    Notes: Abstract A 6,9-nonadecadiene, (Z)-9-nonadecene, andn-nonadecane were identified in extracts of sex pheromone glands of femaleSabulodes caberata Guenée. Potential pheromonal activity of gland components was assessed by electroantennogram measurements of gas-chromatographic fractions of abdominal tip extracts. Chemical identification was based on gas chromatography and mass spectrometry of parent compounds, hydrogenation products, ozonolysis products, dimethyl disulfide adducts, and synthesis. The monoene was synthesized via coupling of alkyl and acetylenic intermediates. (Z,Z)-6,9-Nonadecadiene (Z6,Z9-19∶H) was synthesized from methyl linoleate via chain lengthening. SyntheticZ6,Z9-19∶H had the same retention times on polar and nonpolar capillary gas-chromatographic columns as the 6,9-nonadecadiene from the sex pheromone glands. In field testsZ6,Z9-19:H alone produced trap catch of maleS. caberata and addition of (Z)-9-nonadecene orn-nonadecane had no effect on trap catch.

    Notes: Abstract The most effective lure for maleAmorbia cuneana (Walsingham) in Orange, Ventura, and Riverside counties of California was previously found to be a 1∶1 ratio of (E,E)-10,12- and (E,Z)-10,12-tetradecadien-1-ol acetates. In subsequent field tests in San Diego and Santa Barbara counties, this lure was ineffective. Analysis of sex pheromone glands (SPG) of femaleA. cuneana from these two counties showed theEE:EZ ratio to be about 1∶9 and synthetic lures of this composition were highly attractive in these areas. Analysis of the SPG of a number of females from both areas showed that there were three population types: two in the low ratio areas possessed 37 and 58%EZ, and the third in the high ratio areas possessed 89%EZ.

    Notes: Abstract The major volatile components in the extract of the female sex pheromone gland ofAmorbia cuneana consisted of (E,E)- and (E,Z)-10,12-tetradecadien-1-ol acetates. The identification was based on electroantennogram bioassay of gas Chromatographic effluent from sex pheromone gland extract, relative retention times on polar and nonpolar gas chromatographic columns, chemical degradation (ozonolysis, saponification), mass spectrometry, chemical synthetic methods, and field tests. Based on mass spectrometry and retention times by capillary gas chromatography, traces of (E)-10-tetradecen-1-ol acetate and 1-tetradecanol acetate were also present in the extract. Traps baited with a combination of synthetic (E,E)- and (E,Z)-10,12-tetradecadien-1-ol acetates caught more males than did traps baited with females.

    Notes: The molecular weights and molecular weight distribution of a styrene-butadiene rubber produced at 5°C. according to an incrementally modified recipe with tert-dodecyl mercaptan modifier, do not differ significantly from those of similar conventionally modified rubbers. The limiting effect of diffusion upon the mass transfer of mercaptan in the system provides a consistent explanation of this finding.

    Notes: Based upon its superior catalytic activity for H2O2 decomposition, a bituminous coal-based activated carbon was selected for investigations of pretreatment and enzyme immobilization methods. Pretreatments considered include acid washing, exposure to strong oxidizing agents, contact with concentrated peroxide solutions, nitration and amination, isothiocyanate derivatization, silanization, and stearic acid coating. Effects of these pretreatments on morphology and trace-metal content of the carbon pellets have been studied using scanning electron microscopy and dispersive analysis of x rays. Immobilization of glucoamylase by adsorption, glutaraldehyde crosslinking, and covalent attachment to carbon activated by water-soluble diimide or diazotization have been examined. These different enzyme-carbon catalysts have been characterized by their enzyme loading, enzyme activity, catalytic activity for H2O2 decomposition, or combinations of these measures of performance.

    Notes: Proteins have been immobilized in porous support particles held in a fixed-bed reactor through which protein solution is continuously circulated. Changing the recirculation flow rate alters the observed immobilization kinetics and the maximum enzyme loading which can be achieved for glucose oxidase and glucoamylase on carbodiimide-treated activated carbon and for glucoamylase immobilized on CNBr-Sepharose 4B. Direct microscopic examination of FITC-labelled protein in sectioned Sepharose particles and indirect activity-loading studies with activated carbon-enzyme conjugates all indicate that immobilized enzyme is increasingly localized near the outer surface of the support particles at larger recirculation flow rates. Restricted diffusion of enzymes may be implicated in this phenomenon. These contacting effects may be significant considerations in the scaleup of processes for protein impregnation in porous supports, since apparent activity and stability of the final preparation depend on internal protein distribution.

    Notes: Widely applied selection strategies for plasmid-containing cells in unstable recombinant populations are based upon synthesis in those cells of an essential, selection gene product. Regular partitioning of this gene product combined with asymmetric plasmid segregation produces plasmid-free cells which retain for some time the ability to grow in selective medium. This theory is elaborated here in terms of a segregated model for an unstable recombinant population which predicts population growth characteristics and composition based upon experimental data for stable strain growth kinetics, plasmid content, and selection gene product stability. Analytical solutions from this model are compared with an unsegregated phenomenological model to evaluate the effective specific growth rate of plasmid-free cells in selective medium. Model predictions have been validated using experimental growth kinetics and flow cytometry data for Saccharomyces cerevisiae D603 populations containing one of the plasmids YCpG1ARS1, YCpG1ΔR8, YCpG1ΔR88, YCpG1ΔH103, YCpG1ΔH200, pLGARS1, and pLGSD5. The recombinant strains investigated encompass a broad range of plasmid content (from one to 18 plasmids per cell) and probability α of plasmid loss at division (0.05 ≤ α ≤ 0.42). Experimental data for all strains considered is inconsistent with the hypothesis that plasmid-free cells are unable to grow in selective medium. For a given value of a, the fraction of plasmid-containing cells in the population decreases with increasing plasmid content and increases for less stable selection gene products. This conceptual framework and mathematical model will aid in strain development for greater effective stability.
