    Notizen: Abstract— —This paper presents preliminary results from a large experimental programme to study geometry and size effects in CTOD R-curves. The results presented were obtained from unloading compliance R-curve tests performed at room temperature on different sized single-edge-notch bend specimens made from Ti–3Al–2V alloy. The crack growth resistance was measured in terms of both the conventional CTOD, δ0, (i.e. as defined in BS 5762) and CTOD corrected for crack growth, δR. It was found that the δ0 and δR R-curves were independent of specimen size up to crack extensions corresponding to approximately 10–15% of the original uncracked ligament. Also, after crack extensions of 30–40% of the initial ligament the δ0 and δR R-curves exhibited well defined upswings. The upswing in the CTOD R-curves is thought to be a result of a reduction in crack tip constraint.

It was found that the best agreement was exhibited by the R-curves based on the standard fracture resistance J, which displayed reasonable size independence up to, and in many cases beyond crack growths corresponding to 50% of the initial uncracked ligament.

    Notizen: Abstract The effect of monensin on the secretion of α-amylase and other enzymes from the aleurone layer of barley (Hordeum vulgare L. cv. Himalaya) was studied by electrophoresis followed by fluorography and by pulse-chase and organelle-isolation experiments. Monensin markedly inhibits the secretion, but not the synthesis, of α-amylase, acid phosphatase, and at least four other proteins from the aleurone layer. Monensin treatment causes α-amylase to accumulate within the protoplast, but its effect on the different α-amylase isoenzymes is not equal. The accumulation of isoenzyme 2 is not influenced by monensin while isoenzymes 1, 3 and 4 are not secreted but rather accumulate in the cell when monensin is included in the incubation medium. The α-amylase and acid-phosphatase activities which accumulate within the aleurone cells following treatment with monensin are localized in an organelle having a buoyant density greater than that of endoplasmic reticulum and less than that of mitochondria. In pulse-chase experiments with [35S]methionine, labelled proteins accumulate in this organelle in the presence of monensin and do not appear in the incubation medium. We conclude that monensin inhibits the secretion of proteins from the barley aleurone layer by influencing their intracellular transport.

    Notizen: Summary The behaviour of gibberellins A4, A5 and A7 (GA4, GA5 and GA7) has been studied in various in vitro hydroxylating systems. Incubation of GA4 and GA7 in peroxidase/dihydroxyfumarate or tyrosinase/ascorbate formulations resulted in the production of additional biologically-active zones on thin-layer chromatograms of the reaction mixtures. The product from GA7/peroxidase incubations was conclusively identified as GA7 norketone by gas/liquid chromatography-mass spectrometry and nuclear magnetic resonance spectrometry. A similar reaction is inferred for GA4 in the same system and for both GA4 and GA7 with tyrosinase/ascorbate. By analogy, the product derived from GA5 in the non-enzymatic ascorbate/ferrous iron system may be GA5 norketone. The reaction kinetics and the biological activity of the products are discussed.

    Notizen: Summary The sieve tube wall possesses a broad inner layer often with pronounced radial striations. The plasmalemma of the sieve tube appears to penetrate this wall in the form of a “brush border” of irregular microvilli, greatly increasing its surface area. It is suggested that this is the site of active transport of potassium, which circulates electroosmotically through the sieve plate pores and back through the thick wall. The function of the companion cells is the “care and maintenance” of the active “brush border” sites; in conjunction with their activity in supplying high-energy intermediates movement in the column acts regeneratively and fully polarises the plates. Many of the lamellar stacks and curvilinear membrane aggregates hitherto regarded as endoplasmic reticulum are, it is suggested, plasmalemma displaced from the wall. These findings have important consequences for the electroosmotic theory.

    Notizen: Summary Platelet aggregation to collagen in 12 Type 1 (insulin-dependent) diabetic patients with background retinopathy and 12 Type 1 diabetic patients with proliferative retinopathy was compared with an age- and sex-matched control group. An analogue of prostaglandin H2, 11,9 epoxymethano-prostaglandin H2, which directly stimulates thromboxane receptors, and EP 092, which is a competitive thromboxane A2 receptor antagonist, were used to investigate changes at platelet thromboxane receptor level in these groups. The concentration of collagen (EC50) required to give 50% of maximum aggregation did not differ between the two diabetic groups and the control group. However, platelets from the proliferative retinopathy group were significantly more sensitive to the thromboxane mimetic (11,9 epoxymethano-prostaglandin H2) (p 〈 0.005) than the background retinopathy and control groups. This change may be a factor in the development of proliferative retinopathy.
