    Notes: Photoassisted heteroepitaxy of ZnSe on GaAs by metalorganic molecular beam epitaxy has been performed using sources of diethylselenium and diethylzinc. Illuminating the substrate during growth with an Ar ion laser has been observed to significantly enhance the growth rate. Growth rate enhancement was found to be a function of substrate temperature, VI/II gas flow ratio, and laser wavelength and intensity. Photons having energies sufficient to generate electron/hole pairs in the growing ZnSe film resulted in growth rate enhancement. The photoassisted growth has application for (i) increasing the anomalously low growth rate which is observed, (ii) assisting in tuning the surface stoichiometry, and (iii) providing for selective area epitaxy.

    Notes: Summary It was the aim of this study to investigate the antileukemic activities of recombinant interferon beta (rIFN beta) in chronic-phase CML in vitro and in vivo. Nine patients in the early chronic-phase of CML were treated in a phase-II trial with escalating doses of rIFN beta. In parallel, antiproliferative and immunomodulatory activities of rIFN beta and rIFN alpha 2b were studied in vitro. rIFN beta exhibited a significantly higher antiproliferative activity on hematopoietic progenitor cells of CML patients in vitro than rIFN alpha 2b. In contrast, only very limited clinical antileukemic efficacy of rIFN beta was observed. None of the patients achieved a complete or partial hematologic response (0% response rate, 0–36% 95 C.I.). Primary resistance of CML patients to rIFN beta treatment was caused neither by antibody formation against the recombinant material nor by deficient IFN receptor targeting and/or signaling: Induction of serum levels of beta-2-microglobulin (Β-2-m) and neopterin after administration of rIFN beta was comparable to that seen after administration of rIFN alpha. However, rIFN beta treatment less effectively induced biosynthesis of interleukin-1 receptor antagonist protein (IL-1-Ra) than rIFN alpha 2b. Thus, we conclude that rIFN beta at doses up to 12 MU/day s.c. is ineffective for treatment of chronic-phase CML. Further investigations into divergent biologic responses to various type-I interferons might help to elucidate mechanisms crucial for IFN action in patients with CML.

    Notes: Summary Experimental animal and human in vivo studies have previously demonstrated the impact of exogenous administration of various cytokines on frequencies of circulating myeloid and LAK precursor cells. For the first time we investigated whether exogenous cytokines, in the absence of antigeneic challenge, may also influence frequencies of circulating antigenspecific cytotoxic T-lymphocyte precursor cells. We further asked whether triggering of autoimmune pathways as has been reported for several cytokines can be confirmed on the cellular level by demonstration of induction of autoreactive CTL-p. Limiting dilution analysis was used to determine alloreactive CTL-p frequencies in 31 patients with nonhematologic diseases before and after short-term systemic treatment with either rIL-2 (4.8×106 IU/m2 bid), rIL-3 (2.5, 5.0 or 10.0μg/kg qd), rGM-CSF (5μ/kg qd), rIFN-gamma (200 or 400μg qd), or IFN-alpha (3 or 5×106 IU qod). Simultaneously, autoreactive CTL-p frequencies were determined by split-well analysis in 25 of these patients. We found that rIL-2 significantly expands the circulating precursor pool of alloreactive CTL (p〈0.05). rIL-3 affected CTL-p frequencies in a dosedependent fashion. Low and intermediate doses of rIL-3 did not exhibit significant effects, whereas 10μg/kg rIL-3 led to expansion of alloreactive CTL-p in the same order of magnitude as did rIL-2. This effect was statistically significant when compared with rGM-CSF (p〈0.02), which apparently had no influence on alloreactive CTL-p frequencies. In contrast to rIL-2 and rIL-3, exogenous rIFN-gamma markedly reduced the circulating precursor pool of CTL. This again was statistically significant compared with rIFN-alpha (p〈0.03), which, like rGM-CSF, did not exhibit any effects on the level of alloreactive CTL-p. Frequencies of autoreactive CTL-p were invariably below the limit of detection in our system (〈1/300000). In conclusion, these data demonstrate that (a) short-term systemic administration of rIL-2, rIL-3, and rIFN-gamma differently affects the clone size of circulating precursors of alloreactive CTL in man, while rGM-CSF and rIFN-alpha do not exhibit measurable effects, and (b) none of the cytokines administered is capable of uncovering detectable frequencies of autoreactive CTL-p.

    Notes: Abstract Infrared absorption of the vibrational band of H2 in the bulk is known to arise from overlap and quadrupolar interactions. For adsorbed H2, in addition to intermolecular interactions, an interfacial mechanism of inducing absorption has been identified in porous Vycor glass. A large enhancement of the fundamental vibrational line (by a factor of 60) from the bulk H2 (liquid) has been observed. The vibrational overtone absorption is similarly enhanced by 150. We discuss these results as well as infrared absorption measurements for H2 on MgO and silica gels.

    Notes: Summary Aggressive chemotherapy of advanced myelodysplastic syndrome (MDS), acute myeloid leukemia (AML) evolving from MDS, subacute AML and secondary AML has usually been associated with low complete remission (CR) rates, a high incidence of early death, and low disease-free survival. We therefore have initiated a phase-III trial of aggressive chemotherapy consisting of idarubicin, cytosine arabinoside, and VP-16 to improve the CR rate. Each chemotherapy cycle is followed by G-CSF to accelerate neutrophil recovery and to reduce the incidence of infections. Until now, 19 patients with high-risk AML have been entered. The CR rate is 47%, with only one death during induction. Patients achieving CR are randomized to receive either high-dose or low-dose interleukin-2 to eliminate residual leukemic cells and to prolong the duration of remission.

The tungsten-containing aldehyde oxidoreductase is inactivated to 50% by 3 mM KCN within 1 min, by 1 mM ferricyanide within 5 min, and by 0.05 mM chloralhydrate within 30 s. The molybdenum-containing AOR shows 50% inactivation within 1 min only with 70 mM KCN. The tungsten-containing enzyme is very sensitive to oxygen, especially in the reduced state, whereas the molybdenum-containing enzyme exhibits only moderate oxygen sensitivity without being markedly influenced by the redox state of the enzyme. The tungsten in the aldehyde oxidoreductase is bound to a pterin cofactor (Wco) of the mononucleotide form that is known for molybdopterin cofactor (Moco). The nature of the molybdenum cofactor in the molybdenum-containing aldehyde oxidoreductase is still unclear. The UV/VIS spectrum of the tungsten-containing aldehyde oxidoreductase shows a broad absorption in the range of 400 nm with a millimolar absorption coefficient of 18.1 (reduced form) and 24.8 (dehydrogenated form) at 396 nm. The epr spectrum exhibits two different W(V) signals with the following g values for signal A: 2.035, 1.959, 1.899 and signal B: 2.028, 2.017, 2.002. Dithionite-reduced enzyme shows signals of 4Fe−4S or 2Fe−2S clusters. Initial rate studies with different substrates for the carboxylate reduction led to a Bi Uni Uni Bi mechanism.

    Notes: Abstract Hydrogen adsorption isotherms have been measured at temperatures between 15 K and 20 K for porous Vycor glass. The intermediate adsorption data were analyzed using a modified Brunauer-Emmett-Teller method, which is based on the assumption that two layers are coadsorbed, the first layer being very strongly attached. Our results indicate that, relative to the adsorption energy of the free surface of the bulk liquid, the adsorption energy of the second layer is approximately 60 K, much smaller than the adsorption energy of the first layer but still significant at the low temperature of the experiments. Our results are compared to those for adsorption of4He on Vycor.
