Notizen: Abstract Central mixed venous oxygen saturation (S $$\mathop v\limits^ - $$ O2) monitoring in critically ill patients to estimate adequacy of peripheral perfusion is gaining increasing popularity. However, a number of unexpected responses, one of which is marked depression of regional (splanchnic) venous oxygen saturation which may coexist with normal or high S $$\mathop v\limits^ - $$ O2, makes interpretation pretation of this parameter difficult. The S $$\mathop v\limits^ - $$ O2 and hepatic venous oxygen saturation levels in seven injured (postoperative) and 15 septic patients were measured. No substantial differences between central and hepatic venous oxygen saturation were noted in nonseptic patients, however, septic subjects exhibited a normal S $$\mathop v\limits^ - $$ O2 of 70.5%±8.7% at a time when the hepatic venous saturation was 55.6%±14.4% which is a significant (p〈0.05) reduction. This reduced oxygen saturation was noted to arise from an increased regional metabolic rate rather than reduced perfusion. Nevertheless, we conclude that a flow limited regional oxygen consumption may potentially exist despite the presence of a normal S $$\mathop v\limits^ - $$ O2 in certain patient subgroups such as septic subjects. Therefore, a normal S $$\mathop v\limits^ - $$ O2 should not be considered as sole criteria to insure optimal oxygen delivery in critically ill patients.

Notizen: Abstract The identification of female carriers of Fabry's disease is important for genetic counselling since prenatal diagnosis of affected fetuses is possible. The activities of either total α-galactosidase or α-galactosidase A in cultured fibroblasts were similar in Fabry carriers and controls and cannot therefore be used for carrier detection. Better discrimination between carriers and controls was found when total α-galactosidase activity was expressed as a ratio to β-galactosidase activity, but overlap still occurred. However, there was complete discrimination between the ratio of α-galactosidase A to β-galactosidase in cultured fibroblasts from five carriers of Fabry's disease and either 11 controls, seven hemizygote affected males or two of their female relatives.

Schlagwort(e): Labelled neutrophil kinetics ; Septic shock ; Baboon model

    Thema: Medizin
Notizen: Abstract The purpose of this study was the correlation of neutrophil kinetics with the pathogenic course of septic shock in the baboon model. Radioactively labelled neutrophils were traced in vivo in normal baboons (n= 6) and inEscherichia coli-infected baboons, which were reinjected with labelled autologous neutrophils either 2 h after the onset of theE. coli infusion (procedure A) (n= 3) or simultaneously with the infusion (procedure B) (n=3). Cell isolation was done according to a method developed in this laboratory. The cells were labelled with tropolonate In 111. resuspended in 1–2 ml plasma and reinjected. One-minute images were taken every 5th min and then hourly for 4 h with a gamma camera and analysed with a data processor. Time-activity curves were obtained for neutrophil washout from the lungs and neutrophil accumulation in the liver and spleen. These curves were compared for normal baboons and for those treated according to procedures A and B. A significant retention of neutrophils in the lungs of baboons withE. coli-induced septic shock was noted as well as an abnormally slow rate of accumulation in the liver and spleen. It also seems that any lung injury which could be attributed to changes in neutrophil behaviour should be traced back to events during the early exposure of neutrophils to bacterial infection.

Notizen: Summary This paper reviews biological properties of glycoprotein E (gE) of pseudorabies virus (Aujeszky's disease virus) and homologous proteins in otheralphaherpesvirinae. It focuses on the gene encoding gE, conserved regions in the gE protein and its homologs, the complex of gE and gI, biological functions of gE in vitro and in vivo, the role of gE in latency and the role of gE in the induction of humoral and cellular immune responses. Special emphasis is placed on the use of gE as a marker protein in the control and eradication of pseudorabies virus.
