    Notes: Abstract Oxidation of 2-propanol with hydrogen peroxide catalyzed by TS-1 was strongly retarded in the presence of an equimolar 1-propanol to a level of 1-propanol oxidation, and the competitive oxidation behavior was applied for the selective oxidation of 1-methoxy-2-propanol to afford 1-methoxy-2-propanone in a high yield of 85%.

    Notes: Abstract To characterize the morphological features of the myelinated fibers in the mutant quails deficient in neurofilaments (NF), caused by a nonsense mutation in the NF-L gene, the morphological parameters of the axon and myelin sheath, and their relationships in the peroneal nerve were evaluated. In the mutant, the axonal area was smaller than in the control (P〉0.01), reflecting the lack of large diameter axons. There was no significant difference in the mean number of myelin lamellae and of their spacings between controls and mutants. Therefore, it was decided to analyze the alteration of axonal parameters in relation to the number of myelin lamellae. In the regression analysis, the number of microtubules (MT) per square micrometer of the axonal area was greater in the mutant than in the control (P〈0.05); however, the number of MT per axon was similar in controls and mutants with the same given number of myelin lamellae. The number of MT+NF per axon was smaller in the mutant than in the control only for myelinated fibers with more than 25 myelin lamellae (P〉0.05). These findings indicate that there was a less significant effect of NF deficiency on the smaller than on the larger myelinated fibers. There was no compensatory increase in the numbers of MT per axon of the myelinated fibers in the mutant as found previously in the unmyelinated fibers of the mutant.

    Notes: Abstract Recombinant human α1-antitrypsin (rAAT) was expressed and secreted from transgenic rice cell suspension cultures in its biologically active form. This was accomplished by transforming rice callus tissues with an expression vector, p3D-AAT, containing the cDNA for mature human AAT protein. Regulated expression and secretion of rAAT from this vector was achieved using the promoter, signal peptide, and terminator from a rice α-amylase gene Amy3D. The Amy3D gene of rice is tightly controlled by simple sugars such as sucrose. It was possible, therefore, to induce the expression of the rAAT by removing sucrose from the cultured media or by allowing the rice suspension cells to deplete sucrose catabolically. Although transgenic rice cell produced a heterogeneous population of the rAAT molecules, they had the same N-terminal amino acids as those found in serum-derived (native) AAT from humans. This result indicates that the rice signal peptidase recognizes and cleaves the novel sequence between the Amy3D signal peptide and the first amino acid of the mature human AAT. The highest molecular weight band seen on Western blots (AAT top band) was found to have the correct C-terminal amino acid sequence and normal elastase binding activity. Staining with biotin-concanavalin A and avidin horseradish peroxidase confirmed the glycosylation of the rAAT, albeit to a lesser extent than that observed with native AAT. The rAAT, purified by immunoaffinity chromatography, had the same association rate constant for porcine pancreatic elastase as the native AAT. Thermostability studies revealed that the rAAT and native AAT decayed at the same rate, suggesting that the rAAT is correctly folded. The productivity of rice suspension cells expressing rAAT was 4.6–5.7 mg/g dry cell. Taken together, these results support the use of rice cell culture as a promising new expression system for production of biologically active recombinant proteins.

    Notes: Abstract  A new algorithm of Delaunay Tessellation Particle Tracking Velocimetry (DT-PTV in abbreviation) is proposed for tracking particles in images of a PIV system by making use of the Delaunay tessellation (DT). The algorithm is tested by using numerically simulated particle images. The calculation results based on DT are compared with those obtained by a conventional algorithm of Binary Image Cross-correlation method (BICC). The new algorithm shows higher performance of obtaining more identical particles in two consecutive images correctly with shorter computation time even if the images contain many particles. A further application of DT to elimination of spurious vectors is also discussed.
