    Notes: Abstract We reported that apoptosis occurred in the guinea pig vestibular hair cells after chronic aminoglycoside treatments. In the present study, we used in situ nick-end labeling to determine whether apoptosis was also induced by the acute effects of aminoglycosides in guinea pig ampullar cristae. In addition, we evaluated the effect of zinc supplements upon these ototoxic treatments. After a local application of streptomycin directly to the round window, we found labeled bodies in the vestibular hair cells. The zinc supplement increased the number of labeled bodies resulting in severe hair cell loss. These findings indicate that the acute effects of aminoglycosides also induce apoptosis of the vestibular hair cells, and that zinc enhances aminoglycoside ototoxicity. Consequently, we propose that an interaction with ion channels may play a key mechanism in the processes of apoptosis affecting the vestibular hair cells.

    Notes: Summary We have examined the function and fine structure of the mucociliary system of the eustachian tube in an experimental study of otitis media with effusion induced by X-ray irradiation. Functional examination demonstrated that the ciliary activity was diminished in such a condition, while morphological observations showed pathological findings including compound cilia, vacuolation of ciliated cells and expansion of intercellular space. These findings show that irradiation-induced otitis media with effusion results in impairment of the mucociliary system. As evidenced by these studies, the mucociliary system in the eustachian tube has an important role in the clearance of fluid produced in the tympanic cavity as well as affording improvement in this disease.

    Notes: Abstract Effusion fluid resulting from otitis media contains a variety of inflammatory substances. This middle ear effusion (MEE) may be itself affect the mucosa of the tubotympanum and contribute to chronicity of the condition. The present study was designed to elucidate in vitro and in vivo effects of MEEs on the mucosa of the guinea pig tubotympanum. The results obtained demonstrated that the ciliary activity of the eustachian tube was reduced in the presence of human MEEs. This activity decreased to approximately 80% in the presence of serous MEE at 48 h and was 60% after exposure to mucoid MEE. Intratympanic inoculation of human MEEs resulted in accumulation of a serous effusion in the tympanic cavity. Histologic study of the tubotympanum in inoculated animals demonstrated mucociliary dysfunction as well as a general inflammatory process and increased vascular permeability. This damage was more prominent following inoculation with the mucoid MEEs. The ciliary depression and inflammation found in the tubotympanum suggest that the pathologic nature of MEEs may be, at least partially, responsible for the chronicity of otitis media.

    Notes: Abstract Among nerve cells of vertebrates, the olfactory epithelia are uncommon in their capacity for cell turnover. Apoptosis is well known to play a key role in maintaining homeostasis in continuously renewing tissues. We examined whether true apoptosis occurred in the normal olfactory epithelia of healthy adult guinea pigs using nucleic acid labeling. Subsequently, apoptosis was recognized in olfactory nerve cells, indicating that apoptosis might play a role in turnover of the olfactory epithelium.

    Notes: Summary We have examined the mucociliary function present in a mucocoele of the maxillary antrum and have found certain abnormalities in the tissues studied. Our findings also indicate that the mucocoele's intrinsic pathology is too complex to be improved by any conservative treatment and justifies its surgical removal.
