    Notes: The morphology of the pineal gland was studied in protein-calorie-malnourished (PCM) rats. Twenty-day-old male Sprague-Dawley rats were placed in a 14:10 photoperiod and fed either an 8% low protein diet (LPD) or a standard laboratory diet (SLD) containing 27% protein for 30 d. At 50 d of age, rats from both animal groups were sacrificed at 0900 h and at 2400 h, and the pineal glands were immersion-fixed for either light or electron microscopic analysis. The cytoplasm and nuclei of the pinealocytes from the SLD-fed rats were consistently larger than those of the animals maintained on the LPD. Additionally, the lipid droplets were larger and more prominent in the controls at both 0900 h and 2400 h. Even though the size of these inclusions did not vary among animals given the same diet as a function of the time of sacrifice, they were more numerous in both the well-fed and malnourished rats during the dark phase of the photoperiod. In contrast neither diet nor sampling time affected the size or number of pinealocyte mitochondria. These morphological observations lend further support to the premise than PCM impairs the cellular activity of the pinealocytes.

    Notes: Summary The uptake and retention of a radiolabeled synthetic progestin ORG 2058 by the mammary glands were examined in the baboon. Four estrogen-primed baboons were injected intravenously with 2.5µg/kg body weight of3H-ORG 2058. One animal received an additional injection of 2.5 mg/kg body weight of unlabeled progesterone. One hour after the injections, the animals were killed and the mammary glands were removed and processed for autoradiography. Only 13.4% of the alveolar secretory cells studied demonstrated nuclear uptake and retention of the radiolabeled steroid, while 60% of the alveoli demonstrated at least 1 labeled cell per cross-section. The fibroblasts in the interlobular connective tissue and most of the cells lining the interlobular ducts were labeled; however, no cells in the intralobular connective tissue were labeled. These observations seem to indicate that all alveolar secretory cells do not contain progesterone receptors in the normal mammary gland.

    Notes: The interstitial tissue of the testis of the nine-banded armadillo is composed of blood vessels, clusters of Leydig cells, the usual connective tissue elements, and a network of lymphatic sinusoids. The endothelial walls of the sinusoids are separated from the peritubular contractile cell layer surrounding the seminiferous tubules by a thin layer of collagen. The peritubular contractile cell is characterized by filaments and dense bodies within the cytoplasm, whereas the endothelial cells lack these structures. Within each cluster, several Leydig cells surround one or more blood vessels. Adjacent Leydig cells are joined by 2- to 3-nm wide gap junctions and desmosome-like specializations. The Leydig cell is polygonal in shape with an ovoid nucleus. The cell is characterized by an abundance of smooth endoplasmic reticulum which appears as sheets of membranes, concentric whorls around vacuoles, and a random tubular network. Only a few short cisternae of rough endoplasmic reticulum are observed. Centrioles are closely associated with the Golgi apparatus. Rod-like mitochondria with tubular cristae are scattered throughout the cytoplasm. In addition, the cells contain vacuoles resulting from lipid extraction, filaments, microtubules, and glycogen. The surfaces of the cell exposed to the intercellular spaces exhibit numerous pinocytotic vesicles and cell processes which indicate active movement of material across the plasma membrane. In comparison to other mammalian species, the ultrastructural organization of the interstitium and the fine structure of the Leydig cell of the armadillo resemble those of the guinea pig.

Four estrogen-primed baboons were injected intravenously with 2.5 μ/kg body weight of 3H-ORG 2058. One animal, which served as a control, received an additional injection of 2.5 mg/kg body weight of unlabeled progesterone. One hour after the injections, the animals were killed and the uterus, cervix, oviduct, vagina, and labia were removed and processed for autoradiography. The cells in the germinative layers of the stratified squamous epithelium of the cervix, vagina, and labia demonstrated nuclear localization of the label. The columnar epithelium, both surface and glandular, of the uterus and cervix sequestered the synthetic steroid; however, the nuclei of the epithelium lining the oviduct were unlabeled. The nuclei of the fibroblasts and of the smooth muscle cells were labeled in all the organs studied. These preliminary observations suggest that there is a stage in the reproductive cycle in which progesterone receptors are contained in the stromal cells of the oviduct but are absent in the epithelium.

    Notes: Summary The uptake and retention of radiolabelled dihydrotestosterone by the pituitary gland was examined in the rhesus monkey. Two animals were given an intravenous injection of 1.0μg/kg 3H-dihydrotestosterone (DHT) alone while one monkey received both the labelled androgen and 100μg/kg of unlabelled steroid. One and a half hours later, they were sacrificed. The pituitary glands were removed and processed for autoradiography and immunocytochemistry. Autoradiographic localization of DHT was discernible in the partes nervosa, intermedia and distalis, albeit the highest concentration of radiolabelled cells was noted in the pars distalis. Immunocytochemical staining with antibodies to rat PRL, human TSHβ and ovine LHβ revealed a population of steroid-concentrating cells that contained TSH and a second group that contained LH. None of the cells that reacted with the anti-PRL serum were radiolabelled.

    Notes: Summary The localization of radiolabeled estradiol and dihydrotestosterone was examined in the lymphatic organs of both male and female baboons. A total of 12 baboons were divided into two groups, each containing three males and three females. Each animal in one group, both males and females, was injected intravenously with 1 μg/kg body weight of 3H-estradiol while those in the second group were each injected with 1 μg/kg body weight of 3H-dihydrotestosterone. As controls, one male and one female from each group also received a dose of 100 μg/kg body weight of the corresponding unlabeled steroid. One and a half hours after the injections, the animals were sacrificed and the spleen, thymus, and inguinal lymph nodes removed and processed for autoradiography. The localization of 3H-estradiol was similar in both males and females. In the thymus fibroblasts and epithelio-reticular cells, but not thymocytes, localized 3H-estradiol. In lymph node and spleen, nonlymphoid tissue concentrated the labeled estrogen. Additionally, in the paracortical region of the lymph node, an unknown cell type was labeled with estrogen. Only one male baboon demonstrated nuclear localization of 3H-dihydrotestosterone. This was observed in the reticular cells in the spleen and lymph nodes. The same cell type in the organs of the remaining animals was unlabeled.

    Notes: Summary Nuclear uptake and retention of3H-estradiol by luteinizing hormone (LH) and prolactin (PRL) cells was examined in three species of rodents (guinea pigs, hamsters and gerbils) using the combined techniques of immunocyto-chemistry and autoradiography. Castrated animals were injected with3H-estradiol and decapitated 1.5 h later. The pituitary glands were processed for thaw-mount autoradiography followed by conventional immunocytochemical staining for LH and PRL.3H-estradiol accumulated in more than 80% of the anterior pituitary cells in the gerbils, while only 33 and 22% of the cells accumulated3H-estradiol in the hamsters and guinea pigs, respectively. A varying percentage of immunoreactive LH and PRL cells in all three species were found also to contain binding sites for estradiol. Some LH and PRL cells in hamsters and guinea pigs and only some in PRL cells of gerbils were found to be devoid of grains. Quantitative analysis revealed that the number of grains per nucleus differed considerably from cell to cell. LH cells of guinea pigs accumulated much larger amounts of3H-estradiol than did the PRL cells, while the LH cells in the hamsters and gerbils accumulated only slightly more3H-estradiol than the PRL cells. These results confirm the previous observations in rats and baboons that demonstrated tremendous species differences in percentage of cells in the anterior pituitary gland that accumulated3H-estradiol. Also, these data suggest that there are functionally heterogeneous cell types among the LH and PRL cells in hamsters, guinea pigs and gerbils as has been previously demonstrated in rats and baboons.

    Notes: Summary The cycle of the seminiferous epithelium of the nine-banded armadillo can be divided into ten stages. As in most mammals, only one stage is observed per tubular cross-section. The process of spermiogenesis can be divided into thirteen steps according to the development of the acrosomal system and the flagellum. Four generations of spermatogonia are observed in the germinal epithelium: 1) stem cells, 2) type “A”, 3) intermediate, and 4) type “B” spermatogonia. The stem cell is characterized by a highly irregular nucleus and the presence of glycogen in its cytoplasm. The type “A” spermatogonium contains an oblong nucleus with one or two shallow infoldings of the nuclear membrane. The intermediate spermatogonium contains an ovoid nucleus characterized by one or two nuclei and heterochromatin scattered in the nucleoplasm. The nucleus of the type “B” spermatogonium is more spherically shaped with a centrally placed nucleolus and heterochromatin associated with the nuclear envelope.

    Notes: Summary The uptake and retention of radiolabeled estradiol by the uterus was examined in the armadillo. One pregnant and two non-pregnant armadillos were treated with 1.4 μg/kg body weight of 3H-estradiol (E2) by injection into the left ventricle, and one non-pregnant animal was injected with both the labeled hormone and 140 μg/kg body weight of unlabeled E2. One and a half hour after injection, the animals were sacrificed and the uteri were removed and processed for autoradiography. In the non-pregnant animals, nuclear localization was observed in the interstitial cells and glandular epithelium of the endometrium and the connective tissue cells and smooth muscle of the myometrium. Additionally, there was a gradation of uptake in the epithelial cells of the endometrium in that the glandular cells of the basal region were heavily labeled, while those cells in the sinusoidal, and luminal regions contained successively less label. The luminal cells were poorly labeled. In the pregnant female, the smooth muscle and glandular cells hypertrophied and their nuclei contained less label than was observed in the non-pregnant animals. The arteries of the myometrium were more easily distinguished in the pregnant animals and the nuclei of the endothelial cells and smooth muscle were more consistently labeled than those of the non-pregnant armadillos.
