    Notes: Abstract Connexin45 is a gap junction protein which forms channels with unique characteristics. RNA blots demonstrated that connexin45 is expressed in a number of cell lines including WB, SK Hepl, BHK, A7r5, CLEM, and BWEM cells. Connexin45 was further studied in BWEM cells using specific affinity-purified antibodies directed against a synthetic peptide representing amino acids 285–298 of its sequence. Immunofluorescence experiments demonstrated that the BWEM cells expressed both connexin43 and connexin45 and that these connexins colocalized. Connexin45 polypeptide, immunoprecipitated from BWEM cells metabolically labeled with [35S]-methionine, consisted of a predominant 48 kD polypeptide. Connexin45 and connexin43 contained radioactive phosphate when immunoprecipitated from BWEM cells metabolically labeled with [32P]-orthophosphoric acid. This phosphate label was removed from connexin45 by alkaline phosphatase digestion. Treatment of BWEM cells with the tumor promoting agent 12-O-tetradecanoylphorbol-13-acetate (TPA) inhibited intercellular passage of microinjected Lucifer yellow. While TPA treatment induced phosphorylation of connexin43 in these cells, it reduced the expression of connexin45. Furthermore, the connexin45 expressed after TPA treatment was not phosphorylated. These results suggest that treatments which alter protein phosphorylation may regulate connexin43 and connexin45 in BWEM cells by different mechanisms.

    Notes: Es wird eine neue Synthese des Cyanmelamins aus 2-Chlor-4, 6-diamino-1,3,5-triazin und Cyanamid in Dimethylformamid beschrieben. Cyanmelamin reagiert als schwache Säure mit Metallen, Aminen usw. unter Salzbildung. Die Umsetzung mit HCl-Gas führt in der Kälte zum Chloramidiniumchlorid, in Gegenwart von Alkohol zum Iminoester-hydrochlorid. Bei der sauren Hydrolyse von Cyanmelamin entsteht Ammelin, bei der alkalischen Cyanammelin, das als schwache Säure ebenfalls Salze bildet.Die aus unseren Arbeiten gewonnenen Erkenntnisse der Melamin-Bildung aus Dicyandiamid werden in einem Reaktionsschema zusammengefaßt, in dem das Cyanmelamin eine Position zwischen Guanylmelamin und Melamin einnimmt.

    Notes: Die Umsetzung von 2,4-Dinitro-phenyl-pyridiniumchlorid und 2,4-Dinitro-phenyl-isochinoliniumchlorid mit heterocyclischen Hydrazinen führt zu den entsprechenden Hydrazonen des 1-[2,4-Dinitro-anilino]-pentadien-(1,3)-als-(5) bzw. β-[2,4-Dinitro-anilino]-o-styroaldehyds. Durch Erhitzen der Hydrazone in Eisessig bzw. alkohol. Salzsäure erfolgt Ringschluß zu Pyridinium- bzw. Isochinolinium-betainen, die zur 1,3-dipolaren Addition befähigt sind und mit geeigenten Dipolaphilen zu Pyrazolo [1,5-a]pyridinen bzw. Pyrazolo-[5,1-a]isochinolinen cyclisieren.

    Notes: Die Addition verschiedener elektrophiler Agenzien an ω-Methoxyolefine wurde untersucht, um Aufschluß über eine Methoxygruppenbeteiligung zu erhalten. Eine derartige Beteiligung konnte bei der Addition von Chlor und Brom an 5-Methoxypenten-(1) nachgewiesen werden; hier entstehen zu 10-15% die entsprechenden 2-Halogenmethyltetrahydrofurane, und zu 50-60% tritt Umlagerung unter Verschiebung der Methoxygruppe ein. Die Addition von Chlor oder Brom an 6-Methoxyhexen-(1) liefert zu 15-20% die entsprechenden 2-Halogenmethyltetrahydropyrane, eine Methoxylgruppenwanderung ist nicht nachweisbar. Die Reaktion von Chlor oder Brom mit 4-Methoxybuten-(1) und mit 7-Methoxyhepten-(1) liefert ausschließlich 1, 2-Additionsprodukte.Bei der Anlagerung von Chlor an Allyl-methyl-äther erhält man außer 73% 2,3-Dichlor-1-methoxypropan infolge nebenher ablaufender Dreikomponentenreaktionen 12% 1,2,3-Trichlorpropan, 7% 1,3-Dimethoxy-2-chlorpropan und 8% 1, 2-Dimethoxy-3-chlorpropan. Bei der Addition von 2,4-Dinitrophenylsulfenylchlorid und von Quecksilberacetat an ω-Methoxyolefine tritt weder Umlagerung noch Ringschluß ein. Mit 2, 4-Dinitrophenylsulfenylchlorid entstehen nebeneinander Markownikow- und anti-Markownikow-Produkte, der Anteil des Markownikow-Produktes steigt mit wachsender Kettenlänge vom Allyl-methyl-äther bis zum 7-Methoxyhepten-(1).

    Notes: Die bevorzugte massenspektroskopische Reaktion der 1,2,4-Triazol-4-N-oxide ist die Abspaltung von Sauerstoff (M+-16) aus dem Molekülion. Diese Spaltung wird nur wenig von Methyl- und Benzylgruppen in 3- oder 5-Stellung des s-Triazolringes beeinflußt. Im geringen Umfang spaltet sich aus den N-Oxiden auch Stickstoff (M+-28) ab. Die durch Sauerstoffsabspaltung gebildeten Radikalionen sind mit den Molekülionen der entsprechend substituierten 1,2,4-Triazole identisch und fragmentieren bevorzugt zu Nitrilen und Diaziridinradikalionen. Zwischen photochemischen und massenspektroskopischen Reaktionen der 1,2,4-Triazol-4-N-oxide besteht nur ein bedingter Zusammenhang.

Unter Anwendung der von Schuberth in(2) dargelegten Vorausberechnung isothermer Phasengleichgewichtsdaten für spezielle ternäre Systeme vom Typ S1/S2/L0 auf das in Rede stehende System kann eine Überprüfung der Meßdaten auf thermodynamische Konsistenz vorgenommen werden. Wie sich zeigt, ist die Übereinstimmung von Messung und Vorausberechnung durchaus zufriedenstellend.

    Notes: Gap junctions which comprise a family of proteins called connexins have been implicated in the morphogenesis of the chick limb bud. We have examined the expression patterns of two members of the connexin family, connexin43 (Cx43) and connexin42 (Cx42), during the early development of the chick limb bud and embryo by in situ hybridization. Cx43 mRNA is expressed in high amounts in the apical ectodermal ridge (AER), which promotes the outgrowth of the mesodermal cells of the limb bud, and in the ectopic AER of the limb buds of polydactylous diplopodia-5 mutant embryos. In contrast, little Cx43 expression is detectable in nonridge limb ectoderm at early stages of limb development. These results suggest that Cx43 gap junctions may integrate the activity of the cells comprising the AER and compartmentalize them into a functionally distinct entity capable of directing limb outgrowth. In addition, Cx43 exhibits high expression in the posterior subridge mesoderm of the early limb bud that is growing out in response to the AER, but little expression in the anterior mesoderm. This graded distribution of Cx43 transcripts correlates with a functional gradient of gap junctional communication along the anteroposterior (AP) axis, and suggests that Cx43 gap junctions may be involved in pattern formation across the AP axis. At later stages of development, Cx43 is transiently expressed in high amounts in the precartilage condensations of the carpals and metacarpals, at a time when critical cell-cell interactions are occurring that trigger cartilage differentiation. In contrast, in the developing limb, Cx42 is expressed exclusively by the central artery. In the remainder of the chick embryo, Cx42 is expressed in high amounts by the vessels comprising the arterial vasculature, but is not expressed by the venous vasculature. Thus, Cx42 gap junctions may be involved in specification of the arterial vasculature of the limb and embryo. Cx42, but not Cx43, is expressed in the ventricle of the heart, and by cells along the intrasclerotomal fissure that separates the rostral and caudal halves of the sclerotome of somites into distinct communication compartments. © 1994 Wiley-Liss, Inc.

    Notes: Synthesis, Coordination Chemistry and Mass Spectrometric Fragmentation of New N-(Thiocarbamoyl)-benzamidinesSyntheses of N-(thiocarbamoyl)benzamidines from benzimidoylchlorides and aromatic amino acids as starting materials are reported. The prepared compounds were used as ligands for complexing nickel(II) and copper(II)ions. The benzamidines were characterized by mass spectrometric methods. The fragmentation pattern of the benzamidines was derived from the corresponding MIKE-spectra.
