Description / Table of Contents: The degree of electric charging of metal surfaces due to retarding of neutral metallic and semiconducting particles having various velocities has been determined. The effects are supposed to result from the exo-emission of electrons the amount of which depends on the velocities, masses, and impact angles of the particles. From the strong differences between the reactivities of solid substances during and after mechanical working it is concluded that the observed exo-emission is caused by exitation states at submicroscopic surface points.

Notes: Summary The effect of the indirect sympathomimetic agent tyramine on the isoprenaline-induced increase in plasma renin concentration was investigated in conscious rats. Tyramine caused a dose-dependent decrease in the isoprenaline-induced elevation of plasma renin concentration. Pretreatment of the rats with reserpine abolished this effect of tyramine, indicating that tyramine released catecholamines which acted on the inhibitory adrenoceptors. Pretreatment with phenoxybenzamine, an α-adrenoceptor antagonist, also abolished the inhibitory effect of tyramine on renin release, indicating that α-adrenoceptors mediated the observed inhibition of renin release. In rats with chronically denervated kidneys tyramine did not inhibit renin release. It is concluded that catecholamines which are released from renal sympathetic nerve endings can suppress renin release by activating α-adrenoceptors.

Notes: Summary Bilateral adrenalectomy combined with a sodium-deficient diet caused a time-dependent increase in plasma renin concentration in rats. Seventy-two hours after adrenalectomy the inhibitors of prostaglandin biosynthesis indomethacin and meclofenamate diminished the plasma renin concentration by about 50%. This effect was independent of the amount of renin released. Indomethacin and meclofenamate fully retained their ability to reduce the plasma renin concentration when the renal sympathetic nerves or the macula densa cells of the kidneys no longer contributed to renin release. It is concluded that in adrenalectomized rats renal prostaglandins affect the baroreceptor-mechanism in the afferent arterioles and thus enhance renin release.

Notes: Summary The importance of the sympatho-adrenal system for the isoprenaline-induced increase in plasma renin concentration was investigated in conscious rats. Ganglionic blockade by trimethidinium (10 mg kg−1) increased the dose-dependent elevation of plasma renin concentration induced by isoprenaline (0.03–0.48 μg kg−1 min−1). Also treatment of the rats with guanethidine (6 mg kg−1) or reserpine (2.5 mg kg−1, given 16 and 7 h prior to the experiments) further increased the effect of isoprenaline (0.5 μg kg−1 min−1) on plasma renin concentration. Unilateral renal denervation combined with contralateral nephrectomy doubled the effect of the β-sympathomimetic amine on renin release. The α-adrenoceptor antagonist phenoxybenzamine (3 mg kg−1) also enhanced the effect of isoprenaline on this parameter. It is concluded that apart from a stimulation of renin release via β-adrenoceptors the sympathetic nervous system may inhibit renin release via stimulation of α-adrenoceptors.

Notes: Summary The mechanism of the increase in plasma renin concentration caused by the β-sympathomimetic agent isoprenaline has been further investigated. Rats were pretreated by occluding the left renal artery for 2 hrs, thus rendering the macula densa cells of this kidney nonfunctioning. After contralateral nephrectomy infusion of isoprenaline (1.5 μg/kg min) still caused a strong increase in plasma renin concentration. This increase was significantly suppressed by simultaneous infusion of angiotensin II (1.0 μg/kg min), the α-sympathomimetic amine phenylephrine (60 μg/kg min) or octapressin (10 mU/kg min). The results exclude any mediator-role of the macula densa receptors in the isoprenaline-induced release of renin. The possibility of a stimulation of renin release via the baroreceptors or a direct “secretomotoric” action of isoprenaline is discussed.

Notes: Summary Plasma renin concentrations in rats increase after bilateral adrenalectomy without sodium substitution. The effects of i.v. infused (asp1-β-amid, val5)-angiotensin II (1 μg/kg min), felypressin (phen2, lys8-vasopressin) (40 mU/kg min) and phenylephrine (30 μg/kg min) were investigated on the increase in plasma renin concentration. These effects of the agents were compared with their actions on blood pressure, heart rate and renal hemodynamics. In rats with destroyed macula densa cells the effect of bilateral adrenalectomy without sodium substitution was also studied. Adrenalectomy still increased the plasma renin concentration. Angiotensin II and felypressin, also depressed under these conditions the elevation of plasma renin concentration caused by adrenalectomy. The mechanism of the adrenalectomy-induced reinin release and its suppression by vasoconstrictor agents is discussed.

Notes: Light scattered from a macromolecular solution in a capillary tube is used to determine both the sedimentation and translational diffusion coefficients. The capillary tube is spun in a preparative centrifuge, removed, and placed in a light-scattering photometer equipped with a scanning mechanism. The intensity distribution of scattered light along the tube represents the concentration profile in the tube and provides the measure of boundary migration. The sedimentation coefficient is determined from this measure and the applied centrifugal field. The diffusion coefficient is obtained from a time-autocorrelation analysis of fluctuations in intensity of light scattered from any fixed point of the profile. These coefficients were obtained for two monodisperse systems, R17 bacteriophage and 28s ribosomal rat liver RNA. The molecular weights obtained from ratios of these coefficients are in good agreement with literature values. In the sedimentation analysis, deviations from linearity between boundary displacement and applied field were found to be less than 1%. This precision confirms that the boundary is stable for the capillary geometry even in the absence of a preformed density gradient. The sedimentation coefficients of identical samples were also measured with the Spinco Model E analytical ultracentrifuge; results of the two methods agree to within 4%.As a consequence of the capillary tube geometry and light-scattering detection, sedimentation coefficients can be obtained from sample volumes of less than 100 μl. This detection techniques is thus far demonstrated to be at least an order of magnitude more sensitive than Schlieren optics, thereby useful when uv absorption is not applicable. For diffusion measurements there are also several inherent advantages. The diffusion coefficient is obtained from the identical sample, and scanning provides the capability to measure D from various parts of the sedimentation profiles and thereby directly explore concentration dependence, homogeneity, and integrity of the sample. The capillary tube with a layer of silicone oil over the sample and centrifugation provides an effective method to cleanse the solution and trap all dust.
