    Notes: Summary Nuclear uptake and retention of3H-estradiol by luteinizing hormone (LH) and prolactin (PRL) cells was examined in three species of rodents (guinea pigs, hamsters and gerbils) using the combined techniques of immunocyto-chemistry and autoradiography. Castrated animals were injected with3H-estradiol and decapitated 1.5 h later. The pituitary glands were processed for thaw-mount autoradiography followed by conventional immunocytochemical staining for LH and PRL.3H-estradiol accumulated in more than 80% of the anterior pituitary cells in the gerbils, while only 33 and 22% of the cells accumulated3H-estradiol in the hamsters and guinea pigs, respectively. A varying percentage of immunoreactive LH and PRL cells in all three species were found also to contain binding sites for estradiol. Some LH and PRL cells in hamsters and guinea pigs and only some in PRL cells of gerbils were found to be devoid of grains. Quantitative analysis revealed that the number of grains per nucleus differed considerably from cell to cell. LH cells of guinea pigs accumulated much larger amounts of3H-estradiol than did the PRL cells, while the LH cells in the hamsters and gerbils accumulated only slightly more3H-estradiol than the PRL cells. These results confirm the previous observations in rats and baboons that demonstrated tremendous species differences in percentage of cells in the anterior pituitary gland that accumulated3H-estradiol. Also, these data suggest that there are functionally heterogeneous cell types among the LH and PRL cells in hamsters, guinea pigs and gerbils as has been previously demonstrated in rats and baboons.

    Notes: ---Comprehensive two-dimensional gas chromatography (GC×GC) is an utterly suitable separation technique for the analysis of complex samples, such as oil fractions. Once the two columns and the operating conditions are properly tuned, the technique is able to provide a detailed characterization of such materials. Some considerations applying to the tuning of a GC×GC system for a specific separation are presented and discussed. The authors present a number of different column sets and conditions which allow the separation of a non-aromatic hydrocarbon solvent, a kerosene, the light end of a crude oil, and an olefinic fraction, respectively. The highly structured GC×GC chromatograms, together with chemical knowledge about the samples, provide a much more comprehensive characterization of the samples than hitherto possible.

    Notes: Positive and negative thermospray mass spectra have been recorded for a number of phenazines at the submicrogram level. The spectra are dominated by molecular ion species ([M + H]+, [M - H]-) with minimal fragmentation. The gas chromatographic/electron impact mass spectrometric behaviour of the phenazines was also investigated. A number of phenazines were not amenable to gas chromatographic/mass spectrometric analysis without derivatization; in contrast, thermospray mass spectrometric data were obtained for each phenazine. Thermospray liquid chromatography/mass spectrometry has been applied to identify phenazine-1-carboxylic acid and 2-hydroxyphenazine in culture filtrates of Pseudomonas aureofaciens and 1-amidophenazine from Pseudomonas aeruginosa.

    Notes: Secondary ion intensity from glycerol is measured as a function of 5 keV primary ion current density and is found to be linear over the range 0.1-1 μA cm-2. The possibility that protonated glycerol, or some other condensed-phase precursor to secondary protonated glycerol, exists in solution and enhances secondary ion emission from glycerol is investigated. Kinetics for formation of such precursors by the primary ion beam are described and evaluated by pulsed primary ion beam experiments. Depletion rates are calculated assuming diffusion from the surface as the major loss mechanism Results of this analysis indicate that the mechanism for secondary emission of protonated glycerol does not involve formation of precursors, e.g. solution-phase protonated glycerol, directly or indirectly by the primary ion beam.

    Notes: Incident ion energy E1 and collision gas pressure have been adjusted so as to obtain a satisfactory tandem mass spectrum of the m/z 1882 ion formed from valme-gramicidin A (relative molecular mass 1881). Translational energy losses ΔE have been determined (at Ei = 14.8 keV) from the precise positions of a large number of fragment ion peaks in the spectrum. The variation in ΔE for different fragment ions is great and the magnitudes of ΔE are large (being commonly of the order of 102 eV). It is shown how very large energy losses ΔE can arise, if the parent ion decomposes to a small fragment ion which itself collides and decomposes further. Implications of the dependence of ΔE upon fragment ion mass for the scan laws of four-sector instruments are discussed briefly.

    Notes: Secondary ion mass spectra of singly substituted aromatic hydrocarbon/H2SO4 solutions showed intense aromatic molecular ion and protonated aromatic molecule peaks characteristic of dissolved aromatic compounds from a number of aromatic compound classes, including acids, aldehydes, ketones, nitriles and nitrogen heterocycles. The presence of simultaneously abundant peaks for molecular ions and protonated molecules in secondary ion mass spectra of each aromatic compound/sulfuric acid solution is consistent with known or expected gas-phase proton transfer chemistry. The ratio of intensities, M+·:[M + H]+, appears to be determined by sulfuric acid solution chemistry of the compound. Spectra obtained from 1-2 μl samples were relatively free from chemical noise and persisted for up to 20 min. Detection limits for some substituted aromatic compounds are estimated to be 10-12.
