    Notes: Abstract The distributions of cathepsin B (CB) a lysosomal cysteine proteinase, type IV collagen (CIV) and laminin (LM), which are main components of basement membranes (BMs) were studied in a series of 64 human lung adenocarcinomas using an immunohistochemical technique. Over-expression of CB (〉80% positive cells) was significantly associated with the grade of tumour differentiation (p〈0.01), with lymph node metastasis (p〈0.01) and with BM degradation (p〈0.01) detected by the staining pattern of CIV and LM. It was significantly associated with a prognostic disadvantage (p〈0.01). The immunohistochemical staining pattern of CB has a close relationship with degradation of BM, and may be used as a marker for tumour metastasis and prognosis in lung adenocarcinoma.

    Notes: Abstract Antiinflammatory effects of indomethacin farnesil (IMF), a novel prodrug of indomethacin, was examined after both oral and local administration. In the air pouch carrageenin-induced inflammation, an oral dose of IMF exerted dose dependent inhibitory effects on the accumulation of inflammatory exudate fluid and the migration of leukocytes into the exudate. Both the increased vascular permeability and the prostaglandin E2 levels in the exudate fluid were reduced by IMF. Significant levels of free indomethacin were detected in the pouch fluid. In spite of the inability of IMF to inhibit prostaglandin synthesis in a cell free cyclooxygenase system, IMF injected locally inhibited carrageenin paw edema, and the inhibitory effect was comparable to that of indomethacin itself. When injected locally into the paw together with carrageenin,14C-IMF was effectively converted to its active metabolite, indomethacin. The indomethacin concentration in the paw tissue was comparable to that of indomethacin injected paws with the same molar dose of free indomethacin.

    Notes: Abstract Cine-MRI with presaturation bolus tracking was used in patients with syringomyelia associated with a Chiari malformation to study pulsatile movement of the hindbrain, cervical spinal cord, cerebrospinal fluid and the fluid within the syrinx. Nine patients had 13 examinations, 6 preoperative, 3 after syringosubarachnoid shunting and 4 after posterior fossa decompression. Five controls were also examined. Dynamic display of the acquired images demonstrated downward displacement of the presaturation bolus on the cerebellar tonsils and medulla oblongata (or upper cervical cord) at the C1 level in all preoperative examinations and in two patients after syringo-subarachnoid shunting but with residual foramen magnum obstruction. Downward displacement of the bolus on the cervical spinal cord was also demonstrated in 7 examintions, but not observed in the controls. Thus, the hind-brain-spinal cord axis showed larger pulsatile movements in patients with foramen magnum obstruction. Based on these observations and a review of the literature, a new theory on the mode of extension of syringomyelia, emphasising the role of increased pulsatile movement of the hind-brain-spinal cord axis is proposed: that the pulsatile movements, together with a one-way valve mechanism in the syrinx cavity act as a “vacuum-pump” to enlarge the syrinx.

    Notes: Abstract We produced three monoclonal antibodies, SG1, SG2 and SG3, specific for human seminal γ-glutamyltransferase when characterized by enzyme-linked immunosorbent assay and immunoblotting. Seminal γ-glutamyltransferase was localized, by immunostaining, to the epithelial cells of the ductus epididymidis, seminal vesicle and prostate gland with SG1, those of the prostate gland with SG2, and those of the seminal vesicle with SG3. Rabbit polyclonal anti-seminal γ-glutamyltransferase serum reacted with the proximal convolution of the kidney and the bile capillaries of the liver, and with the epithelial cells of the reproductive organs. However, immunoreactivity was not observed in the kidney or liver with the monoclonal antibodies. Thus, these monoclonal antibodies are probably all specific to seminal γ-glutamyltransferase but recognize different epitopes.

    Notes: Summary When corn endosperm is gently homogenized in a microfilament-stabilizing buffer and filtrates centrifuged at very low g forces, abundant protein bodies are present in the pellet. When stained with both rhodamine-phalloidin and fluorescein-S1-myosin and viewed under epifluorescence, individual protein bodies appear to be enclosed in F-actin and groups of protein bodies appear to be enmeshed within a network of fine actin filaments. The yield of actinenclosed protein bodies is not affected by the addition of non-ionic detergents to the grinding medium, but it is severely decreased with the addition of cytoskeleton-disrupting agents including Tris-HCl, KCl, KI, and ammonium sulphate, and the interconnecting fine filaments are totally abolished. Heparin, which interacts with membrane-cytoskeleton complexes, causes apparent coagulation of the protein bodies. The evidence from fluorescent microscopy suggesting that F-actin surrounds and enmeshes the protein bodies is supported by electron microscopy, gel electrophoresis, and Western blot analysis.

    Notes: Summary Frozen corn endosperm was homogenized in a cytoskeleton-stabilizing buffer and stained directly (without pelleting) with rhodamine-phalloidin for actin and either thiazole orange to stain RNA or DiOC6 to stain membranes prior to examination under the fluorescence microscope. Other samples were treated with a non-ionic detergent alone or in conjunction with a ionic detergent prior to staining and fluorescence microscopy. Very gentle homogenization in unsupplemented buffer yielded a massive aggregate containing protein bodies that fluoresced after treatment with the ER stain DiOC6. This aggregate was capped by an aggregate of unstained starch grains. More vigorous homogenization yielded more disperse patterns showing almost identical co-localization of ER, actin and RNA (polysomes). Homogenization in buffer plus non-ionic detergent removed most of the membrane yet maintained co-localization of actin and polysomes, while homogenization in double detergent removed the last traces of membrane and actin, and released over 70% of the polysomes. We interpret these results to suggest that protein bodies are surrounded by membranes, cytoskeleton and RNA (polysomes) and that the majority of the polysomes are attached more firmly to the cytoskeleton than to the membrane. This provides evidence from fluorescence microscopy to supplement that from biochemical analyses for the existence of cytomatrix-bound polysomes in plants.

    Notes: Abstract Forest community dynamics were studied for 4 years in a 6 ha permanent plot of species rich, old-growth, temperate deciduous forest in Ogawa Forest Reserve, central Japan. The gap formation rate, recruitment, mortality, gain and loss rate in basal area during 4 years were 42 m2 ha−1 yr−1, 1.74% yr−1, 1.19% yr−1, 1.12% yr−1 and 0.88% yr−1, respectively. The turnover time calculated from them ranged from 58 to 240 years. Both the mortality and mortality factors were size dependent; trees in middle size class had smallest mortality, and the proportion of the trees killed by disturbances increased with size. Gap creations were concentrated in a particular year, suggesting a large heterogeneity in time. Spatial distribution of recruited trees were biassed to the old gaps (older than 4 years), especially that of the species with Bell-shaped dbh distribution (shade intolerant) strongly associated with the gaps. Recruitment in tree stems and the loss of basal area, thus had the larger variability than mortality of stems and this forest, and the species with L-shaped dbh distribution seemed to going to increase the importance in the future if the present trend continues to be held. The turnover time of population is positively correlated with the maximum dbh size of the species, indicating the slow change of the population of large sized species.
