    Notes: A fifty five year-old man had experienced itching sensation when he applied analgesic plasters for his lumbago. He tried to use several kinds of analgesic plasters, however he felt itching sensation whenever he used them. He applied a plaster containing ferbinac for his left elbow joint’s pain on April 2003. On the next day, an itching erythema developed on the area where the plaster was applied. He was treated with a difluprednate ointment, and his dermatitis gradually improved. He visited our clinic for precise medical examination for finding out the causative agent of his dermatitis on May 13, 2003. We conducted 48 hours closed patch testing with the plaster he used and it’s ingredients. He reacted positively to the plaster containing ferbinac and crotamiton 5%p that was used for dissolute the active drug. He also reacted positively to a cream containing ketoprofen, however he reacted negatively to ketoprofen 1%p. Crotamoton was also used in the ketoprofen cream.

    Notes: We investigated a 4-year-old Japanese boy with oculocutaneous albinism who had a solitary pigmented mole measuring 5 mm in diameter on his back. An electron microscopic tyrosine incubation test and a DOPA reaction test clearly demonstrated the presence of tyrosinase activity in the patient's hypopigmented skin. The presence of tyrosinase activity was confirmed by tests on hair bulb samples. Histopathological evidence showed that the mole was a typical compound cellular naevus with melanin pigmentation. Although no reports to date have focused on the relationship between pigmented naevi in albinism and tyrosinase activity, our findings suggest that the occurrence of pigmented naevi in an albino may indicate the presence of tyrosinase activity.

    Notes: Summary The ultrastructural localization of the pemphigus vulgaris (PV) and pemphigus foliaceus (PF) antigens in cultured human squamous carcinoma cells was observed using immunogold electron microscopy. Both the PV and PF autoantibodies bound only to the extracellular portion of the desmosomal structures. After incubation at 37°C, the PV antigen–antibody complexes were observed within the cultured cells. PV and PF antigen expression was markedly reduced when the cells were cultured in medium with a low Ca2 concentration.

    Notes: The ultrastructural localization of Brazilian pemphigus foliaceus (BPF) (fogo selvagem) antigen(s) in cultured human squamous cell carcinoma cells was studied using immunogold electron microscopy. Five of six BPF sera, which showed positive cell-surface reactivity on immunofluorescence, bound to the cell-cell contact area of cytoplasmic projections. This binding pattern was apparently different from that of non-endemic pemphigus foliaceus and pemphigus vulgaris sera, and mouse monoclonal anti-buman E-cadherin antibody. The results suggest tbat BPF autoantibodies recognize a molecule(s) which is different from non-endemic pemphigus antigens, or different epitope(s) of a molecule identical with non-endemic pemphigus antigens, and that the epitope(s) to which BPF autoantibodies bind is expressed on cell-cell contact areas at a relatively early stage of cell-cell adhesion formation.

    Notes: Summary A 76-year-old man presented with a 3-month history of a cutaneous nodute on the right thigh. The tumour was composed of CD3+, large atypical cells, and CD20+, small normal-appearing cells. Flow cytometry showed that CD20+ cells outnumbered CD3+ cells. By Southern blot hybridization analyses, the malignant cells were shown to be of T-cell origin, because of the presence of rearranged bands for the β chain of the T-cell receptor, but not for the immunoglobulin heavy chain. This case represents a T-cell lymphoma intermingled with a large number of non-neoplastic B cells.

    Notes: Background  We observed nishikinezumi, cinnamon-coloured (NC)/Fujita (F) mice aged between 5 and 28 weeks. These NC mice have skin eruptions that resemble human atopic dermatitis (AD) under conventional circumstances.Object  We investigated the skin of eruptive and non-eruptive lesions in NC/F mice by using haematoxylin–eosin (H&E) staining, toluidine blue staining and immunohistopathological study with immunoglobulin (Ig)EɛRI, CD23, interleukin (IL)-4, IL-5, interferon (INF)-γ and Ia antigen.Results  Histological examination of the eruptive lesions revealed the perivascular infiltration of many lymphocytes and mast cells into the upper dermis. Intracellular oedema of the epidermis, lymphocyte infiltration into the epidermis and liquefaction degeneration of the basal layer were also observed. The numbers of IL-4 and IL-5 positive cells in the eruptive lesions were larger than those of the non-eruptive lesions. IL-4 and IL-5 positive cells in the eruptive lesions increased weekly. Some IFN-γ positive cells were observed in the eruptive lesions after 21 weeks. IFN-γ positive cells were scarce in the skin of both the non-eruptive and eruptive lesions before 21 weeks. Serum IgE increased from 7 weeks to 21 weeks.Discussion  We confirmed that these findings indicated that T helper (Th)2-dominant immunological activation transformed to a Th1-dominant situation. Many IgEɛRI positive cells were recognized in the dermis of the eruptive lesions by the time IgE had decreased. We assumed that the dermatitis before 21 weeks was an IgE-mediated allergy. We have previously reported that older NC/F mice had positive patch-test reactions to mites. Because serum IgE decreased after 21 weeks, dermatitis after 21 weeks might be associated more with cell-mediated delayed hypersensitivity than with IgE-mediated immediate allergy.

    Notes: Abstract A recent investigation has indicated that reactive oxygen species (ROS) may play an important role in the activation of lymphocytes. The main aim of the present study is to find out what effect on lymphocyte activation is attributable to radical scavengers (RSs). AD0261 (AD) is a novel RS which displays strong inhibitory action on the generation of lipid peroxides and superoxide anions. This compound has been shown to be effective in inhibiting tritiated thymidine ([3H]TdR) incorporation into, IL-2 release from and IL-2 receptor (Rc) expression on Con A-stimulated mouse spleen cells and PHA-P-stimulated lymphocytes of human peripheral blood, IC50 values being in the range 1–10 μM. It also inhibited erythema formation elicited by transferring PHA-P-stimulated lymphocytes into guinea pigs (30–40% inhibition at 10 μM,p〈0.05). Further study showed that the activity of ornithine decarboxylase (ODC), which is a critical enzyme for cell proliferation, was inhibited by AD in a dose-dependent manner (IC50=1.0 μM). In summary, AD would seem to affect the lymphocyte activation by inhibiting ODC activity, probably through the action of ROS trapping.

    Notes: Abstract Cyanobacteria are prokaryotes that carry out plant-type photosynthesis and contain several eukaryotic-type RNA-binding proteins. Using a single-stranded DNA column, a 33-kDa protein was isolated and characterized from Synechococcus sp. PCC6301. This protein of 293 amino acids is similar in overall structure to the ribosomal protein S1 found in the same species, and contains three repeated units that are highly similar to the S1 motif originally found in the ribosomal protein S1 of Escherichia coli. However, the 33-kDa protein was found not to be associated with ribosomes and its nucleic acid binding specificity is distinct from that of the ribosomal protein S1. As this protein has high affinity for both single- and double-stranded DNA, as well as for poly(G) and poly(A), we tentatively named it nucleic acid-binding protein 1 (Nbp1).
