Notes: Summary An experimental animal model of human ulcerative colitis using lipopolysaccharide (LPS) was studied. Rabbits were skin-sensitized by LPS and challenged with intrarectal instillation of LPS after 1% formalin enema. The course of experimental colitis was followed by performing serial colonofiberscopic examinations and biopsy. Petechiae appeared from the 8th hour, and ulcers and bleeding on the 3rd day. Mild macroscopic changes continued for about 2 weeks. By repeating the LPS enema after the initial treatment, the colitis was maintained for over 1 month. Control groups without formalin enema revealed no macroscopic changes, and the groups with only formalin enema showed mild transient changes. The endotoxin level in the blood during the experiment increased (36 pg/ml) at 24 h after the treatment in the LPS-sensitized group, while non-sensitized control rabbits had higher levels of endotoxin. Though fibrinogen and PTT levels had increased at 24 and 72 h, these levels were more marked in the control rabbits. The direct reaction of LPS was minimal, and local immune reaction by LPS seems to play an important role in the perpetuation of experimental colitis. Tissue fibrinolysis of the colon increased significantly as the mucosal damage appeared. This experimental colitis with LPS may be useful as a model of human ulcerative colitis.

Notes: Summary One hundred and twenty definite or classical rheumatoid arthritis (RA) patients with an average duration of the disease of 12.1 years were studied. Sixty-two patients had distinct atrophy of the first dorsal interosseous of the hand without definite signs of carpal, cubital, or ulnar tunnel syndrome (group A); 43 patients showed neither distinct atrophy nor sensory disturbance of either hand (group B). Other patients had sensory and/or motor disturbances due to carpal, cubital, or ulnar tunnel syndrome and other neuropathies. Electrodiagnostic examinations revealed that there were differences in the distal latency to the first dorsal interosseous muscle from the wrist between 24 group-A patients and 14 normal controls (P 〈 0.05), and between the group-A patients and 12 group-B patients (P 〈 0.1). The results of this study indicate that some RA patients with atrophy of the thumb web space may have compression neuropathy of the most distal branches of the ulnar nerve.

Notes: Summary We describe a family from Japan displaying the mutant insulin syndrome with hyperinsulinaemia and an increased insulin: C-peptide molar ratio. Serum insulin isolated from several family members showed reduced in vitro biological activity, and analysis by high performance liquid chromatography revealed a peak co-eluting with human insulin and a second species of increased hydrophobicity co-migrating with the previously reported Insulin Wakayama. The insulin genes from the propositus were cloned and sequenced, revealing one normal allele; the second allele, encoding a leucine for valine amino acid substitution at position 3 of the insulin A chain, was similar to that previously described for Insulin Wakayama. Synthesized [LeuA3] insulin showed 0.14% of receptor binding activity on rat adipocytes and a 10-fold prolonged half-life in a somatostatin-infused dog compared with human insulin. The finding of the same mutant gene in two unrelated Japanese families suggests that Insulin Wakayama may be discovered in additional Japanese families with hyperinsulinaemia and/or diabetes.

Notes: Summary To clarify whether activity of the sympathetic nervous system is decreased in streptozotocin-induced diabetic rats, noradrenaline turnover, which is a reliable indicator of sympathetic nervous system activity, was measured in the interscapular brown adipose tissue, heart and pancreas of streptozotocin diabetic rats. Results from studies using inhibition of noradrenaline biosynthesis with α-methyl-p-tyrosine demonstrated significant reductions (p〈0.05-0.001) in sympathetic nervous system activity in the interscapular brown adipose tissue, heart and pancreas of streptozotocin (65 mg/kg) diabetic rats, compared with measurements in streptozotocin (35 mg/kg) diabetic and saline-control rats. The daily injections of neutral protamine Hagedorn insulin to streptoz/otocin (65 mg/kg) diabetic rats prevented the decrease of noradrenaline turnover in the interscapular brown adipose tissue and heart significantly (p〈0.02), but this was less marked in pancreas, compared with non-treated streptozotocin (65 mg/kg) diabetic rats. Furthermore reduced noradrenaline turnover was also observed in the control rats which showed comparable changes in body weight to the rats injected with streptozotocin (65 mg/kg). These results suggest that poorly controlled streptozotocin diabetic rats may have reduced sympathetic nervous function, and that insulin therapy might prevent this.

Notes: Summary A method is described which records efferent motor nerve action potentials (MNAPs), evoked by trans-skull motor area stimulation, directly from a nerve root or peripheral nerve fibres during operation. The records of MNAPs from the root show whether function of the anterior root is intact and its continuity retained. When recorded from the proximal stump of a divided nerve at the wrist the motor funiculus can be identified, enabling funicular nerve suture to be carried out more easily.

Notes: Abstract The trials to use Azolla as a green manure for rice culture were made in the Niger basin.Azolla pinnata (Niger isolate) was used for the experiments. The effect of phosphorus on the growth and N2-fixation was examined in the field and in the laboratory. The growth rate and N content were maximum with P 3.1 ppm culture solution under laboratory conditions. The threshold P content for the growth was 0.5–0.6% in the dry matter. Maximum N content was 4.1% in the laboratory culture. In the field culture, the effect of P fertilizer on the growth and N yield of Azolla was tested. The split application of 6.5 kg P ha−1 per 13 days was most effective in stimulating the growth of Azolla. One kg of P as triple superphosphate produced 3.66 kg N in the Azolla. Maximum growth rate and N content in the field trials was 4.3 days (doubling time) and 2.3%, respectively. The lower productivity in the field in comparison with the laboratory culture was considered to be due to higher temperature and light intensity. the growth of Azolla was suppressed in the hot season in the Niger basin. The growth rate and N content were reduced during the high temperature period over 30°C on an average. The effect of inoculation of Azolla on rice yield was tested in the field experiment. The grain yield was increased 27% by Azolla incolation over the treatment without Azolla inoculation in — N fertilizer treatments. While the growth of Azolla with rice plants did not attain saturated density (1.8 kg fresh weight m−2), the effect on the grain yield was comparable to 40 kg N ha−1 as urea.
