    Notes: The effect of heat treatment during fish meal processing on amino acid racemization was studied. The hydrolysis-induced racemization rate (R) and the d-isomer content in the sample before hydrolysis (I) were differentiated by means of deuterium labelling and gas chromatography-mass spectrometry (GC-MS) analysis in selected ion monitoring mode. A preliminary experiment on laboratory-made herring meals cooked at 125°C for different time intervals showed aspartic acid (Asp) as the only amino acid with significant racemization before hydrolysis. Aspartic acid racemization rate appeared to be a nearly linear function of the duration of thermal treatment (R2= 0.93; P 〈 0.01). Analyses were carried out on several samples of commercial fish meals from different origin. Low-temperature-dried fish meals had a d-Asp content, expressed as I= 100 [d-isomer concentration before hydrolysis / (d- +l-isomer concentration before hydrolysis)], at less than 1%, while the d-isomer content of high-temperature dried fish meals exceeded 2%. Differences between the two commercial categories were statistically significant (P 〈 0.001). Further studies are required in order to evaluate the effects of d-Asp in protein of fish feeds and the role of the raw material and processing parameters in inducing amino acid racemization in protein of fish meals.

    Notes: Abstract Blood glucose, insulin, C-peptide and glucagon were evaluated in 36 newborn term infants at birth, and before and 60 min after the first feed during the first day of life. Under basal conditions glycaemia diminished during the first day of life and glucagon increased, while insulin and C-peptide did not show any variation. The C-peptide: insulin molar ratio was higher in the newborn than in adults because of the longer half-life of C-peptide, probably due to reduced renal function in the neonatal period. The subjects were divided into two groups: 18 newborn infants were given a feed of commercially available milk powder reconstituted in water at 10% (5 ml/kg); the other 18 were given a feed of 5 ml/kg 10% glucose solution. In each group 6 were given the first feed after a fast of 6 h, 6 after a fast of 12 h, and 6 after a fast of 24 h from birth. After the first feed with milk, the average increase of glycaemia was 19.83 mg%, of insulin 6.06 μU/ml, and of C-peptide 1.88 ng/ml. After the first feed with glucose the average increase of glycaemia was 13.59 mg%, of insulin 2.46 μU/ml, and of C-peptide 0.59 ng/ml. Glucagon did not show significant changes after the first feed.

    Notes: Abstract In six infants aged between 5 and 8 months with vitamin D deficient rickets, we have studied blood levels of calcium (Ca), phosphorus (P), alkaline phosphatase, immunoreactive parathyroid hormone (PTH) and calcitonin (CT), as well as urinary excretion of Ca, P, hydroxyproline and cyclic AMP, both under basal conditions and during a 4 h infusion of 20 mg/kg 10% Ca gluconate in normal saline. Under basal conditions all the infants had high alkaline phosphatase (range: 470–770 U.I/l); PTH (range: 620–1200 pg Eq/ml) and CT (range: 440–750 pg/ml) but two infants had hypocalcaemia and four had normocalcaemia and hypophosphataemia. The urinary Ca excretion was low whereas the urinary P, hydroxyproline and cyclic AMP excretions were high. During Ca infusion the total serum Ca and CT levels increased, while alkaline phosphatase and PTH fell. After the end of the infusion, CT levels fell perceptibly; phosphaturia, hydroxyprolinuria and cyclic AMP decreased on the day of the infusion.

    Notes: Summary Bacillus subtilis strain PB 2427 temperature sensitive in the synthesis of RNA during spore germenation and outgrowth has been characterized to some extent. At non permissive temperature (46°C) strain PB 2427 synthesizes stable and unstable RNA for 50 min from the beginning of germination and then stops. Most of the stable RNA is degraded to shorter molecules but can be identified as ribosomal RNA by hybridization-competition experiments. At non permissive temperature, in the presence of chloramphenicol, synthesis of RNA proceeds, though at a reduced rate, for at least 90 min. By hybridization-competition experiments it can also be shown that the RNA synthesized at 46° C in the presence of chloramphenicol includes transcripts that are absent, from the RNA synthesized at 46° C in the absence of drug. The RNA polymerase (holo and core) purified from vegetative cells of the mutant strain does not appear to have a greater heat-lability as compared with the enzyme purified from the parental strain. At non permissive temperature only six polypeptide chains with MW ranging from 47,000 to 78,000 daltons are synthesized by the germinating spores of the mutant.

    Notes: Abstract The performance and clinical relevance of a qualitative PCR-based assay for the detection of HIV-1 DNA sequences in peripheral blood mononuclear cells (PBMCs) was evaluated by two different laboratories. Four hundred and one samples were obtained from 397 individuals from different risk populations. All blood donors tested had negative results; positive signals were obtained from all infected patients. HIV-1 DNA was detected in 3 of 17 infants born to seropositive mothers; Western blot indeterminate blood donors and exposed healthcare workers had negative results. Our results demonstrate that this PCR assay provides both sensitive and specific results and is suitable for testing large numbers of samples and for rapid identification of HIV-1 infection.
