    Notes: Background:  In situ hybridization techniques allow a cell-type-specific messenger RNA (mRNA) analysis in complex tissues such as human skin.Methods: To evaluate both the expression of mRNA and protein within the same tissue section, we developed a protocol of combined non-radioactive in situ hybridization and immunohistochemistry for use in dermatohistopathology. To validate the technique, we assessed the distribution of vasoactive intestinal polypeptide (VIP) protein and its receptor, VPAC2 mRNA, in human skin samples.Results: Simultaneous detection of VPAC2 mRNA and VIP immunoreactivity led to abundant staining for both signals in a variety of cell types. There was marked staining for VPAC2 mRNA in epidermal cells, with most pronounced hybridization signals found in keratinocytes of the basal layer and in glandular cells surrounded by VIP-immunoreactive nerve fibers. Hair follicle cells next to VIP-positive fibers also exhibited hybridization signals. Specific staining was also detected in endothelial and mononuclear cells. The findings of simultaneous in situ hybridization and immunohistochemistry were identical to results obtained by the single application of both methods.Conclusions:  The combination of in situ hybridization and immunohistochemistry appears to be a promising technique to assess the expression of both protein and mRNA in skin samples and may be used for various purposes in experimental and clinical dermatopathology.

    Notes: Abstract.  The physiological properties of transgenic tobacco plants (Nicotiana tabacum L.) with decreased or increased transport capacities of the chloroplast triose phosphate/phosphate translocator (TPT) were compared in order to investigate the extent to which the TPT controls metabolic fluxes in wild-type tobacco. For this purpose, tobacco lines with an antisense repression of the endogenous TPT (αTPT) and tobacco lines overexpressing the TPT gene isolated from the C4 plant Flaveria trinervia (FtTPT) were used. The F. trinervia TPT expressed in yeast cells exhibited transport characteristics identical to the TPT from C3 plants. Neither antisense TPT plants nor FtTPT overexpressors showed a phenotype when grown in a greenhouse in air. Contents of starch and soluble sugars in upper source leaves were similar in TPT underexpressors and FtTPT overexpressors compared to the wild type at the end of the photoperiod. The FtTPT overexpressors incorporated more 14CO2 in sucrose than the wild type, indicating that the TPT limits sucrose biosynthesis in the wild type. There were only small effects on labelling of amino acids and organic acids. The mobilisation of starch was enhanced in αTPT lines but decreased in FtTPT overexpressors compared to the wild type. Enzymes involved in starch mobilisation or utilisation, such as α-amylase or hexokinase were increased in αTPT plants and, in the case of amylases, decreased in FtTPT overexpressors. Moreover, α-amylase activity exhibited a pronounced diurnal variation in αTPT lines with a maximum activity after 8 h in the light. These changes in starch hydrolytic activities were confirmed by activity staining of native gels. Activities of glucan phosphorylases were unaffected by either a decrease or an increase in TPT activity. There were also effects of TPT activities on steady-state levels of phosphorylated intermediates as well as total amino acids and malate. In air, there was no or little effect of altered TPT transport activity on either rates of photosynthetic electron transport and/or CO2 assimilation. However, in elevated CO2 (1500 μl · l−1) and low O2 (2%) the rate of CO2 assimilation was decreased in the αTPT lines and was slightly higher in FtTPT lines. This shows that the TPT limits maximum rates of photosynthesis in the wild type.

    Description / Table of Contents: Abstract Peripheral opiod receptors have been found in inflamed synovia and the analgesic effect of intra-articularly administered morphine after arthroscopic knee surgery has been proven. There is controversy about effiacy of intraarticular morphine after shoulder arthroscopy. Thirtytwo patients with impingement syndrome underwent subacromial decompression in the course of arthroscopic shoulder surgery. At the end of the operation morphine (5 mg) or saline was injected periarticularly. Pain intensity (rest and passive mobilisation) was recorded after recovery and after 1, 2, 4, and 24 hours (Numeric Rating Scale); the use of rescue medication (piritramide by patient controlled analgesia) was noted. No relevant pain reduction was apparent in the morphine group. Piritramide consumption was identical in both groups (19,7±16 mg vs. 19,8±19 mg). We conclude that periarticularly administered morphine in arthroscopic subacromial decompression in the dosage applied in this study does exert no relevant analgesic effect. This is possibly due to the fact that either subacromial tissue, despite of chronic inflammation, does not show the same reagibility as synovia or it is a problem of the nearly complete resection of the subacromial bursa.
    Notes: Zusammenfassung Fragestellung. Die Existenz peripherer Opiatrezeptoren in entzündlicher Kniegelenkssynovia ist nachgewiesen und die analgetische Wirksamkeit intraartikulär applizierten Morphins nach Kniegelenksarthroskopien belegt. Ob ein solcher Therapieansatz auch für Schulterarthroskopien gilt, ist ungeklärt. Methodik. Bei 32 Patienten mit Impingementsyndrom wurde im Rahmen einer Schulterarthroskopie eine subakromiale Dekompression durchgeführt. Zum Operationsende wurden 5 mg Morphin oder NaCl 0,9% arthroskopisch in den Subakromialraum instilliert. Der postoperative Ruhe- und Belastungsschmerz wurde nach Erwachen sowie nach 1, 2, 4 und 24 h erhoben (Numeric Rating Scale, NRS) und der Bedarf einer Ausweichmedikation (Piritramid als Patientenkontrollierte Analgesie, PCA) erfaßt. Ergebnisse. Es zeigte sich keine klinisch relevante Schmerzreduktion in der Verumgruppe. Der Piritramidverbrauch über 24 h war in beiden Gruppen gleich (19,7±16 mg vs.19,8±19 mg). Schlußfolgerungen. Periartikulär verabreichtes Morphin bei Schulterarthroskopien erzeugt in der hier verwendeten Dosierung keine klinisch relevante analgetische Wirkung. Dies kann in der Operationstechnik begründet sein, aber auch in einer im Vergleich zur Synovia anderen Reagibilität des subakcromialen Bindegewebes.

    Notes: We report the zero kinetic energy photoelectron spectrum of the propargyl radical, C3H3. From the spectrum an ionization energy of 69 953±10 cm−1 (8.673 eV) is deduced. Vibrational frequencies are obtained for the totally symmetric normal modes of the propargyl cation, as well as some combination and overtone bands. Both the frequencies and the relative intensities agree well with the predictions from recent ab initio calculations. © 2000 American Institute of Physics.

    Notes: Wide angle x-ray scattering and classical molecular-dynamics simulations were employed to study structural and dynamic properties of two low-molecular weight glass-forming liquids, propylene carbonate, and salol. We observe pronounced changes in the liquid structure as a function of temperature in a wide temperature range bracketing the glass transition temperature. The experimental and simulation results compare the x-ray scattering from the liquid slightly and far above the experimental glass transition temperature, Tgexp. The simulations largely reproduce the characteristic behavior of the experimental x-ray intensities, which is interpreted in terms of clusterlike heterogeneities forming in the liquid as Tgexp is approached. © 2000 American Institute of Physics.
