    Notes: Abstract  The temperature-sensitive mutant cells (tsFT101) derived from a mouse mammary carcinoma cell line, FM3A, become multinucleated at a non-permissive temperature of 39°C. To further understand the molecular mechanism of such cytokinetic disturbance, we examined the expression of profilin, the main regulator of the transition of globular actin (G-actin) to filamentous actin (F-actin). RT-PCR analysis of mouse profilin cDNA from tsFT101 showed a point mutation (177 A → G) which was a wobble mutation causing no change in the encoded amino acid. The expression level of profilin mRNA was, however, diminished in cultured tsFT101 cells under non-permissive temperatures compared with wild-type FM3A cells in association with multinucleation. A stable transfection of profilin cDNA expression vector to tsFT101 cells prevented multinuclear cell formation when cultured at 39°C. In contrast, antisense profilin cDNA expression vector did not alter multinuclear cell formation. The primary cause of the cytokinetic disturbance of tsFT101 cells may be due to the diminished level of profilin gene expression.

    Notes: Abstract  Cells of a temperature-sensitive mutant line (tsFT101) derived from a mouse mammary carcinoma cell line (FM3A) become multinucleated at a non-permissive temperature of 39°C because of disturbed cytokinesis. To explore how this relates to thermotolerance, we examined the proliferative activity of, and heat shock protein (HSP) expression in, FM3A and tsFT101 cells cultured at 37°C and 39°C after heat shock pretreatment (15 min exposure at 45°C). FM3A cells developed thermotolerance when cultured at both 37°C and 39°C, but whereas tsFT101 cells developed thermotolerance at 37°C, this was markedly reduced at 39°C. Western blot analysis showed similar degrees of expression of constitutive HSP70 (HSP73) in FM3A and tsFT101 cells after heat shock pretreatment at both 37°C and 39°C. However, expression of inducible HSP70 (HSP72) was reduced in tsFT101 cells at 39°C compared to 37°C and to FM3A cells at both 37°C and 39°C. Heat shock pretreatment activated DNA binding of heat shock transcription factor (HSF) in FM3A cells at 37°C and 39°C, but only at 37°C in tsFT101 cells. These results indicate that (1) multinucleation caused by disturbed cytokinesis increases temperature sensitivity, (2) HSP70 is critical for the development of thermotolerance in both FM3A and tsFT101 cells, and (3) decreased expression of inducible HSP70 parallels deficient development of thermotolerance in tsFT101 cells cultured at a non-permissive temperature.

    Notes: Abstract  Ultraviolet B (UVB) alters the expression of heat shock protein 70 (HSP70) in cultured fibroblast cells derived from human skin. However, the nature of the signal transduction pathway remains to be determined. Transforming growth factor-β (TGF-β) has a large variety of biological functions, including cell growth control, modulation of inflammation and immunoregulation. In this study, we examined whether TGF-β is associated with the process of HSP70 expression induced by UVB irradiation. The constitutive expression of TGF-β1 mRNA and HSP70 expression in human skin fibroblast cells were detected using reverse transcription polymerase chain reaction (RT-PCR) and Western blot analysis. The results indicate that: (1) UVB irradiation stimulates HSP70 expression in a dose- and time-dependent manner, (2) constitutive expression of TGF-β1 mRNA is detected after UVB irradiation, the level of which peaks at 4 h after 10 mJ cm–2 of UVB irradiation, (3) HSP70 expression is induced by TGF-β1 without UVB irradiation, and (4) HSP70 expression induction with UVB irradiation is inhibited by preincubation of the cells with the anti-TGF-β type II receptor antibody. Our results suggest that HSP70 expression induced by UVB involves the autocrine signalling of TGF-β production.

    Notes: Abstract  Epithelial mucins are present at the apical membranes of gastrointestinal epithelial cells or in their secretions. In this study, we examined the occurrence of peptide epitopes of the mucins MUC1 and MUC3 and of three mucin-associated glycotopes (TF, Tn, and s-Tn) in a series of colorectal tissue samples (normal colon, adenomas with different grades of dysplasia, carcinoma in situ, and invasive carcinomas). A new monoclonal antibody to a conformation-dependent peptide epitope of MUC1 was employed, which does not react with the fully glycosylated mucin as found in normal gastrointestinal mucosa. We found that adenomas acquired the ability to expose Tn, s-Tn, TF and MUC1 epitopes, and this correlated with increasing malignant potential. The secretory mucin, MUC3, revealed a different pattern: it was detectable in all sections, with maximum expression in adenomas and decrease in carcinomas. Most importantly, normal mucosa and benign lesions showed supranuclear and/or apical distribution of these antigens, but malignant lesions and lesions with a very high risk of malignancy revealed diffuse cytoplasmic and basolateral membrane localization. The immunohistological response to a combination of MUC1-related antibodies may assist in assessing the malignant potential and status of lesions of the colon.

    Notes: Abstract Advances in the synthesis of organic conducting polymer systems has increased the electrical conductivity of these systems by several orders of magnitude in the last decade. Several practical applications are envisioned for such systems, but a thorough understanding of the conduction mechanisms and identification of the charge carriers is lacking, making design and implementation for bulk synthesis difficult. In order to clarify our understanding of the electrical properties of these systems, the resistivity and magnetoresistivity of various polymers doped near the metal - insulator transition, such as polyaniline protonated by camphor sulfonic acid (PANi-CSA) and polypyrrole doped with PF6 (PPy-PF6), have been studied down to 25 mK in magnetic fields up to 16 T.

    Notes: Abstract There has been increased research interest in systems composed of multiple autonomous mobile robots exhibiting cooperative behavior. Groups of mobile robots are constructed, with an aim to studying such issues as group architecture, resource conflict, origin of cooperation, learning, and geometric problems. As yet, few applications of cooperative robotics have been reported, and supporting theory is still in its formative stages. In this paper, we give a critical survey of existing works and discuss open problems in this field, emphasizing the various theoretical issues that arise in the study of cooperative robotics. We describe the intellectual heritages that have guided early research, as well as possible additions to the set of existing motivations.

    Notes: Abstract Hg-1223 thick films on flexible Ni substrate coated with Cr(Ag,Pd) have been fabricated by a two-step spray/press process. The effect of possible interfacial diffusion between Hg-1223/Ni has also been examined. By properly adjusting the processing parameters, a self-field Jc ∼ 7×104 A/cm2 at 77 K has been achieved inthese Hg-1223 thick-film tapes, comparable to that of the well-known BSCCO tapes prepared by the more complex powder-in-tube technique.

    Notes: Abstract We have observed a δ-independent linear pressure effect on Tc of HgBa2CuO4+δ under hydrostatic pressures up to 1.7 GPa for δ≤0.30 but a nonlinear effect for δ〉0.30. These results, together with previous doping data, show a possible non-rigidity of the electron bands of the compounds, based on which previous phenomenological models of the pressure effect on Tc are built, and thus suggest only a restricted universality for the inverse parabolic relation between Tc and charge carriers per CuO2-layer. In other words, factors in addition to charge carriers can affect high temperature superconductivity.

    Notes: Abstract The magnetic penetration depth λ(T) of three HgBa2CuO4+δ samples with 0.16 〈 δ ≤ 0.27 has been determined from the reversible magnetization. The obtained λ follows a BCS-like correlation of 1/λ2 ∝ 1−(T/Tc)2 over whole measured temperature range in an underdoped sample with Tc ∼ 90 K, but deviates significantly from similar fits in an overdoped sample with the same Tc and an optimum doped sample, whose 1/λ2 's depends on T nearly linearly below Tc/2. This asymmetry between the underdoped and overdoped samples suggests that the T-dependence of 1/λ2 is affected by doping in a complicated way.

    Notes: Abstract Root exudates were collected from radish and rape plants grown in P sufficient and P deficient nutrient solution. In radish, tartaric, malic and succinic acids were the dominant organic acids which increased between 15 times (succinic acid) and 60 times (malic acid) under P deficient conditions. In another experiment in quartz sand culture supplied with either Ca3(PO4)2 or AlPO4, radish utilized P from AlPO4 much better than from Ca3(PO4)2 whereas the opposite was true for rape. The results demonstrated the role of a particular organic acid in mobilizing sparingly soluble P and were in accordance with the preferential growth of two plants on acid (radish) and calcareous (rape) soils in China.
