    Notes: Summary The oral bioavailability of methotrexate is variable and may be dose-dependent. The absorption of ‘interval’ oral methotrexate, which is given between cycles of chemotherapy, is unknown. The bioavailability of oral methotrexate has been studied in eight patients, acting as their own controls, to assess the effect of subdivision of the dose, the formulation, and the timing of the methotrexate within the chemotherapy cycle. The mean bioavailability for all the oral methods of administration was 28.2%±3.7% compared with the same dose given IV. Absorption was uninfluenced by subdivision of the dose, liquid or tablet formulation, or administration on day 1 or day 10 of the chemotherapy cycle.

    Notes: Summary The administration of cytosine arabinoside (araC) by continuous IV infusion requires the patient to be in hospital and have prolonged IV cannulation. In this study the pharmacokinetics of araC during continuous IV infusion were compared with those of continuous SC infusion in six patients with acute myelogenous leukaemia. Each patient acted as his own control. The mean plasma levels of araC reached a plateau within 2 h and the plasma concentrations and the area under the curve were similar for both methods of administration. The mean area under the curve (AUC) was 1147±230 ng/ml for the IV infusion and 1017±238 ng/ml for the SC infusion. The plasma araC concentrations showed wide interpatient variation, and there was also considerable variability in the plasma concentrations of araC within the individual patients after a plateau had apparently been reached. Subcutaneous infusion was well tolerated by the patients without any local discomfort or excoriation and SC infusion of araC is thus a feasible alternative to IV infusion. It allows the patients the benefits of being at home, while avoiding unnecessary thrombophlebitis.

    Notes: Summary A range of clinical and industrial gloves have been evaluated to determine their ability to exclude penetration by cytotoxic drugs. Radiolabelled cyclophosphamide, methotrexate, daunomycin, cytosine arabinoside, and vincristine sulphate were studied in an equilibrium dialysis system in which the glove material was used as the dialysis membrane. The thicker gloves were the most effective, but very little drug crossed any of the gloves tested under laboratory conditions. Further studies to evaluate gloves in conditions of clinical use are now needed.

    Notes: Summary Experimental and clinical evidence indicates that bleomycin by continuous infusion is superior to intermittent administration. Continuous infusion is less convenient, however. It has been suggested that a suspension of bleomycin in sesame oil, given by IM injection, simulates a continous infusion. The pharmacokinetics of this formulation have been compared with those of bleomycin in saline following IM injection, in six patients. The pharmacokinetic profiles of the two formulations were similar. The only difference between the profiles was the long terminal half-life at very low concentrations between 12 and 48 h after injection of the oil suspension. This difference is of unknown, but doubtful, clinical significance.

    Notes: Abstract Experiments were conducted at 1 GPa on four starting materials to investigate the effects of variable mineral proportions on the melting systematics of compositionally fertile peridotitic assemblages. Starting materials were constructed by recombining Kilbourne Hole xenolith mineral separates by weight into four mixtures with mineral proportions olivine (Ol): orthopyroxene (Opx): clinopyroxene (Cpx): spinel (Sp) of 0.50:0.07:0.40:0.03 (FER-B), 0.50:0.46:0.01:0.03 (FER-C), 0.50:0.30:0.10:0.10 (FER-D), and 0.50:0.235:0.235: 0.03 (FER-E). Experiments were performed on a 1.27-cm (0.5 in.) piston-cylinder apparatus over the temperature interval 1270–1390 °C, using a variation of the diamond aggregate melt extraction technique employing vitreous carbon spheres in place of diamonds as the melt extraction layer. The solidus temperatures are similar for all the starting materials, with an average value of 1250 °C. In FER-D and -E, the near-solidus melting reaction for a lherzolite assemblage was determined to be of the form Cpx + Opx + Sp → melt + Ol. A subsequent reaction of the form Opx + Sp → melt + Ol was determined for FER-D after the exhaustion of Cpx. Over the entire temperature interval investigated for FER-B and -C, reactions were determined to be of the form Cpx + Sp → melt + Ol and Opx + Sp → melt + Ol, respectively. Melt percent (F) vs temperature (T) curves are concave up for all starting materials, demonstrating that isobaric melt productivity increases with progressive batch melting. At any given melt fraction, (dF/dT)P increases with increasing amount of Cpx in the starting material, indicating that the modal proportion of Cpx is one of the primary controls on isobaric melt productivity of upwelling peridotite. The concave up melt productivity functions for peridotitic assemblages determined in this study suggest that assuming linear or concave down melt productivity functions for modeling mantle melting may not be appropriate.
