    Notes: Abstract Objective: The correlations between steady-state plasma concentrations of mianserin and its active metabolite desmethylmianserin and those of trazodone and its active metabolite m-chlorophenylpiperazine (m-CPP) were examined in 19 depressed patients. Methods: Ten patients received first mianserin (30 mg per day) and second trazodone (150 mg per day), while 9 patients received these treatments in the opposite sequence, with at least 2-week intervals between the two phases. Blood was sampled at steady state, 1–3 weeks after initiation of each treatment. Plasma concentrations of mianserin, the separate enantiomers S(+)- and R(−)-mianserin, desmethylmianserin, trazodone and m-CPP were measured by means of high-performance liquid chromatography. Results: There was a significant correlation between steady-state plasma concentrations of trazodone and total mianserin (r = 0.59) or S(+)-mianserin (r = 0.57), but not R(−)-mianserin (r = 0.33). Conclusion: The present study thus suggests that the metabolic capacity of mianserin, especially the more active S(+)-enantiomer, and that of trazodone correlate to each other. This finding supports the previous suggestions that cytochrome P4502D6 is involved in the metabolism of mianserin and trazodone.

    Notes: Abstract  The effects of clozapine and other antipsychotic drugs on noradrenaline (NA) transport were examined in cultured bovine adrenal medullary cells and in transfected Xenopus laevis oocytes expressing the bovine NA transporter. Incubation of adrenal medullary cells with clozapine (30–1000 ng/ml) inhibited desipramine (DMI)-sensitive uptake of [3H]NA in a concentration-dependent manner (IC50=110 ng/ml or 336 nM). Other antipsychotic drugs such as haloperidol, chlorpromazine, and risperidone also decreased [3H]NA uptake (IC50= 144, 220, and 210 ng/ml or 383, 690, and 512 nM, respectively). Eadie-Hofstee analysis showed that clozapine reduced Vmax of uptake of [3H]NA and increased Km. Furthermore, clozapine inhibited specific binding of [3H]DMI to plasma membranes isolated from bovine adrenal medulla (IC50=48 ng/ml or 146 nM). Scatchard plot analysis of [3H]DMI binding revealed that clozapine decreased both Bmax and Kd. Other antipsychotic drugs, including haloperidol, chlorpromazine, and risperidone, also reduced [3H]DMI binding to the membranes. In transfected Xenopus oocytes expressing the bovine NA transporter, clozapine inhibited [3H]NA uptake in a concentration-dependent manner similar to that observed in adrenal medullary cells. These results suggest that clozapine and haloperidol directly inhibit transport of NA by acting on the site of an NA transporter that influences both substrate transport and binding of tricyclic antidepressants.

    Notes: A new type of microelectrode array device has been developed using semiconductor-processing techniques. An array of 15 × 15 square electrodes as small as 1 μm, spaced 100 μm apart, has been fabricated on a silicon chip of dimensions 1.5 × 1.5 mm2. Steady state electrochemistry was performed using these devices in both aqueous and non-aqueous media.

    Notes: Two commercial styrene-butadiene (SBR) latexes were used to prepare a model filled material consisting of glassy SBR filler particles about 1000 Å in diameter embedded in a rubbery SBR matrix crosslinked by γ-radiation. When transparent specimens of this material were extended, voiding occurred, as evidenced by stress whitening and greatly enhanced X-ray scattering intensity. More voids were formed at higher rates of extension, but voids disappeared when specimens were relaxed. The effects of filler content and cure time of the matrix on the size and number of voids formed were determined by low-angle X-ray scattering for a constant extension rate and a constant extension ratio λ = 1.6. The number of voids measured by X-ray scattering intensity decreased rapidly with time over the 3-h period of measurement. The number of voids remaining 1 h after extension increased about 40 times as filler content was increased from 15% to 50%. Increasing the cure time from 24 to 96 h increased the number of voids about four times. In contrast, the radius of gyration of the voids formed (250-350 Å) did not depend strongly on time, nor did it depend strongly on the filler content or the cure time of the matrix. Stress relaxation measurements made under the same conditions as X-ray scattering measurements showed effects typical of filled materials. However, the relaxation of stress (which followed a power law decay) was much slower than the decay of the number of voids as measured by X-ray scattering intensity.
