    Notes: The energetics of Pb film grown at 300 K on two well-defined oxides [a MgO(100) thin film and a p(2×1)-oxide on Mo(100)] have been measured using single-crystal adsorption microcalorimetry. The evolution of the film morphology was followed using Auger electron spectroscopy (AES). An initial heat of adsorption of 103 kJ/mol is observed for the Pb/MgO(100) system. Auger results indicate that this is due to the formation of an ∼20 atom Pb island in the first pulse of Pb gas incident on the MgO(100) surface. This allows the extraction of a Pb–MgO(100) bond energy of ∼32±2 kJ/mol in the small two-dimensional particles formed in the first pulse. As more Pb is deposited onto the MgO(100) surface, the Pb forms three-dimensional islands. The integral of the heats of adsorption up to high coverage indicates an adhesion energy of 76.5±∼20 μJ/cm2 for large 3D Pb particles to the MgO(100) substrate. This indicates a Pb–MgO(100) bond energy of 49±13 kJ/mol at the 3D interface. Similar analyses of the Pb/p(2×1)-oxide on Mo(100) surface give an initial heat of adsorption of 146.2 kJ/mol and an adhesion energy of 82.5±20 μJ/cm2 for thick Pb films. This indicates a bond energy of 52±12 kJ/mol for a Pb atom to this surface at the interface of a thick Pb film. The metal's sticking probabilities as a function of coverage and the coverage-dependent changes in optical reflectivity were measured. For the Pb/MgO(100) system, an initial sticking probability of 0.70 at 300 K was observed, whereas for the Pb/p(2×1)-oxide on Mo(100) surface unit sticking probability was observed. The relationship between these quantities and the adsorption energetics is discussed. © 2001 American Institute of Physics.

    Notes: Urethral epithelial cells are invaded by Neisseria gonorrhoeae during gonococcal infection in men. To understand further the mechanisms of gonococcal entry into host cells, we used the primary human urethral epithelial cells (PHUECs) tissue culture system recently developed by our laboratory. These studies showed that human asialoglycoprotein receptor (ASGP-R) and the terminal lactosamine of lacto-N-neotetraose-expressing gonococcal lipooligosaccharide (LOS) play an important role in invasion of PHUECs. Microscopy studies showed that ASGP-R traffics to the cell surface after gonococcal challenge. Co-localization of ASGP-R with gonococci was observed. As ASGP-R-mediated endocytosis is clathrin dependent, clathrin localization in PHUECs was examined after infection. Infected PHUECs showed increased clathrin recruitment and co-localization of clathrin and gonococci. Preincubating PHUECs in 0.3 M sucrose or monodansylcadaverine (MDC), which both inhibit clathrin-coated pit formation, resulted in decreased invasion. N. gonorrhoeae strain 1291 produces a single LOS glycoform that terminates with Gal(β1–4)GlcNac(β1–3)Gal(β1–4)Glc (lacto-N-neotetraose). Invasion assays showed that strain 1291 invades significantly more than four isogenic mutants expressing truncated LOS. Sialylation of strain 1291 LOS inhibited invasion significantly. Preincubation of PHUECs in asialofetuin (ASF), an ASGP-R ligand, significantly reduced invasion. A dose–response reduction in invasion was observed in PHUECs preincubated with increasing concentrations of NaOH-deacylated 1291 LOS. These studies indicated that an interaction between lacto-N-neotetraose-terminal LOS and ASGP-R allows gonococcal entry into PHUECs.

    Notes: Abstract Background: Single-agent bi-weekly paclitaxel was studied as first-line metastatic treatment for breast cancer in a phase I–II trial. Patients and methods: Thirty-eight women with metastatic breast cancer were enrolled. Thirty-seven are evaluable for toxicity, 35 for response. Results: The MTD was defined at 160 mg/m2 q two weeks with dose limiting toxicity in two patients consisting of hematological toxicity (1) and neurotoxicity (2). Twenty patients were treated at 150 mg/m2, the recommended dose. Response rates were two CRs and nine PRs (overall 61%) at the RD of 150 mg/m2 and three CRs and 11 PRs for an overall RR of 67% for the two top doses. Conclusions: The good drug tolerance, response rates, and convenience over weekly treatment suggest this may be a worthwhile regimen.

    Notes: Abstract We present the results of a microscopic theory of the scattering, transmission, and sticking of 4 He atoms impinging on a zero temperature 4 He slab at near normal incidence. The theory includes coupling between different modes and allows for inelastic processes. The present work focuses on the elastic reflection and transmission of a 4 He atom in the sense that we examine these intensities for atoms which have the same energy as the incident atoms. We find a considerable loss of total intensity due to scattering into multiple excitations. The reflected signal is in qualitative and semi-quantitative agreement with experimental results for 4 He atoms scattered from the surface of bulk helium. The transmission intensity — which has not been measured — shows a very strong energy dependence. Moreover, we show that this dependence is substantially different from the Feynman level theory, which doesn't permit the decay of the single excitation into multiple excitations, and thus cannot describe a reduction in total intensity. In our theory, the major source of decay of elastic transmission and reflection (i.e., sticking) is from the production of ripplons at the liquid-vacuum interfaces.

    Notes: Abstract Recent studies on the nature of global warming indicate the likelihood of an asymmetric change in temperature, where night-time minimum temperature increases more rapidly than the day-time maximum temperature. We used a physically based scenario of asymmetric warming combined with climate change scenarios from General Circulation Models (GCMs) outputs and the EPIC (Erosion Productivity Impact Calculator) plant process model to examine the effects of asymmetric temperature change on crop productivity. Our results indicated that the potential effects of global change on crop productivity may be less severe with asymmetric day-night warming than with equal day-night warming.

    Notes: Abstract Indicators of the condition and sustainability of agricultural lands in five Mid-Atlantic states were measured in 1994. Indicators were selected to reflect crop productivity and land stewardship on annually harvested herbaceous crop (AHHC) land, which covers almost 10% of the land area in this region. Overall, condition of agricultural lands in the region is good. Crops generally yielded more than those grown in the 1980s, with a mean observed/expected yield index greater than 1. The mean soil quality index was slightly better than a "moderate" rating for crop growth. Almost 2/3 of the AHHC land is covered by crop rotation plans, with the remaining land mostly in hay fields. Insecticides were applied to less than 20% of AHHC land, and less than 20% of the land where pesticides were applied has high to moderately high potential for pesticides leaching into groundwater. However, integrated pest management (IPM) is practiced on less than 20% of AHHC land. Hay showed more efficient use of nitrogen than seed crops, and non-tilled sites, which are mostly hay, had more microbial biomass (suggesting more nutrient cycling) than tilled sites. This information could provide a baseline for a long-term monitoring program for agroecosystems in the region.

    Notes: Use of electrosynthetic methodology allows the production of hen egg-white lysozyme (HEWL) either mononitrated at tyrosine 23 or bisnitrated at tyrosines 20 and 23, but never nitrated at tyrosine 53. This is a different sequence from that obtained by the chemical nitrating agent tetranitromethane, and when reduced by dithionite, the selectively modified enzyme can be anchored at pH 5 via the unique aromatic amino group to magnetic beads or other suitable matrices. HEWL so immobilised loses less than 10% of cell-wall lytic activity compared with the approximately 50% loss of activity when immobilised by conventional methodology at pH 9 via essentially random reaction at lysine residues and other functionalities which are nucleophilic at this pH. This result offers promise as a general method for selective protein immobilisation in biosensors and similar applications. © 1998 John Wiley & Sons, Ltd.
