Summary The authors use the method ofErkoçak andBucher (1961, 1962) in order to produce — by a single intraperitoneal injection of a weak dose of alloxan — an overloading of the proximal tubule of the nephrons due to an increased reabsorption of glucose. The injected dose of alloxan (4 mgs. for a mouse of 25 grs.) has no toxic effect on the proximal tubule; there are no pathological alterations, neither at the light microscope nor at the electron microscope level. However, the overloading of the proximal tubules is discernible by some minute modifications of their ultrastructure. So one finds, four days after the start of the experiment, an augmentation of the juxtamembranous vesicles which, for the first time, are also described in the kidney of untreated animals. Moreover, the lysosomes seem to be more frequent and the endoplasmic reticulum better developed.

Zusammenfassung Die Verfasser benutzen die Methode vonErkoçak u.Bucher (1961, 1962), um — durch eine einmalige intraperitoneale Einspritzung einer kleinen Dosis von Alloxan — eine gezielte Mehrbelastung des Hauptstückes der Nephrone zu erhalten; diese beruht auf der erhöhten Rückresorption von Glucose. Die eingespritzte Alloxanmenge (4 mg je Maus von 25 g) hat keine toxische Wirkung auf das Hauptstück; sie erzeugt keine pathologischen Veränderungen, weder im lichtmikroskopischen, noch im elektronenmikroskopischen Bereich. Die Mehrbelastung der Hauptstücke äußert sich jedoch in gewissen geringgradigen Änderungen der Ultrastruktur. So findet man, vier Tage nach Versuchsbeginn, eine Häufigkeitszunahme der juxtamembranösen Bläschen, auf deren Vorkommen in Nierenzellen auch der gesunden Kontrolltiere erstmals hingewiesen wird. Ferner scheinen die „Lysosomen“ häufiger und das endoplasmatische Reticulum besser entwickelt zu sein.

Résumé Les auteurs se servent de la méthode d'Erkoçak etBucher (1961, 1962) pour créer — par une seule injection intra-péritonéale d'une faible dose d'alloxane — une surcharge du segment initial des néphrons due à la réabsorption accrue du glucose. La dose alloxanique injectée (4 mg par souris de 25 g) n'a pas d'effet toxique sur le segment initial; elle ne produit pas d'altérations pathologiques, ni à l'échelle de la microscopie à lumière, ni même à celle de la microscopie électronique. En revanche, la surcharge des segments initiaux s'exprime par certains menus changements ultrastructuraux. Ainsi, on y trouve, quatre jours après le début de l'expérience, une augmentation des vésicules juxtamembraneuses que les auteurs décrivent les premiers aussi pour les reins d'animaux témoins. En outre, les „lysosomes“ semblent être plus nombreux et le réticulum endoplasmique plus développé.
