    Notes: In ulcerative colitis, hydrogen sulphide is postulated to impair colonocyte butyrate metabolism, leading to cellular energy deficiency and dysfunction.〈section xml:id="abs1-2"〉〈title type="main"〉Aims:To determine the effects of sulphide exposure on butyrate metabolism and adenosine triphosphate levels of HT29 colonic epithelial cancer cells, and to establish whether energy deficiency can be prevented by increased butyrate concentrations or the presence of glucose.〈section xml:id="abs1-3"〉〈title type="main"〉Methods:HT29 cells were maintained in medium containing 3 mM butyrate, 5 mM glucose, or both substrates. Oxidation rates were measured by 14CO2 release from 14C-labelled substrates. Cellular adenosine triphosphate was assayed using the luciferin/luciferase chemiluminescent method. The effects of sulphide (0–5 mM) on substrate oxidation and adenosine triphosphate levels and of increasing butyrate concentration (0–30 mM) with sulphide were observed.〈section xml:id="abs1-4"〉〈title type="main"〉Results:HT29 cells showed similar energy substrate usage to primary colonocyte cultures. Sulphide exposure inhibited butyrate oxidation and led to a reduction in cellular adenosine triphosphate. This fall was prevented by co-incubation with glucose, but not by increasing concentrations of butyrate.〈section xml:id="abs1-5"〉〈title type="main"〉Conclusions:HT29 cells utilize butyrate as an energy substrate and represent a useful in vitro model of the effects of sulphide on colonocytes. Sulphide inhibits butyrate oxidation and leads to demonstrable energy deficiency, prevented by the presence of glucose but not by increased butyrate concentrations.

    Notes: Interspecific hybridization is known to improve productivity and resistance to diseases in many crops. Therefore, an attempt was made to introgress productivity and disease resistance into chickpea from wild Cicer species. The true F1 hybrids of cultivated chickpea genotypes ‘L550’ and ‘FGK45’ with C. reticulatum were backcrossed twice to their cultivated female parents to minimize the linkage drag of undesirable wild traits. The pedigree method was followed to advance the segregating populations from straight crosses (without backcross) and BC1/BC2 generations to F5–F7. The interspecific derivatives recorded up to a 16.9% increase over the check cultivars and a 25.2% increase over the female parent in a preliminary yield evaluation trial. Of the 22 interspecific derivatives thus derived, four desi and two kabuli lines were further evaluated for seed yield in replicated trials at three diverse locations. These lines possess a high degree of resistance to wilt, foot rot and root rot diseases, and recorded a 6.1–17.0% seed yield increase over the best check cultivars.

    Notes: Intergeneric hybridization between wheat (Triticum aestivum L.) and a wild weedy species, Imperata cylindrica (2n = 20) resulted in the recovery of a high frequency of wheat haploids, which were obtained through the elimination of I. cylindrica chromosomes. Cytological analysis of the root tips revealed the somatic chromosome count of the regenerants equalled 21. Haploid regenerants were also obtained in all the crosses of wheat F1s with I. cylindrica, implying the genotype nonspecific nature of wheat x I. cylindrica hybridization. Variation among wheat F1 hybrids was observed with respect to seed formation (44.9-84.5%), embryo formation (15.1-47.7%) and regeneration (27.0-75.0%) in crosses with I. cylindrica. Comparisons based on the efficiency of I. cylindrica and maize (Zea mays) as pollen sources indicated that Imperata-mediated haploid production is equally efficient. There is natural coincidence of flowering period of I. cylindrica with that of wheat under sub-temperate conditions which is advantageous compared with maize, which cannot be grown during the winter season in the sub-temperate regions.

    Notes: Helminthosporium leaf blight (HLB) is the most important disease constraint to wheat (Triticum aestivum L.) cultivation in the eastern Gangetic Plains of South Asia. A Helminthosporium Monitoring Nursery (HMN) including potential adapted and exotic sources of HLB resistance was developed in Bangladesh, India and Nepal to assess the stability of genetic resistance across locations. The 8th, 9th and 10th HMN assessed the HLB resistance and agronomic traits of 17 wheat genotypes across 20 environments of Bangladesh, India and Nepal in the 1999-2000, 2000-2001 and 2001-2002 cropping seasons, respectively. The area under the disease progress curve (AUDPC) for HLB, grain yield, thousand-kernel weight (TKW), days to heading, days to maturity, and plant height were examined. The 17 genotypes showed a range of variability for disease and agronomic characters. Disease severity (AUDPC) differed in the 3 years and showed the highest values in 2002. The increase in AUDPC in 2002 caused the lowest grain yield, with an average 18% reduction due to HLB. A few genotypes (SW 89-5422, Yangmai-6 and Ning 8201) appeared to have stable HLB resistance across environments. However, most of the higher-yielding genotypes, except BL 1883, were unstable. The results suggest that careful selection of HLB resistance with acceptable grain yield, TKW and plant height may be possible using the wheat genotypes included in the HMN.

    Notes: Experiments were conducted in eight ponds of 0.08 ha each for a period of 6 months to evaluate the production performance combinations of Indian major carps and exotic carps at varied stocking densities and species. The four different treatments evaluated were stocking densities of 5000 and 10 000 fingerlings/ha with three Indian major carps (catla, rohu, mrigal) and combination of three Indian major carps with three exotic carp species (silver carp, grass carp and common carp) in replicate ponds. The mean net production levels recorded in different treatments were 1791 kg/ha, 2385 kg/ha, 2532 kg/ha and 3472 kg/ha in 6 months in treatments with three species combinations at 5000/ha, six species at 5000/ha, three species at 10 000/ha and six species at 10 000/ha, respectively. Correspondingly, the mean growth, survival and feed conversion ratios recorded in four treatments were 432 g, 84.7% and 1.49; 567 g, 86.2% and 1.47; 318 g, 82.1% and 1.69; and 444 g, 80.4% and 1.67, respectively.

    Notes: This paper presents the steady-state and transient behavior of a single-phase self-excited induction generator (SEIG) using a three-phase machine with one shunt and one series excitation capacitors for resistive and inductive loads. The generation scheme consists of one three-phase delta connected induction machine and two capacitors - one connected in parallel with one winding and the other in series with a single-phase load. The dynamic model of the system has been developed as a hybrid model considering the stator phase currents in abc reference frame and the rotor currents in stationary d-q axes reference frame as state variables. The simulated and experimental results are presented for different dynamic conditions such as initiation of self-excitation, load perturbation and short-circuit. The simulated results of the steady-state analysis have been compared with the transient and experimental results and a close agreement between them indicates the accuracy and effectiveness of the approach.
