    Notes: Summary Otsuka Long-Evans Tokushima Fatty (OLETF) rats are reported to be obese Type II (non-insulin-dependent) diabetic rats with insulin resistance and impaired insulin secretion. To investigate the contribution of intestinal glucose absorption to postprandial hyperglycaemia, we determined the plasma xylose concentrations after an 0.8 g/kg oral xylose load which was used as a test of small intestinal glucose absorption in 6-week-old OLETF rats and weight-matched Long-Evans Tokushima Otsuka (LETO) rats. An oral glucose tolerance test showed that OLETF rats developed hyperglycaemia at 60 and 90 min after the glucose load, though the fasting plasma glucose concentration, insulin concentration and insulin-induced in vivo glucose utilization rate were similar. Consistently, in an oral D-xylose loading test, the peak concentration of plasma xylose in OLETF rats was increased by 58.7 % compared with that of LETO rats (p 〈 0.005). The disappearance rate of plasma xylose concentrations after intravenous xylose loading did not differ between the two strains. Co-treatment with 0.4 g/kg phlorizin, a specific inhibitor of sodium-dependent glucose transporter 1 (SGLT1), abolished both plasma glucose and xylose concentrations after the loads. Morphological studies showed that both the small intestinal wet weight and surface area were 30 % larger in the OLETF rats than in the LETO rats. Furthermore, the SGLT1 mRNA content of OLETF rats also increased compared with LETO rats. These results suggest that an increased SGLT1 expression concomitant with intestinal hypertrophy in OLETF rats is partly associated with postprandial hyperglycaemia before the onset of insulin resistance and hyperinsulinaemia.     Notes: Abstract Inorganic phosphate (Pi) uptake by the marine cyanophyte Synechococcus sp. NIBB 1071 was studied using cells grown in an artificial seawater medium. The phosphate uptake was markedly enhanced in cells grown in the medium of low phosphate concentrations (phosphate-limited cells) than in cells grown in the phosphate-rich medium (phosphate-replete cells). The diagnosis of kinetics of instantaneous phosphate-uptake showed that V max of the former was more than two orders of magnitude greater than that of the latter, and the k m of the former was about 1/20 of that of the latter. The enhancement of the phosphate uptake was completed after a 40-h incubation of phosphate-replete cells in the phosphate-free medium. The activation was suppressed by chloramphenicol, an inhibitor of protein synthesis. The uptake developed in phosphate-limited cells was energy dependent and susceptive to osmotic shock, which suggests the involvement of a periplasmic phosphate-binding protein, analogous to that found in heterotrophic gram-negative eubacterial cells. The relationship between phosphate quota and growth rate, together with the kinetical data for phosphate uptake, predicted that ambient phosphate as low as 0.5 nM could support cell growth at a rate of one division per day. Results indicate that cells can grow rapidly even at phosphate concentrations as low as nanomolar levels. A possible regulatory mechanism of phosphate uptake in marine Synechococcus spp. is discussed in relation to a wide distribution of this picophytoplankton in the ocean environment.

    Notes: Abstract TheBacillus subtilis gnt operon is negatively regulated via interaction of thegnt repressor (GntR) with an operator upstream ofgntR, which is antagonized by gluconate. An 8 bp insertional operator mutation (gntOi) of thegnt operon was constructed which affected the expression level of this operon. Two suppressors of thisgntOi mutation, exhibiting normal expression, were also isolated; one involved a threonine substitution for the Ala-48 residue (gntR48T) within the helix-turn-helix DNA-binding motif of GntR, and the other an adenine substitution for the guanine at nucleotide — 4 within thegntOi operator (gntOiM4A) (+1 is the transcription initiation site). ThegntR48T mutation by itself rendered thegnt operon partially constitutive. When thegntR43L mutation, which renders thegnt operon fully constitutive, was introduced into thegntOi orgntOiM4A mutant, the operator mutations were found not to affect the promoter activity of thegnt operon. These in vivo results indicate that thegntOi mutation affects the operator interaction with GntR, causing a low expression level even in the presence of gluconate. In vitro gel retardation and DNase I footprint analyses demonstrated that even when gluconate was present, GntR still bound to thegntOi operator region.

    Notes: Summary Intracellular location of cytochrome oxidase (cytoxidase) active in vivo was studied cytochemically with four strains of cyanophytes, using 3,3′-diaminobenzidine (DAB) oxidation. DAB was oxidized in the dark bySynechocystis sp. PCC6714 and two strains ofAnacystis nidulans (Tx20 and R2) grown under photoautotrophic, heterotrophic and salt-stressed conditions, respectively. Electron microscopic observations showed that DAB-oxidation in the dark occurred in the thylakoids, but was insignificant on or around the cytoplasmic membrane. However, deposition of DAB-oxidation product around the cytoplasmic membrane was observed with cells of the thylakoid-less cyanophyteGlaeobacter violaceus ATCC29082. All DAB oxidations observed with the four strains were inhibited completely by cyanide, the inhibitor of cyt-oxidase, but not by 3-amino-1,2,4-triazole, the inhibitor of peroxidase. The results show that (1) DAB was oxidized by the cyt-oxidase functioning in the respiratory system, and that (2) cyt-oxidase in thylakoids was active in vivo.

    Notes: Abstract Short-lived ions12B (beta-radioactive, T1/2=20.3 ms) sustaining nuclear spin polarization were introduced into superfluid helium at 1.7 K. It was found that the12B ions were transported as charged entities under a static electric field and that the nuclear polarization was maintained throughout the lifetime of12B nuclei. Polarization of12B was determined through beta-NMR method. “Snowball”, a singly charged microcluster of helium atoms formed around an impurity ion, is responsible for the behaviour and thus constitutes a suitable environment for preserving nuclear polarization of the core ions12B. In a separate experiment snowballs were produced by implanting8Li (T1/2=830 ms) into liquid helium and detected by means of alpha particles from the core ions to guarantee that the snowballs survive longer than the lifetime of12B.

    Notes: Abstract: The β decay of 61Ga to its mirror nucleus 61Zn has been measured for the first time by using on-line mass separation and β-delayed gamma-ray spectroscopy. The observed decay strength to the ground state implies superallowed character in accordance with the systematics of the mirror decays in the sd and fp shell. The β feedings observed to four excited states in 61Zn are consistent with earlier spin-parity assignments based on in-beam experiments. The ground-state spin and parity for 61Ga were determined to be 3/2−.
