    Description / Table of Contents: Zusammenfassung Das feste β-Carotin ist gegen γ-Strahlung ziemlich stabil. Bei Bestrahlung unter Sauerstoffbegasung treten deutliche β-Carotin-Verluste erst bei der hohen Dosis von 12 Mrad auf; bei Bestrahlung an der Luft werden ähnliche Radiolyseerscheinungen erst durch wesentlich höhere Strahlendosen ausgelöst. Spaltungsprodukte, die bei 12 Mrad und Sauerstoffbegasung auftraten und dünnschichtchromatographisch aufgetrennt werden konnten, waren Isozeaxanthin, β-Carotin-5,6-5′, 6′-diepoxid, β-Carotin-5,8-5′,8′-diepoxid, β-Apo-12′-carotinal, β-Apo-10′-carotinal, 3,3′,6′-Trihydroxy-α-carotin-5,8-epoxid und Vitamin-A-Alkohol. Es ist bemerkenswert, daß ein Teil der Radiolyseprodukte (z. B. Vitamin-A1-Alkohol, β-Apo-12′-carotinal und β-Apo-10′-carotinal) Vitamin-A-Wirksamkeit besitzt.
    Notes: Abstract Solid β-carotene is remarkably stable to γ-irradiation. In an oxygen atmosphere doses as high as 12 Mrad were required to bring about significant losses of β-carotene, whereas in air even larger doses had to be applied in order to effect a comparable degree of radiolysis. Cleavage products which arose in O2 at a dose of 12 Mrad and which could be separated from each other by thin-layer chromatography were isozeaxanthin, β-carotene-5,5-5′,6′-diepoxide, β-apo-12′-carotenal, β-apo-10′-carotenal, 3, 3′, 6′-trihydroxy-α-carotene-5,8-epoxide and vitamin A1. It is notable that some of the radiolysis products (e.g., vitamin A1, β-apo-12′-carotenal and β-apo-10′-carotenal) possess vitamin A activity.

    Notes: Summary The daily rhythm (8–14 h, 14–20 h, 20–2 h, 2–8 h) of HVA, VMA, (VA) and 5-HIAA was studied in 21 depressed patients and was compared to the values of 13 healthy subjects. In healthy subjects all compounds trend to high values during day-time and significant lower values in the nightphase (2–8 h). Depressed patients did not show a significant rhythm during the four fractions; the values of HVA and VMA were significant lowered in the morning-phase (8–14 h). This demonstrates a good connexion to the clinical feature with the morning listlessness. Normal concentrations were reached in the night-phase (2–8 h). 5-HIAA did not show any significance between healthy subjects and depressed patients during the circadian rhythm. The ratio VMA/HVA in healthy subjects fell significant from morning-phase (8–14 h) to night-phase (20–2 h, 22–8 h). In depressed patients the ratio was approximately equal in all fractions. Whereas the ratio 5-HIAA×100/ VMA〉+HVA increased from the morning-phase to the night-phase in healthy persons, this ratio did not show any significance in the four fractions of depressed patients. Also these results suggest some importance in explaining the lost of drive in the activity of depressed patients during morning hours and the remission of these patients often observed in the evening. A very good correlation to the clinical feature is emphasized. Preliminary studies on 6 patients with different depressive diagnosis during depression- and remission-phase gave a different biochemical pattern. Further extended studies have to be carried out to differentiate exactly between nosological different depressions.

In various areas of the brainstem: nucleus caudatus, putamen, gyrus cinguli, oral and caudal part of substantia nigra, nucleus amygdalae, globus pallidus, raphe, and nucleus ruber L-tyrosine, dopamine, noradrenaline, L-tryptophan, serotonin and 5-hydroxyindoleacetic acid were investigated in eight cases of deceased L-Dopa psychosis, six cases of parkinsonian patients without psychotic symptoms, and in nine controls. 2. The values of L-tyrosine in all investigated areas were increased in parkinsonian patients as well as in L-Dopa psychosis. 3. The dopamine levels in all areas were decreased in both groups. 4. Noradrenaline shows moderate slight increase in six out of nine regions in the psychotic group, when compared to parkinsonian patients without psychotic symptoms. 5. L-tryptophan levels in the psychotic group are decreased in all areas, whereas levels in parkinsonian patients without psychotic symptoms are equal to those of the Controls. 6. The 5-hydroxytryptamine levels in the psychotic group are increased in eight of nine regions, when compared to parkinsonian patients without psychotic symptoms, predominantly in the nucleus ruber, somewhat less in the raphe, and in the globus pallidus. 7. 5-hydroxyindoleacetic acid levels show a similar pattern as 5-HT. 8. The loss of balance will be discussed as trigger of the psychotic symptoms.

    Notes: Summary An accumulation of norepinephrin, serotonin and dopamin exists in the brainstem. A decrease of biogenic amines in the brains of Parkinson's cases has been demonstrated. A substitution by the precursor L-DOPA produces an increase of dopamin in the brain and a decrease of serotonin. The high level of dopamin improves the akinesia and the depletion of serotonin causes a toxic delir. A substitution by l-tryptophan restores the balance and the Dopa-psychosis disappears.

    Notes: Summary There were determined free plasma tyrosine and tryptophan spectral-photofluorometrically at 22 healthy controls and 81 depressed patients (13 of them were controlled in a long-time-study; mean time: 11 months, and 68 patients were controlled ambulant). The biochemical data were demonstrated in a tyrosine-tryptophan diagram and were correlated with the clinical behaviour of the patients. This correlation only was possible with the postulation of a balance between catecholamine-and indolamine metabolism. Three biochemical parameters are necessary. The values of 1. tyrosine that of 2. tryptophan and not only 3. the resulting quotient. With help of such demonstration we showed this tyrosine-tryptophan balance graphically. We think, that both catecholamine-and the indolamine-hypothesis of depression seem to have their availability.

    Notes: Summary Intravenous or oral application of L-Dopa increased not only the plasma level but led at the same time to an improvement of the kinetic behaviour. A combined intravenous or oral administration of L-Dopa and the decarboxylase inhibitor Ro 4-4602 stop the damage of this peripheral L-Dopa-metabolism for the most part and therefore a higher L-Dopa-plasma-level is reached and the increase lasts longer. A reduction of L-Dopa dosages is the clinical result. A potentiation of clinical and biochemical effects we found in three of ten failures after a medication of L-Dopa plus amantadine-sulfate-infusion. Two of the ten failures showed a clinical deterioration and also diminished L-Dopa-plasma levels after L-Dopa plus amantadine-sulfate-infusions; five of ten failures demonstrated no change in the plasma level after application of amantadine-sulfate-infusions plus L-Dopa. For producing kinetic effects a sufficient availability of L-Dopa in plasma is necessary and a sufficient utilization in the brain has to be. In cases of akinetic crisis a combined infusion of L-Dopa plus amantadine-sulfate is a vital indication.

    Description / Table of Contents: Zusammenfassung Es wird über eine neue dünnschichtchromatographische Methode berichtet, mit der eine Bestimmung des Coffeingehaltes im Kaffee möglich ist. Der nach dem Verfahren von Cortes hergestellte Rohcoffeinextrakt wird auf Kieselgel GF254-Platten gereinigt und die Coffeinfleckenfläche in Abhängigkeit von der Konzentration ermittelt. An Hand einer Eichkurve, die bei bestimmten Coffeinmengen und geeignet gewählten Diagramm-Maßstäben linear verläuft, wurde die Coffeinmenge in fünf verschiedenen Kaffeesorten bestimmt. Das dünnschichtchromatographische Verfahren ist der Methode von Cortes, nach welcher der Stickstoffgehalt im Rohcoffeinextrakt bestimmt und daraus die Coffeinmenge ermittelt wird, an Zeit und Arbeitsaufwand insofern etwas überlegen, da auf einer Dünnschichtchromato-graphie-Platte mehrere Probeextrakte zugleich untersucht werden können. Dies kompensiert die etwas geringere Analysengenauigkeit (±10%) der Dünnschichtchromatographie-Methode gegenüber dem Verfahren von Cortes (±5%).
    Notes: Summary It is referred about a new thin layer chromatographic method, by which it is possible to determine the content of caffeine in coffee. The raw caffeine extract, prepared by the procedure of Cortes, is purified on silica gel GF254 plates and the area of the caffeine spots in relation of the concentration is investigated. By means of a standard curve, which progresses linear by determined quantities of caffeine and by suitable selected diagram scale, the quantity of caffeine in five different sorts of coffee was examined. The thin layer chromatographic procedure is, with regard to time and work expanse a little better than the method of Cortes, by which the quantity of nitrogen is determined, because it is possible to examine several extracts of samples together on one thin layer chromatographic plate. By that the smaller accuracy of analysis (±10%) of the thin layer chromatographic method compared with the procedure of Cortes (±5%) is compensated.
