    Notes: Use of a monoclonal antibody against Lafora bodies for the immunocytochemical study of ground-glass inclusions in hepatocytes due to cyanamide Aims: Ground-glass inclusions (GGIs) in hepatocytes are known to be associated with cyanamide treatment in patients with alcohol dependency. The purpose of this study was to assess the reactivity of a monoclonal antibody (MAb) raised against polyglucosan and to detect early events in GGI formation. Methods and results: Formalin-fixed paraffin-embedded liver tissues from four patients treated with cyanamide were used. Sections were stained with haematoxylin and eosin and periodic acid–Schiff with and without diastase digestion, and were immunohistochemically stained with the MAb. For electron microscopic study, routinely processed liver tissue from one patient was examined with conventional and immunoelectron microscopy with use of the MAb. All specimens from the four cyanamide-treated patients contained GGIs in the cytoplasm of hepatocytes, and these GGIs reacted intensely with the MAb. Fully developed GGIs contained various organelles, whereas early ones consisted primarily of glycogen granules and dilated smooth endoplasmic reticulum. In immunoelectron microscopic preparations, gold particles were located within GGIs, and the immunolabelled organelles appeared to be glycogen granules. Conclusions: This novel MAb is useful for the detection of GGIs caused by cyanamide. Our results support the idea that GGI formation may result from specific abnormalities in glucose metabolism.

    Notes: Expression of MUC1 and MUC2 and carbohydrate antigen Tn change during malignant transformation of biliary papillomatosis Aim: Biliary papillomatosis is characterized by papillary proliferations of biliary lining cells without invasion or metastasis. The neoplastic character and biological behaviour of this disease remain still speculative. These issues were examined in this study. Methods and results: Mucin core protein MUC1, MUC2, MUC3, MUC5AC and carbohydrate antigens (T, Tn and sialosyl Tn) were immunohistochemically examined, using 11 lesions of biliary papillomatosis from seven patients, and five lesions of biliary papillomatosis with foci of carcinoma from four patients. Five cases of papillary intrahepatic cholangiocarcinoma and 12 histologically normal livers were used as a control. Patients with biliary papillomatosis alone or with carcinoma were middle-aged or elderly (five men and six women). Microscopically, biliary papillomatosis showed a villous, papillo-tubular, papillary, or papillo-villous pattern with a thin fibrovascular core. Cytologically, they were classifiable into biliary epithelial or pyloric gland-like type. The former was frequent in the cases associated with carcinoma. Expression of MUC1, Tn antigen and sialosyl Tn antigen was frequent and marked in biliary papillomatosis alone and with carcinoma and also intrahepatic papillary carcinoma. In addition, marked expression of MUC1 and Tn antigen were rather frequent in biliary papillomatosis with carcinoma and intrahepatic biliary papillary carcinoma compared with biliary papillomatosis. MUC2 was rather frequent and marked in biliary papillomatosis alone compared to other two disease groups. Focal expression of MUC5AC and MUC2 was rather frequent and infrequent irrespective of disease group, respectively. Focal expression of T antigen was frequent in papillary ICC. Conclusion: Biliary papillomatosis could undergo overt malignant transformation along with altered phenotypic expression of MUC proteins and mucin carbohydrate antigens.

    Notes: Six packings of symmetry-related cylinders, with cylinder axes in invariant positions (coordinates completely determined by symmetry), are described. Two have axes along 〈100〉 and four have axes along 〈111〉. It is shown that there can be no cubic cylinder packing with axes along 〈110〉. Earlier errors concerning the numbers of such packings and their symmetries are corrected.

    Notes: Abstract. The feasibility of three-dimensional (3-D) photonic crystals made using textile technology was investigated. Three different textures consisting of the cotton-yarn and TiO2 dispersed resin; a crossed linear-yarn laminated fabric, a multi layered woven fabric, and a 3-D woven fabric, were fabricated. The microwave attenuation of the transmission amplitude through these photonic crystals was measured. The straight cotton-yarn as well as the wavy cotton-yarn/TiO2 dispersed resin photonic crystals exhibited band gaps in the 6 to 15 GHz range. Thus, we could fabricate successfully 3-D photonic crystals using textile technology.

    Notes: Abstract Pancreaticobiliary maljunction (PBM), a congenital anomaly, causes regurgitation of pancreatic juice into the biliary tract, where it exerts a hazardous influence. However, changes in the common bile duct (CBD) remain obscure due to a lack of suitable experimental models. Using cats, we have developed an experimental model of PBM without bile-duct dilatation that allows the pure effects of PBM to be studied. Histologic and cellular kinetic changes in the CBD were analyzed in 6 controls and 9 experimental animals that survived for more than 6 months. CBD sections were stained with hematoxylin and eosin and a monoclonal antibody to the proliferating cell nuclear antigen (PCNA). Invaginations of the bile-duct epithelium or parietal sacculi increased, and peribiliary glands were well-developed. PCNA-positive cells significantly increased in the CBD, especially in the parietal sacculi and glands. It is concluded that PBM increases the cell cycle in CBD epithelium and subsequently developed peribiliary glands. These developed glands may be associated with the formation of protein plugs, often seen in patients with PBM.

    Notes: Abstract We have produced a new model of pulmonary super-infection with Aspergillus fumigatus and Pseudomonas aeruginosa in immunosuppressed mice. Male ICR mice were given an intratracheal inoculation of 4 × 105 conidia of A. fumigatus in agar beads, and were immunosuppressed with 100 mg/kg subcutaneous injections of cortisone acetate on days 7, 9, 12, 14, and 16 after inoculation. Twelve days after inoculation, with the agar beads, the mice were challenged with the intranasal instillation of 2 × 106 CFU of P. aeruginosa. The survival rates of superinfected, A. fumigatus-alone, P. aeruginosa-alone, and non-infected mice were 50%, 30%, 90%, and 100% 14 days after pseudomonal infection (26 days after inoculation of A. fumigatus), respectively. In the superinfected mice, both A. fumigatus and P. aeruginosa were detected more than 10 days after pseudomonal infection (22 days after inoculation of A. fumigatus). Histopathological examination revealed peribronchial necrosis around A. fumigatus hyphae and inflammation by P. aeruginosa. This infection model in mice would be useful for studying the pathogenesis of superinfection.

    Notes: Abstract We administered multidisciplinary treatment to a 37-year-old man with poorly differentiated adenocarcinoma of the rectum which exhibited a fistula between the rectum and the urinary bladder; the tumor was considered unresectable because of extensive local spread at initial evaluation, including a laparotomy. The patient received radiotherapy, consisting of a total dose of 50 Gy, given as fractions of 2 Gy per day 4 days a week. Concurrently with the radiation, the patient also received twice daily a tegafur [1-(2-tetrahydrofuryl)-5-fluorouracil] suppository (750 mg) and 5-fluorouracil ointment (2.5 g) applied to the perianal involved skin lesion. In addition, 5 KE of OK-432 was injected into the tumor nine times during the course of radiation. Because the tumor responded well to this radio-chemo-immunotherapy, the patient was discharged, and was treated only with tegafur suppositories for almost 3 years on an outpatient basis (total dose of tegafur, approximately 1500 g). As a result of the combined therapy, the tumor completely disappeared, while only the recto-urinary fistula remained. The patient has survived without recurrence for 50 months after his initial presentation, and his levels of carcinoembryonic antigen and immunosuppressive acidic protein have decreased from pretreatment values of 85 ng/ml and 1220 μg/ml, respectively, to 0.8 ng/ml and 459 μg/ml, respectively. Although this is only one case, it shows that appropriate multidisciplinary treatment may achieve excellent results even in patients with unresectable poorly differentiated adenocarcinomas of the rectum.
