-protein interactions.

    Notes: Abstract We examined chemosensitivity to 5-fluorouracil (5-FU) in four human gastric cancer cell lines, by analyzing the expression of p53 and its related genes. Treatment with 1mM 5-FU induced variable degrees of apoptosis in the cultured cells. The apoptotic indices 72 h after treatment were approximately 14% in MKN-74 (wild-type p53 gene), 12% in MKN-45 (wild-type), 3% in MKN-28 (mutated) and 0.5% in KATO-III cells (deleted), respectively. On the other hand, 50 μM 5-FU had little effect on the induction of apoptosis in MKN-74 cells, the value being approximately 2% after 72 h. Induction of P53 expression was noted 3 h after initiating the treatment, followed by the induction of P21/Waf1 after 6 h in both MKN-74 and MKN-45 cells. The same expression mode was noted in MKN-74 treated with 50 μM 5-FU. Conversely, the level of P53 expression was constant in MKN-28 cells and absent in KATO-III cells, in which P21/Waf1 had never been induced. The Bax/Bcl-2 expression ratio was gradually elevated for up to 72 h in MKN-74 and MKN-45 cells treated with 1mM 5-FU; in contrast, it was unchanged in MKN-28 and KATO-III cells, and MKN-74 treated with 50 μM 5-FU. These results might indicate that (1) 1mM 5-FU induces apoptosis in cultured gastric cancer cells carrying the wild-type p53 gene, but not those carrying the mutated type or a gene deletion, and (2) the elevated Bax/Bcl-2 expression ratio plays a more crucial role than the higher expression of P21/Waf1 in the induction of p53- gene dependent apoptosis.

    Notes: Abstract Phyllostachys pubescens Mazel ex Houzeau de Lehaie is one of the largest bamboo species with a leptomorph root system in the world. The species originates in China and has been naturalized in the neighboring countries. It was introduced in 1746 into Japan because of the economic value of the young sprouts and culm woods. It escaped from the planted areas and expanded by invading the original vegetation. In order to clarify the basic ecological characteristics of the species, carbon fixation and cycling were determined in a stand of Phyllostachys pubescens. The standing culm density and average DBH in 1991 were 7100 ha(-1) and 11.3 cm, respectively. The above-ground biomass was 116.5 t ha(-1) for culms, 15.5 t ha(-1) for branches, 5.9 t ha(-1) for leaves and 137.9 t ha(-1) in total. The total above-ground biomass was one of the largest among the world's bamboo communities. The biomasses of rhizomes and fine roots were 16.7 t ha(-1) and 27.9 t ha(-1), respectively. Annual soil respiration was 52.3 t CO(2) ha(-1) yr(-1), the highest among those determined in Japan. The gross production was high: 32.8 t C ha(-1) yr(-1). Allocation of the products to its root system was also high: 34% to gross production and 46% to the fluxes out of the leaves into other compartments of the ecosystem. This resulted in the reduced above-ground net production of 18.1 t ha(-1) yr(-1), which fell within the average range of productivity of forests under similar climate conditions. This paper discusses the correspondence of the allocation pattern with the successful range expansion.
