Notes: In Channa punctatus, day-night variations in hypothalamic 5-HT (serotonin) and monoamine oxidase (MAO) activity were noticed in preparatory but not other phases (prespawning and postspawning) of the annual reproductive cycle. Hypothalamic MAO, 5-HT, and norepinephrine (NE) activity was found to be high in the prespawning phase and low in the postspawning phase. Dopamine (DA) activity, on the other hand, was high in the postspawning season and low in the prespawning phase. Pinealectomy caused season-dependent effects on hypothalamic monoaminergic activity, with a significant increase in serotonergic activity and a significant reduction in MAO activity at midscotophase during the preparatory phase (March) but not in the prespawning (May-June) or postspawning (September) phases. Hypothalamic catecholaminergic (CA) activity was not influenced by pinealectomy during any of the seasons. To determine whether or not the photoperiodic influences on daily variations of 5-HT and MAO in the preparatory phase are mediated via pineal and/or lateral eyes, fish were pinealectomized and/or blinded in January, when there is no rhythm, and sacrificed in February, when a day-night variation normally sets in. The day-night difference in 5-HT content and activity and MAO activity was not abolished by pinealectomy or blinding alone; but the combination (pinealectomy + blinding) obliterated the daily variation only in 5-HT content and in MAO activity. However, pinealectomy and blinding, alone or in combination, caused a significant elevation of 5-HT activity (not its level) and a significant decrease in MAO activity at midscotophase, with the combination having an additive effect. Hypothalamic CA content or activity was not affected by these regimes. The results show that photoperiodic influence on the daily pattern of 5-HT and MAO activity is mediated through and by the interaction of the pineal and lateral eyes.

Notes: Summary We report a new high-resolution absorption spectrum of BaD in the wavelength region (3580÷3760) Å using the third order of a 21 ft. concave grating spectrograph. Two new electronic states of BaD are reported named asK 2∃-X 2 ∃ at 3750 Å andL 2 II-X 2 ∃ at 3680 Å, respectively. The (0,0),(1,1),(1,0) and (2,1) bands of theL 2-II-X 2∃ state and (0,0) and (1,0) bands of theK 2∃-X 2∃ state have been observed. Vibrational and rotational analysis of these newly observed bands have been carried out and the corresponding effective vibrational and rotational constants are reported. TheK 2∃ andL 2 II states are tentatively assigned as originating from the 7p atomic state of barium.

Notes: Abstract Heart tissue contains large amounts of the protein encoded by the Ca2+ pump gene SERCA2. The SERCA2 RNA can be spliced alternatively to produce mRNA encoding the proteins SERCA2a and SERCA2b which differ in their C-terminal sequences. In this study we report the tissue distribution of SERCA2a and SERCA2b isoforms byin situ hybridization to rabbit heart and stomach. The expression of SERCA2 mRNA was high in myocardial cells, being the highest in the atrial region. In contrast, there was more SERCA2 protein in Western blots in ventricles than in atria. Myocardial cells expressed predominantly the mRNA for the isoform SERCA2a. Whereas the stomach smooth muscle and the neuronal plexus expressed SERCA2 at levels much lower than myocardial cells, the expression was very high in the stomach mucosa. Mucosa contained mainly the mRNA for SERCA2b. From immunocytochemistry it was concluded that the anti-heart SR Ca2+ pump antibody IID8 reacted much better with heart and surface mucosal cells in the stomach than with the stomach smooth muscle, and that IID8 reactivity was intracellular. In contrast PM4A2B, an antibody against the plasma membrane Ca2+ pump, reacted well with heart and stomach smooth muscle, plexus and mucosa, and its localization appeared to be in the plasma membrane. Thus, stomach smooth muscle expressed SERCA2b mRNA and protein at low levels, mucosa expressed SERCA2b mRNA and protein at high levels, atria and ventricle expressed SERCA2a mRNA and protein at high levels, mRNA being more in atria, but protein being more in ventricles.

Notes: Abstract Although the role of adenosine deaminase (ADA), adenylate deaminase (AMP-DA), purine nucleoside phosphorylase (PNP) is well documented in gastric and intestinal carcinoma, their role in inflammatory bowel diseases remains unknown. In the present study, we investigated the profile of these enzymes in blood and intestinal tissues during colitis. Colitis induced in Wistar rats by acetic acid was monitored by a marker enzyme myeloperoxidase (MPO). The tissue levels of MPO increased on 1, 2, 5 and 6 days post-administration (PA) of acetic acid and declined to the control levels by day 7 PA. In parallel the blood levels of ADA and AMP-DA decreased on days 1, 2 and 5 without any significant change on days 6 and 7 PA. Similar observations were recorded for these enzymes in the cytosolic extracts of colonic tissue specimens. In contrast, PNP remained unaltered in both blood and tissue samples. These findings suggest an inverse-relationship between inflammation and purine deaminases in both blood and tissues.

Notes: Abstract Four commercial alloys-Hastelloy C-4, alloy 1.4306S (SS 304L), Incoloy 800H, and Incoloy 825-were studied for their oxidation behavior at elevated temperatures. Specimens were exposed to air from 600 to 1200°C for 1 to 400 hr. Reaction kinetics of oxidation were determined, and the morphology of the surface-oxide scales was investigated. Hastelloy C-4 showed better resistance to oxidation dor exposure temperatures up to 1000°C in comparison with the other three alloys. In this temperature range, it follows a cubic rate law of oxidation due to formation of uniform, protective, and adherent oxide scales. The latter three alloys obeyed the parabolic rate law at 1000°C and 1200°C, but for lower temperatures a mixed behavior was shown. The oxide layer developed on the alloy 1.4306S was always in the form of stratified nodules/warts. For longer exposures the nodules joined each other to form continuous but discrete layers. Incoloy 800H and Incoloy 825 behaved in an almost identical manner, their reaction kinetics being governed by the parabolic rate law throughout the temperature range. Oxide spalling was observed at all temperatures. In contrast to Incoloy 800H the Incology 825 was totally oxidized for longer exposures at 1200°C.

Notes: Abstract In continuation of previous work, the oxidation behavior of three more alloys, namely UHBA 25L, Sanicro 28, and Inconel 690, was studied at high temperatures (600 to 1200°C). The oxidation kinetics of UHBA 25L and Sanicro 28 followed the parabolic-rate law at 800 and 1000°C. At 600°C oxidation rates were very low, while at 1200° C the parabolic-rate law was initially observed, followed by “breakaway.” Both alloys suffered extensive spalling at all temperatures, despite their high Cr contents of about 25% and 27%, respectively. This is atrributed to significant amounts of Mn present. Sanicro 28, with higher Cr, suffered an early breakaway at 1200°C because of the presence of Mo, which forms a low-melting oxide. Inconel 690 showed mixed behavior at 600 and 800°C, parabolic at 1000°C and cubic at 1200°C. Inconel 690 and Incoloy 800H gave the best overall performance for the range of temperature and exposure time under study. This may be ascribed to the absence of Mo, the presence of small amounts of Ti and Al, and relatively small amount of Mn in these alloys.

Notes: Abstract A gene encoding a sulphur-rich, sunflower seed albumin (23% cysteine plus methionine) was modified to contain the promoter for the 35S RNA of cauliflower mosaic virus, in order to obtain leaf expression in transgenic plants. In addition, a sequence encoding an endoplasmic reticulum-retention signal was added to the 3′ end of the coding region so as to stabilize the protein by diverting it away from the vacuole. The modified gene was introduced into subterranean clover (T. subterraneum L.) and its expression was detected by northern and western blots and by immunogold localization. The albumin was accumulated in the lumen of the endoplasmic reticulum, and, among six independent, transformed lines, it accumulated in the leaves of T0 transgenic plants at varying levels up to 0.3% of the total extractable protein. The level of accumulation of the sunflower albumin increased with increasing leaf age, and in the older leaves of the most highly expressing plants of the T1 generation it reached 1.3% of total extractable protein. Expression of the SSA gene was stable in the first and second generation progeny. These results indicate that there is potential for significantly improving the nutritional value of subterranean clover for ruminant animals such as sheep by expressing genes that code for sulphur-rich, rumen-stable proteins in leaves.

Notes: Abstract In Channa punctatus maintained under ambient photothermal conditions (10.5L:13.5D; 13 ± 2°C) in the postspawning season, hypothalamic dopamine (DA) and norepinephrine (NE) content varied diurnally, but serotonin (5-HT) and monoamine oxidase (MAO) did not. Exposure of the fish to high temperature (25 ± 2°C), with or without altering the photopheriod, increased the NE and DA content, and NE turnover but decreased DA turnover. Increasing the photoperiod (16L:8D) alone had no significant effect on catecholaminergic (CA) content or activity. When the fish were exposed to long photoperiod, with or without altering the temperature, a day-night variation in 5-HT content and activity was induced in the postspawning season. An increase in temperature alone had no significant effect on 5-HT content or activity. Hypothalamic MAO activity was elevated in fish exposed to high temperature alone, or in combination with long photoperiod, but was not affected significantly by long photoperiod treatment. These results indicate that CA activity is influenced largely by ambient high temperature, whereas serotonergic activity is controlled primarily by photoperiod, with high temperature having an additive effect. MAO activity seems to be influenced by both temperature and photoperiod in this species.
