    Notes: The potential use of RAPD markers for taxonomic studies in Malus was investigated using 18 accessions of wild species and 27 apple cultivars. 29 preselected random decamer primers were applied to three sets of Malus genotypes. Random amplified polymorphic DNA (RAPD) ‘fingerprints’ were analysed for polymorphic amplification fragments, and coefficients estimating genetic similarity were calculated on the basis of about 50 polymorphic RAPD loci in each set of genotypes. Cluster analysis by an unweighted pair-group method with arithmetic averages (UPGMA) revealed that, in the cultivars, the molecular classification was in good agreement with the known lineage. A dendrogram generated for the wild species gave relationships that were, in principle, in accordance with the known phylogenetic information. Closely related species from section I were clearly distinguishable from those of sections III and IV. On the molecular level, a high degree of genetic diversity was found among both different apple cultivars and wild species of the genus Malus. The results gave additional evidence for the hypothesis that M. pumila and M. sylvestris were involved in the origin of the cultivated apples.

    Notes: Abstract The α-aminoadipate-semialdehyde dehydrogenase (EC 1.2.1.31) of Trichosporon adeninovorans, an enzyme of lysine biosynthesis, was partially purified, some properties of the enzyme were studied and a novel regulatory pattern was found. The Km values of the enzyme were estimated to be 0.78 mM for α-aminoadipate, 1.0 mM for ATP, 0.23 mM for NADPH and 0.77 mM for MgCl2. It is demonstrated that the enzyme can be regulated by lysine and lysine analogues. l-Lysine (Ki of 0.09 mM), S-(B-aminoethyl)-l-cysteine (Ki of 0.007 mM and δ-hydroxylysine (Ki of 1.65 mM) inhibited the enzyme activity. The inhibition was competitive with respect to α-aminodipate and non-competitive with respect to both ATP and NADPH.

    Description / Table of Contents: Summary The long-term variation of the mean δ13C-value of breath-CO2 of Europeans (base value −26‰) was ±1‰ over a period of several months. However, the ingestion of products from corn, a C4-plant, caused remarkably higher shifts. In a fasted state and during rest a stability of the base value of ±0.3‰ could be attained for several hours. Longer fasting periods imply a shift of the CO2-δ-value due to the metabolism of body products. — By a change of the main energy source to corn products the δ13C-base line could be shifted to −18‰ within one day. After this shift even the metabolism of products from C3-plants could be investigated. The degradation curve and the integral degradation of fats were largely dependent on the method of application, and the base value largely interfered with the results. The period of maximum disintegration for a naturally and uniformly labelled fat was different from that of an artificially labelled trilinoleate. The results shall be used for the adjustment and the correction of the base line in13C-breath tests, especially with patients under parenteral nutrition.
    Notes: Zusammenfassung Der mittlere δ13C-Wert im CO2 der Atemluft von Europäern (Basiswert −26‰) zeigt eine Langzeitschwankung über mehrere Monate von ±1,0‰; der Verzehr von Produkten aus C4-Pflanzen (z. B. Mais) hat allerdings erheblich höhere Änderungen zur Folge. Im Nüchternzustand und in Ruhe wird während 6 h eine Stabilität des Basiswertes von ±0,3‰ erreicht; nach längeren Nüchternphasen kann aber der Abbau körpereigener Substanzen zu Verschiebungen führen. Durch Umstellung der Energieversorgung auf Maisprodukte ist es möglich, den Basiswert innerhalb eines Tages auf −18‰ zu heben; dann läßt sich auch der Abbau von Substanzen aus C3-Pflanzen verfolgen. Der Verlauf und die Rate des Abbaues von markierten Fetten sind stark von der Art der Applikation abhängig, und die Messung des Gesamtumsatzes wird durch den Untergrund stark beeinträchtigt. Der Zeitpunkt des maximalen Abbaues von uniform markiertem Fett (Sojaöl) unterschied sich von dem eines in der Carboxylgruppe markierten Trilinoleats. Die Resultate sind Grundlage zur Einstellung bzw. Korrektur von Basiswerten bei13C-Atemtests, besonders bei Patienten mit parenteraler Ernährung.

    Notes: Abstract The ultrastructural differentiation of capsule and its relation to tube development is described in several Octocorallia species(Alcyonaria: Alcyonium digitatum, Parerythropodium coralloides, Cornularia cornucopiae, Paralcyonium elegans; Pennatularia: Pteroeides spinosum, Veretillum cynomorium; Gorgonaria: Pseudopterogorgia aerosa), all of which have only one type nematocyst. In the Octocorallia, capsule and tube are secreted successively by the Golgi apparatus associated with a primary centriolar complex. During the secretion of the external tube, the outer capsular wall (sclera) is structurally differentiated; inside the capsule the material of the inner capsular wall is separated from the later capsular content (matrix). The primary wall differentiation enables the capsules to “grow” after capsular secretion has been completed. Following tube secretion, the external tube is completely transferred into the capsule, without the tube wall being transformed into capsular wall, as previously suggested (Westfall, 1966; Ivester, 1977). During early invagination of the tube wall, the coarse, granulated matrix of the external tube is transferred into the internal tube. From this material the spines are developed, which are observed before the tube is completely transferred into the capsule. By a secondary wall differentiation the previously structureless inner capsular wall changes to a complex structure, extending again the capsule, thus mixing the capsular content and enabling the tube to shift to a position, which corresponds with that of mature capsules. These observations demonstrate for the first time the differentiation of the capsule and its close relationship to the differentiation of the tube in nematocysts of Octocorallia.
