    Notes: Five yellow 2-hydroxy-5-methoxybenzoquinones with C15 and C17 alkyl side chains were isolated from fruitbodies of the wood-rotting fungus Phlebia chryoscrea. The compounds, named phlebiachrysoic acids A-E are strong inhibitors of leukotriene biosynthesis and are responsible for the red colour reaction when the fruitbodies are touched with aqueous alkali.

Geburtstag gewidmet.Two novel diterpenoids trichaurantin (1) and its 6-O-acetyl derivative 2 have been isolated from Tricholoma species. Their structures were established by spectroscopic methods and an X-ray crystal analysis, 1 and 2 appear to be biosynthetically related to the neodolabellane and dolastane group of diterpenes and can be designated as 2,3-seconeodolastane derivatives.

    Notes: In 4-Stellung substituierte 2-Trifluormethyl-oxazolone-(5) (I) sind aus α-Aminosäuren durch Erhitzen mit Trifluoressigsäureanhydrid oder aus N-TFA-α-Aminosäuren mit POCl3 in Pyridin erhältlich. Sie entstehen als Nebenprodukte auch beim Versuch, die Methode der Peptidsynthese über die gemischten Anhydride auf N-TFA-Aminosäuren anzuwenden. In anomaler Reaktion werden die 2-Trifluormethyl-oxazolone-(5) zu α-Ketosäuren hydrolysiert, wodurch viele Verbindungen dieser Gruppe leicht zugänglich werden, darunter auch die dem L-Isoleucin entsprechende (+)-2-Keto-3-methyl-valeriansäure in optisch aktiver Form. Durch Umsetzung von I mit Äthylmercaptan in HBr/Eisessig werden in vielen Fällen direkt die Mercaptole der α-Ketosäuren zugänglich; liegt jedoch im 4-ständigen Substituenten eine α-ständige Verzweigung vor, so bilden sich auch die α-Äthylmercapto-acrylsäure-Derivate. Das 2-Trifluormethyl-4-brom-4-methyl-oxazolon-(5) verhält sich bei der Umsetzung mit Äthylmercaptan in HBr/Eisessig wie die bromfreie Verbindung, da das Brom zunächst reduktiv entfernt wird.  -  Mit o-Phenylendiamin in Eisessig reagieren die Verbindungen I direkt zu 3-substituierten 2-Hydroxy-chinoxalinen. Lediglich mit Monoaminen findet normale Ringöffnung zu N-TFA-Aminosäureamiden statt, mit Anilin hingegen beobachtet man beim 2-Trifluormethyl-4-methyl-oxazolon-(5) eine gemischte Reaktionsweise.

Die Ausgangsverbindungen 7 sind aus α-Aminosäuren über α-Acylaminoketone 6 leicht zugänglich.

    Description / Table of Contents: Dakin West Reaction, IV1). - Reaction of α-Amino Dicarboxylic Acids with Carboxylic Acid Anhydrides2)The Dakin West reaction of N-acylaspartic anhydrides yields furo[3,2-d]oxazoles 5 and 14, 2(5H)-furanones 8 or 6H-1,3-oxazin-6-ones 9, depending on the reaction conditions. N-Thiobenzoylaspartic acid is converted into 6H-1,3-thiazin-6-one (12) by treatment with acetic anhydride and 3-picoline. Glutamic acid gives 1,5-diacetyl-2-pyrrolidone (22) in high yield on warming with acetic anhydride and 4-(dimethylamino)pyridine. Possible mechanisms for the formation of the different products are discussed.
    Notes: N-Acylasparaginsäureanhydride liefern bei der Dakin-West-Reaktion je nach den Reaktionsbedingungen Furo[3,2-d]oxazole 5 und 14, 2(5H)-Furanone 8 oder 6H-1,3-Oxazin-6-one 9. Aus N-Thiobenzoylasparaginsäure, Acetanhydrid und 3-Picolin entsteht das 6H-1,3-Thiazin-6-on (12). Glutaminsäure wird von Acetanhydrid und 4-(Dimethylamino)pyridin in guter Ausbeute in 1,5-Diacetyl-2-pyrrolidon (22) übergeführt. Mechanismen für die Bildung der verschiedenen Produkte werden diskutiert.

The substitution pattern of these new derivatives of (+)-dunnione (1) is in accord with a polyketide biogenesis of the quinone system.
    Notes: Aus Kulturen des Mycosymbionten der tropischen Rindenflechte Trypethelium eluteriae Sprengel wurden die antibiotisch aktiven 1,2-Naphthochinone Trypethelon (2), Trypethelon-methylether (3), 8-Methoxytrypethelon-methylether (5) und 4′-Hydroxy-8-methoxytrypethelon-methylether (6) isoliert. Es handelt sich um Derivate des (+)-Dunnions (1), deren Substituentenanordnung mit einer Polyketidbiogenese des Chinonsystems im Einklang ist.

    Description / Table of Contents: α-Amino Acids as Nucleophilic Acyl Equivalents, III.  -  Synthesis of (E)-3-Acylacrylic Esters and 4-Oxocarboxylic EstersBase catalyzed addition of methyl propiolate to oxazolin-5-ones 1 yields mixtures of the diastereomeric methyl 3-(5-oxo-2-phenyl-2-oxazolin-4-yl)acrylates 2 which may be converted into (E)-3-acylacrylates 5 via hydrolytic ring opening to 3 and subsequent oxidation with lead tetraacetate. Alternatively the vinylogous monomethyl malonates 3 are converted into 4-oxocarboxylates 4, by thermal decarboxylation followed by hydrolysis with oxalic acid dihydrate in acetic acid.
    Notes: Basenkatalysierte Addition von Propiolsäure-methylester an Oxazolin-5-one 1 ergibt Gemische der diastereomeren 3-(5-Oxo-2-phenyl-2-oxazolin-4-yl)acrylsäure-methylester 2, die nach hydrolytischer Ringöffnung zu 3 mit Bleitetraacetat die (E)-3-Acylacrylsäureester 5 liefern. Alternativ lassen sich die vinylogen Malonsäurehalbester 3 thermisch decarboxylieren und anschließend mit Oxalsäure-dihydrat in Eisessig zu den 4-Oxocarbonsäureestern 4 hydrolysieren.

    Description / Table of Contents: Fungus Pigments, 4141).  -  Naphthoquinone Pigments from the Slime Moulds Trichia floriformis and Metatrichia vesparium (Myxomycetes)Trichione (1) is the red main pigment of fruiting bodies from the slime mould Trichia floriformis. It contains a 2,5-dihydroxynaphthoquinone chromophore, substituted in position 3 by an unsaturated side chain carrying a terminal hemimalonate moiety. Homotrichione (7) differs by the presence of two additional CH2 groups in the side chain. It co-occurs with arcyriaflavin B (5), arcyriaflavin C (6) and further pigments in Metatrichia vesparium.
    Notes: Die Fruchtkörper des Schleimpilzes Trichia floriformis enthalten als roten Hauptfarbstoff Trichion (1), ein 2,5-Dihydroxynaphthochinon, das in 3-Stellung eine ungesättigte Seitenkette mit terminaler Malonhalbester-Gruppierung trägt. Homotrichion (7) unterscheidet sich nur durch eine um zwei CH2-Gruppen längere Seitenkette. Es kommt neben 1, Arcyriaflavin B und C (5 bzw. 6) und weiteren Farbstoffen in Metatrichia vesparium vor.

    Description / Table of Contents: Antibiotics from Basidiomycetes, XII.  -  Cristatic Acid, a Modified Farnesylphenol from Fruiting Bodies of Albatrellus cristatusFruiting bodies of Albatrellus cristatus contain high concentrations of cristatic acid (3), a farnesylphenol modified by incorporation of a furan ring in its side chain. A closely related American Albatrellus species contains grifolic acid (6) instead of 3. Biological screening of 3, 6 and related meroterpenoids revealed remarkable cytotoxic activities especially for dimethylcristatic acid methyl ester (4).
    Notes: Fruchtkörper von Albatrellus cristatus enthalten in hoher Konzentration Cristatsäure (3), ein Farnesylphenol, dessen Seitenkette durch Einbau eines Furanringes modifiziert ist. In einer nahe verwandten amerikanischen Albatrellus-Art kommt anstelle von 3 Grifolsäure (6) vor. Die biologische Aktivität von 3, 6 und verwandten Meroterpenoiden wird untersucht, wobei besonders die cytotoxische Wirkung von Dimethylcristatsäure-methylester (4) bemerkenswert ist.
